Interferon gamma boosts the nucleotide oligomerization domain 2-mediated signaling pathway in human dendritic cells in an X-linked inhibitor of apoptosis protein and mammalian target of rapamycin-dependent manner

The cytoplasmic nucleotide oligomerization domain 2 （NOD2） receptor recognizes the bacterial cell wall component muramyl dipeptide （MDP）. NOD2 ligation initiates the nuclear factor kappa B and the mitogen-activated protein kinase cascades. However, administering MDP alone is insufficient to elicit strong cytokine responses in various immune cells, including dendritic cells （DCs）. Because the simultaneous presence of various microbial products and cytokines in inflamed tissues modulates DC function, we initiated this study to examine how interferon gamma （IFNy）, a central modulator of inflammation, affects the NOD2-mediated signaling pathway in human conventional DCs （cDCs）. Synergistic stimulation of DCs with MDP and IFNy increased the expression of CD40, CD80, CD83, CD86, and human leukocyte antigen DQ proteins and significantly elevated the production of pro-inflammatory cytokines IL- 1β, IL-6, IL-12, and tumour necrosis factor （TNF）, as well as anti-inflammatory cytokine IL-10. Furthermore, the simultaneous presence of MDP and I FNy was necessary to decrease IkBe protein levels. By investigating various mechanisms implicated in MDP- and IFNγ-mediated signaling pathways, we revealed that the increased production of pro-inflammatory cytokines is highly dependent on the X-linked inhibitor of apoptosis protein （XIAP） but not on cellular lAP1 and lAP2. We also found that the NOD2 signaling pathway is regulated by the mammalian target of rapamycin （mTOR） but is not affected by phosphatidylinositol-3 kinase or signal transducer and activator of transcription 1 inhibition. Our results demonstrate, for the first time, that IFNy positively affects NOD2-mediated signaling in human cDCs, in a manner considerably dependent on XIAP and partially dependent on mTOR.

Methionine(Met)is an essential and first limiting amino acid in the poultry diet that plays a significant role in chicken embryonic development and *** present study examined the effect of in ovo injection of DL-Met and L-Met sources and genotypes on chicken embryonic-intestinal development and *** eggs of the two genotypes,TETRA-SL layer hybrid(TSL)—commercial layer hybrid and Hungarian Partridge colored hen breed(HPC)—a native genotype,were randomly distributed into four treatments for each *** treatment groups include the following:1)control non-injected eggs(NoIn);2)saline-injected(SaIn);3)DL-Met injected(DLM);and 4)L-Met injected(LM).The in ovo injection was carried out on 17.5 d of embryonic development;after hatching,eight chicks per group were sacrificed,and the jejunum was extracted for *** results showed that both DLM and LM groups had enhanced intestinal development as evidenced by increased villus width,villus height,and villus area(P<0.05)compared to the *** DLM group had significantly reduced crypt depth,glutathione(GSH)content,glutathione S-transferase 3 alpha(GST3),occludin(OCLN)gene expression and increased villus height to crypt depth ratio in the TSL genotype than the LM group(P<0.05).The HPC genotype has overexpressed insulin-like growth factor 1(IGF1)gene,tricellulin(MD2),occludin(OCLN),superoxide dismutase 1(SOD1),and GST3 genes than the TSL genotype(P<0.05).In conclusion,these findings showed that in ovo injection of Met enhanced intestinal development,and function,with genotypes responding differently under normal *** also influenced the expression of intestinal antioxidants,tight junction,and growth-related genes.

Holocene Climate Evolution of the Ugii Nuur Basin, Mongolia

In order to evaluate the Holocene palaeoenvironmental evolution of the Ugii Nuur basin, central Mongolia, investigations on chemical and mineralogical properties of lacustrine sediments were carried out on a 630 cm sediment core from lake Ugii Nuur. The interpretation of the record is based on a principal component analysis （PCA） of the elemental composition of the samples. The results show that lacustrine deposition started at 10.6 kyr BP. Low lake level conditions were identified during the Early Holocene （10.6-7.9 kyr BP）. The Mid Holocene （7.9-4.2 kyr BP） was characterized by generally higher lake levels and thus higher moisture supply, but it experienced strong climatic fluctuations. Arid conditions prevailed from 4.2-2.8 kyr BP and were followed by a stable, more humid phase until today.

