We previously demonstrated that endogenous phosphatidic acid(PA)promotes liver regeneration after acetaminophen(APAP)***,we hypothesized that exogenous PA is also *** test that,we treated mice with a toxic APAP dose at 0 h,followed by PA or vehicle(Veh)*** then collected blood and liver at 6,24,and 52 ***-treatment with PA 2 h after APAP protected against liver injury at 6 h,and the combination of PA and n-acetyl-L-cysteine(nAC)reduced injury more than nAC ***,PA did not affect canonical mechanisms of APAP ***,transcriptomics revealed that PA activated interleukin-6(IL-6)signaling in the *** with that,serum IL-6 and hepatic signal transducer and activator of transcription 3(Stat3)phosphorylation increased in PA-treated ***,PA failed to protect against APAP in IL-6-deficient ***,IL-6 expression increased 18-fold in adipose tissue after PA,indicating that adipose is a source of PA-induced circulating ***,however,exogenous PA did not alter regeneration,despite the importance of endogenous PA in liver repair,possibly due to its short *** data demonstrate that exogenous PA is also beneficial in APAP toxicity and reinforce the protective effects of IL-6 in this model.

Modeling the Influence of Salt on the Hydrophobic Effect and Protein Fold Stability

Salt influences protein stability through electrostatic mechanisms as well as through nonpolar Hofmeister *** the present work,a continuum solvation based model is developed to explore the impact of salt on protein *** model relies on a traditional Poisson-Boltzmann(PB)term to describe the polar or electrostatic effects of salt,and a surface area dependent term containing a salt concentration dependent microscopic surface tension function to capture the non-polar Hofmeister *** model is first validated against a series of cold-shock protein variants whose salt-dependent protein fold stability profiles have been previously determined *** approach is then applied to HIV-1 protease in order to explain an experimentally observed enhancement in stability and activity at high(1M)naCl *** inclusion of the salt-dependent non-polar term brings the model into quantitative agreement with experiment,and provides the basis for further studies into the impact of ionic strength on protein structure,function,and evolution.

Examining Electrostatic Influences on Base-Flipping:A Comparison of TIP3P and GB Solvent Models

Recently,it was demonstrated that implicit solvent models were capable of generating stable B-form DnA ***,generalized Born(GB)implicit solvent models have improved regarding the solvation of conformational sampling of DnA[1,2].Here,we examine the performance of the GBSW and GBMV models in CHARMM for characterizing base flipping free energy profiles of undamaged and damaged DnA *** sampling of the base flipping process was performed for the bases cytosine,uracil and *** umbrella sampling simulations were carried-out with both explicit(TIP3P)and implicit(GB)solvent in order to establish the impact of the solvent model on base ***,base flipping potential of mean force(PMF)profiles generated with GB solvent resulted in a greater free energy difference of flipping than profiles generated with *** of the significant differences between implicit and explicit solvent models is the approximation of solute-solvent interactions in implicit solvent *** calculated electrostatic interaction energies between explicit water molecules and the base targeted for *** interaction energies were calculated over the base flipping reaction coordinate to illustrate the stabilizing effect of the explicit water molecules on the flipped-out *** is known that nucleic base pair hydrogen bonds also influenced the free energy of flipping since these favorable interactions must be broken in order for a base to flip-out of the *** Watson-Crick base pair hydrogen bond fractions were calculated over the umbrella sampling simulation windows in order to determine the effect of base pair interactions on the base flipping free *** is shown that interaction energies between the flipping base and explicit water molecules are responsible for the lower base flipping free energy difference in the explicit solvent PMF profiles.

The MYC transcription factor network： balancing metabolism, proliferation and oncogenesis

Transcription factor networks have evolved in order to control, coordinate, and separate, the functions of distinct network modules spatially and temporally. In this review we focus on the MYC network （also known as the MAX-MLX network）, a highly conserved super-family of related basic-helix-loop-helix-zipper （bHLHZ） proteins that functions to integrate extracellular and intracellular signals and modulate global gene expression. Importantly the MYC network has been shown to be deeply involved in a broad spectrum of human and other animal cancers. Here we summarize molecular and biological properties of the network modules with emphasis on functional interactions among network members. We suggest that these network interactions serve to modulate growth and metabolism at the transcriptional level in order to balance nutrient demand with supply, to maintain growth homeostasis, and to influence cell fate. Moreover, oncogenic activation of MYC and/or loss of a MYC antagonist, results in an imbalance in the activity of the network as a whole, leading to tumor initiation, progression and maintenance.

Physical activity and nutrition attitudes in obese Hispanic children with non-alcoholic steatohepatitis

numerical simulation on inclusion transport in continuous casting mold

Turbulent flow, the transpor＇t of inclusions and bubbles, and inclusion removal by fluid flow, transport and by bubble flotation in the strand of the continuous slab caster are investigated using computational models, and validated through comparison with plant measurements of inclusions. Steady 3-D flow of steel in the liquid pool in the mold and upper strand is simulated with a finitedifference computational model using the standard k-εturbulence rondel. Trajectories of inclusions and bubhles tire calculated by integrating each local velocity, considering its drag and buoyancy forces, A ＂random walk＂ model is used to incorporate the effect of turbulent fluctuations on the particle motion. The attachment probability of inclusions on a bubble surface is investigated based on fundamental fluid flow simulations, incorporating the turbulent inclusion trajectory and sliding time of each individual inclusion along the bubble surface as a function of particle and bubble size. The chunge in inclusion distribution due to removal by bubble transport in the mold is calculated based on the computed attachment probability of inclusions on each bubble and the computed path length of the bubbles. The results indicate that 6%-10% inclusions are removed by fluid flow transport. 10% by bubble flotation, and 4% by entrapment to the submerged entry nozzle （SEn） walls. Smaller bubbles and larger inclusions have larger attachment probabilities. Smaller bubbles are more efficient for inclusion removal by bubble flotation, so Inng as they are not entrapped in the solidifying shell A larger gas flow rate favors inclusion removal by bubble flotation. The optimum bubble size should be 2-4mm.
