Although genome-wide association studies have identified more than eighty genetic variants associated with non-small cell lung cancer(NSCLC)risk,biological mechanisms of these variants remain largely *** integrating a large-scale genotyp. data of 15581 lung adenocarcinoma(AD)cases,8350 squamous cell carcinoma(SqCC)cases,and 27355 controls,as well as multip.e transcrip.ome and ep.genomic databases,we conducted histology-sp.cific meta-analyses and functional annotations of both rep.rted and novel suscep.ibility *** identified 3064 credible risk variants for NSCLC,which were overrep.esented in enhancer-like and p.omoter-like histone modification p.aks as well as DNase I hyp.rsensitive *** factor enrichment analysis revealed that USF1 was AD-sp.cific while CREB1 was *** annotation and genebased analysis imp.icated 894 target genes,including 274 sp.cifics for AD and 123 for SqCC,which were overrep.esented in somatic driver genes(ER=1.95,p.0.005).p.thway enrichment analysis and Gene-Set Enrichment Analysis revealed that AD genes were p.imarily involved in immune-related p.thways,while SqCC genes were homologous recombination deficiency *** results illustrate the molecular basis of both wellstudied and new suscep.ibility loci of NSCLC,p.oviding not only novel insights into the genetic heterogeneity between AD and SqCC but also a set of p.ausible gene targets for p.st-GWAS functional exp.riments.

To evaluate the effectiveness of a p.ogramme of asthma clubs in imp.oving quality of life in p.imary school children with asthma. Methods: A cluster randomised intervention trial was undertaken in 22 p.imary schools within the urban area of south and east Belfast, Northern Ireland. Schools were randomised in p.irs to immediate or delayed group.. The study subjects comp.ised 173 children aged 7- 11 years whose p.rents had notified the school of their asthma diagnosis. Children attended school based weekly clubs over an 8 week p.riod. The main outcome measures were the interview administered p.ediatric Quality of Life Questionnaire scores, ranging from 1 (worst) to 7 (best), sp.rometry, and inhaler technique. Results: Over 15 weeks, small but non-significant imp.ovements in the overall quality of life score (mean 0.20; 95% confidence interval (CI) - 0.20 to 0.61) and in each of its three comp.nents, activity limitation (0.20; - 0.43 to 0.84), symp.oms (0.23; - 0.23 to 0.70), and emotional function (0.17; - 0.18 to 0.52), were observed in the immediate comp.red with the delayed group. Inhaler technique at week 16 was markedly better in the immediate group. with 56% having correct technique comp.red with 15% in the delayed group. No significant effect of the intervention on sp.rometry results could be demonstrated. Conclusion: This p.imary school based asthma education p.ogramme resulted in sustained imp.ovements in inhaler technique, but changes in quality of life scores were not significant.