Coffea(coffee) species are grown in almost all countries along the Equator. Many members of the genus have a large production history and an important role both in the global market and researches. Seeds(Coffeae semen) are successfully used in food, cosmetic, and pharmaceutical industries due to its caffeine and high polyphenol content. Nowadays, the three best-known coffee species are Arabic(Coffea arabica L.), Robusta(Coffea robusta L. Linden), and Liberian coffees(Coffea liberica Hier.). Even though, many records are available on coffee in scientific literature, wild coffee species like Bengal coffee(Coffea benghalensis Roxb. Ex Schult.) could offer many new opportunities and challenges for phytochemical and medical studies. In this comprehensive summary, we focused on the ethnomedicinal, phytochemical, and medical significance of coffee species up to the present.

Comparative phytochemical analysis of Coffea benghalensis Roxb. Ex Schult, Coffea arabica L. and Coffea liberica Hiern

Objective: To make phytochemical studies of the leaf, pericarp and seed of Coffea benghalensis(C. Benghalensis) compared with those of the widely known Coffea arabica and Coffea liberica. Methods: The sample extracts were prepared by Soxhlet-extraction. Polyphenol content was analyzed by HPLC-ESI-MS/MS, the identification was carried out based on the retention time, UV and mass spectra of standards and literature data of the detected compounds. Results: Phenolic acids like caffeoylquinic acids, dicaffeoylquinic acids, feruloylquinic acids and coumaroylquinic acid, as well as mangiferin were detected as main constituents in all extracts. Procyanidin trimers were present exclusively in the leaves. In C. benghalensis, main constituents were 5-caffeoylquinic acid and 4-caffeoylquinic acid. Flavan-3-ols were described in all immature and mature pericarp and leaf extracts. Even though 4-feruloylquinic acid was described in both immature and mature seed, dicaffeoylquinic acids were identified only in the mature seed extracts. Mangiferin was present in the leaf, mature pericarp and seed. Conclusions: These analyses provide new chemotaxonomical data for the selected coffees, especially for C. benghalensis. Due to its high polyphenol content, our results indicate its significance of providing new data as a possible source for industry.

Soil Properties in Two Soil Profiles from Terraces of the Nam Co Lake in Tibet,China

This article investigates the soil properties in two soil profiles from the terraces of the Nam Co Lake in Tibet,China. Profile 1,with a slope aspect to east,is 37 m,and profile 2,to southwest,is 32 m above the lake level. Twelve and nine layers were identified in profile 1 and profile 2,respectively,based on the color,texture,structure,moisture content,presence of carbonate,and root density. Grain sizes and measurements of water content,total organic carbon,total inorganic carbon,total carbon,pH value and electrical conductivity were all analyzed for each sample. The results show that soil is poorly developed because of sparse vegetation coverage and weak chemical weathering in the cold and arid area. The results also indicate that the soil properties are different between the two profiles and suggest that soil is better developed in profile 1 than in profile 2. These differences are ascribed to the dissimilarity of landform,vegetation and timing. Profile 1 faces to east as compared with profile 2 facing to southwest and gets less insolation and therefore less evaporation. Thus,vegetation grows better in profile 1 than in profile 2. Radiocarbon dating and OSL dating show that profile 1 underwent a longer soil forming process than profile 2,which is helpful for the soil development in profile 1.

Prophylactic transcatheter arterial embolization reduces rebleeding in non-variceal upper gastrointestinal bleeding: A meta-analysis

BACKGROUND Despite the improvement in the endoscopic hemostasis of non-variceal upper gastrointestinal bleeding(NVUGIB),rebleeding remains a major *** To assess the role of prophylactic transcatheter arterial embolization(PTAE)added to successful hemostatic treatment among NVUGIB *** We searched three databases from inception through October 19th,*** controlled trials(RCTs)and observational cohort studies were *** compared patients with NVUGIB receiving PTAE to those who did not get *** outcomes were rebleeding,mortality,reintervention,need for surgery and transfusion,length of hospital(LOH),and intensive care unit(ICU)*** the quantitative synthesis,odds ratios(ORs)and weighted mean differences(WMDs)were calculated with the random-effects model and interpreted with 95%confidence intervals(CIs).RESULTS We included a total of 3 RCTs and 9 observational studies with a total of 1329 patients,with 486 in the intervention *** was associated with lower odds of rebleeding(OR=0.48,95%CI:0.29–0.78).There was no difference in the 30-d mortality rates(OR=0.82,95%CI:0.39–1.72)between the PTAE and control *** who underwent PTAE treatment had a lower chance for reintervention(OR=0.48,95%CI:0.31–0.76)or rescue surgery(OR=0.35,95%CI:0.14–0.92).The LOH and ICU stay was shorter in the PTAE group,but the difference was non-significant[WMD=-3.77,95%CI:(-8.00)–0.45;WMD=-1.33,95%CI:(-2.84)–0.18,respectively].CONCLUSION PTAE is associated with lower odds of rebleeding and any reintervention in ***,further RCTs are needed to have a higher level of evidence.
