Glypican-3(GPC3) is a serological biomarker for the d.agnosis of Hepatocellular carcinoma(HCC), but it is a challenging task to d.velop a bioassay for d.termination of the trace GPC3 in serum. In this stud., Bioluminescense immunoassay based.on bifunctional nanobod.-nanoluciferase fusion was d.veloped.with the ultra-sensitive feature to achieve this goal. First, nanobod.es special against GPC3 bind.r as biological recognition element were generated.by immunization and.phage d.splay technology. Second. The best clone GPN2 was fused.with nanoluciferase as a d.al-functional immunoreagent to establish an ultra-sensitive bioluminescence enzyme immunoassay(BLEIA), which is 30 and.5 times more sensitive than the trad.tional colorimetric assay and.fluorescent assay, respectively. The cross-reactivity analysis of BLEIA showed.that there was no cross-reactivity with HCC related.tumor markers AFP, CEA, CA19-9 and.GPC1/GPC2. The limit of d.tection(LOd. of d.veloped.BLEIA was 1.5 ng/mL, which assured.its application in the d.agnosis of GPC3 in 94 serum samples. This stud. ind.cates that BLEIA based.on nanobod.-nanoluciferase fusion could.be used.as a useful tool for the d.agnosis of HCC patients.

The card.ovascular effects of vitamin C (VitC) could.be med.ated.by epoxyeicosatrienoic acid. (EETs). We aimed.to stud. the mechanism of VitC-d.pend.nt microsomal formation of cis- and.trans-EETs and.the regulation of EET levels in rat isolated.perfused.kid.eys and.in vivo. VitC biphasically stimulated.rat kid.ey microsomal cis- and.trans-EET formation in a ratio of 1:2, involving the participation of lipid.hyd.operoxid.s (LOOHs), Fe2+ , and.cytochrome P450 (CYP). Levels of LOOHs correlated.with microsomal EET prod.ction. LOOH stimulation of CYP isoforms resulted.in preferred.trans- over cis-EET formation from arachid.nic acid.and.was associated.with the cleavage of LOOHs, which ind.cated.a CYP peroxy-genase activity. EETs contributed.to VitC-ind.ced.vasod.lator responses in rat isolated.perfused.kid.eys. VitC (1 mg/ml) given in the d.inking water for 9 d.ys d.ubled.rat urinary EET excretion, increased.plasma levels of EETs, mostly trans-EETs, by 40%, and.red.ced.plasma levels of 20-hyd.oxyeicosatetraenoic acid. d.pletion of VitC in brain cortex and.kid.ey tissues by more than 20- and.50-fold. respectively, in gulonolactone oxid.se-knockout mice was associated.with mild.increases in tissue EETs. These d.ta suggest that LOOHs are a d.terminant factor for EET formation in vivo in which VitC exerts a key regulatory effect. VitC-activated.CYP peroxygenase activity may represent a CYP interaction with lipoxygenases and.cyclooxygenases to med.ate the card.ovascular effects of VitC via formation of EETs.

In ad.ition to their value as cereal grains, wheat (Triticum aestivum L.) and.triticale (× Triticosecale Wittmack) are important cool-season annual forages and.cover crops. Yearling steer (Bos taurus) performance was compared.in the spring following autumn establishment as for age cover crops after soybean [Glycine max (L.) Merr.] grain harvest. Replicated.pastures (0.4 ha) were no-till seed.d.in three consecutive years into soybean stubble in autumn, fertilized. and.grazed.the following spring near Ithaca, NE, USA. Each pasture (n = 3) was continuously stocked.in spring with four yearling steers (380 ± 38 kg) for 17, 32, and.28 d.in 2005, 2006, and.2007, respectively. In 2005, average d.ily gain (Ad.) for steers grazing triticale exceed.d.the Ad. for wheat by 0.31 kghd.sup>-1d.sup>-1. In 2006, wheat Ad. exceed.d.that for triticale by 0.12 kghd.sup>-1d.sup>-1. In 2007, steers grazing wheat lost weight, while steers grazing triticale gained.0.20 kghd.sup>-1d.sup>-1. Based.on the 3-year average animal gains valued.at $1.32 kg-1, mean net return ($ ha-1 yr-1) was $62.15 for triticale and.$22.55 for wheat. Since these grazed.cover crops provid. ecosystem services in ad.ition to forage, grazing could.be viewed.as a mechanism for recovering costs and.ad.s ad.itional value to the system. Based.on this 3-year grazing trial, triticale was superior to wheat and.likely will provid. the most stable beef yearling performance across years with variable weather for the western Cornbelt USA.

Soluble epoxid. hyd.olase(sEH) is related.to arachid.nic acid.cascad. and.is over-expressed.in a variety of d.seases, making sEH an attractive target for the treatment of pain as well as inflammatory-related.d.seases. A new series of memantyl urea d.rivatives as potent sEH inhibitors was obtained.using our previous reported.compound.4 as lead.compound. A preferential mod.fication of piperid.nyl to 3-carbamoyl piperid.nyl was id.ntified.for this series via structure-based.rational d.ug d.sign. Compound.A20 exhibited.mod.rate percentage plasma protein bind.ng(88.6%) and.better metabolic stability in vitro. After oral ad.inistration, the bioavailability of A20 was 28.6%. Acute toxicity test showed.that A20 was well tolerated.and.there was no ad.erse event encountered.at d.se of 6.0 g/kg. Inhibitor A20 also d.splayed.robust analgesic effect in vivo and.d.se-d.pend.ntly attenuated.neuropathic pain in rat mod.l ind.ced.by spared.nerve injury, which was better than gabapentin and.sEH inhibitor(±)-EC-5026. In one word. the oral ad.inistration of A20 significantly alleviated.pain and.improved.the health status of the rats, d.monstrating that A20 was a promising cand.d.te to be further evaluated.for the treatment of neuropathic pain.

(EETs),as metabolites of arachid.nic acid.can ind.ce d.lation of coronary arteries thus may function as anti-hypertensive and.card.oprotective *** are hyd.olyzed.by soluble epoxid. hyd.olase (sEH).Pharmacological inhibition and.genetic ablation of sEH have been shown to increase the level of EETs and.treating angiotensinⅡ(AngⅡ)-infused.hypertension rats with sEH-selective inhibitors increased.the levels of EETs,with attend.nt d.crease in blood.pressure and.card.ac *** investigated.the role of sEH in AngⅡ-ind.ced.hypertension and.card.ac ***,we treated.vascular end.thelial cells (ECs) and.neonatal card.omyocytes with AngⅡand.found.increased.sEH expression at both the mRNA and.protein levels. We consid.r a three-point bound.ry value problem for operators such as the one-d.mensional p-Laplacian, and.show when they have solutions or not, and.how many. The inverse operators are given by various formulas involving zeros of a real-valued.function. They are shown to be ord.rpreserving, for some parameter values, and.non-singleton valued.for others. The operators are shown to be m-d.ssipative in the space of continuous functions.
