Evaluation of Cytotoxicity and Genotoxicity of Insecticide Carbaryl to Flounder Gill Cells and Its Teratogenicity to Zebrafish Embryos

In this study, we determined the cytotoxicity and genotoxicity of carbamate insecticide carbaryl to flounder gill （FG） cells and its teratogenicity to zebrafish embryos. The cytotoxicity of carbaryl to FG cells was determined with methods including MTT and neutral red uptaking （NRU）, lactate dehydrogenase （LdH） releasing and Hoechst 33342 and propidiurn idodide （PI） double staining. Moderate cytotoxicity in a concentration-dependent manner was observed. The 24h-IC50 value of 53.48 ± 1.21, 59.13 ± 1.19 and 46.21 ± 1.24 mgL 1 carbaryl was obtained through MTT, NRU and LdH assays, respectively. double fluorescence staining dem- onstrated that carbaryl induced the death of FG cells mainly through necrosis. There was no significant genotoxicity found in the FG cells exposed to the highest testing concentration of carbaryl （20mgL-1, P〉0.05） as was demonstrated by Comet assay. Zebrafish embryos exposed to carbaryl at concentrations 〉10 mg L-1 displayed moderate toxic effects on the survival, spontaneous movement, hatching, heart rates of the embryos and their development, which were evidenced by yolk and pericardial sac edemas, body length reduction and tail flexure in time- and concentration-dependent manners at specific stages. The 24h-, 48h- and 96h-LC50 values of carbaryl to zebra fish embryos were 41.80 ± 1.10, 17.80 ± 1.04 and 14.46 ± 1.05 mg L-1, respectively. These results suggested that car- baryl is moderately toxic to FG ceils cultured in vitro and zebrafish embryos, and the FG cells were similar to zebrafish embryos in their sensitivity to carbaryl as 24h-IC50 and LC50 indicated.

Central nervous system and peripheral cell labeling by vascular endothelial cadherin-driven lineage tracing in adult mice

Understanding the contribution of endothelial cells to the progenitor pools of adult tissues has the potential to inform therapies for human *** address whether endothelial cells transdifferentiate into non-vascular cell types,we performed cell lineage tracing analysis using transgenic mice engineered to express a fluorescent marker following activation by tamoxifen in vascular endothelial cadherin promoter-expressing cells(VEcad-CreERT2;B6 Cg-Gt(ROSA)26Sortm9(CAG-tdTomato)Hze).Activation of target-cell labeling following 1.5 months of ad libitum feeding with tamoxifen-laden chow in 4–5 month-old mice resulted in the tracing of central nervous system and peripheral cells that include:cerebellar granule neurons,ependymal cells,skeletal myocytes,pancreatic beta cells,pancreatic acinar cells,tubular cells in the renal cortex,duodenal crypt cells,ileal crypt cells,and hair follicle stem *** Nestin expression has been reported in a subset of endothelial cells,Nes-CreERT2 mice were also utilized in these *** tracing of cells in adult Nes-CreERT2 mice revealed the labeling of canonical progeny cell types such as hippocampal and olfactory granule neurons as well as ependymal ***,Nestin tracing also labeled skeletal myocytes,ileal crypt cells,and sparsely marked cerebellar granule *** findings provide support for endothelial cells as active contributors to adult tissue progenitor *** information could be of particular significance for the intravenous delivery of therapeutics to downstream endothelial-derived cellular *** animal experiments were approved by the Boise State University Institute Animal Care and Use Committee(approval No.006-AC15-018)on October 31,2018.

discovery of nigral dopaminergic neurogenesis in adult mice

Parkinson＇s disease is characterized by the loss of dopaminergic neurons in the substantia nigra. As a result, intensive efforts have focused upon mechanisms that facilitate the death of mature dopaminergic neurons. Unfortunately, these efforts have been unsuccessful in providing an effective treatment to address neurodegeneration in this disease. Therefore, alternative theories of pathogenesis are being explored. Adult neurogenesis of dopaminergic neurons is an attractive concept that would provide a possible mechanism of neurodegeneration as well as offer an endogenous means to replenish affected neurons. To determine whether dopaminergic neurons experience neurogenesis in adult mice we developed a novel cell lineage tracing model that permitted detection of neurogenesis without many of the issues associated with popular techniques. Remarkably, we discovered that dopaminergic neurons are replenished in adult mice by Nestin＋/Sox2- progenitor cells. What＇s more, the rate of neurogenesis is similar to the rate of dopaminergic neuron loss reported using a chronic, systemic inflammatory response mouse model. This observation may indicate that neuron loss in Parkinson＇s disease results from inhibition of neurogenesis.

The impact of alloantibodies directed against the second donor on long-term outcomes of repeat liver transplantation

However, literature has identified an association between endurance exercise and atrial fibrillation with the implication that the incidence of atrial fibrillation increases in endurance athletes.1,2 To this point, researchers have grouped diverse activities （running, biking, etc.） of varying intensity in arriving at these conclusions,z＇ Proposed mechanisms for the associa- tion between prolonged physical activity and atrial fibrillation include left atrial remodeling, increased atrial ectopy, and increased vagal tone induced by endurance exercise.

Investigating the Prevalence and Risk Factors of depression Symptoms among NCAA division I Collegiate Athletes

This study aimed to determine an overall prevalence rate for depression symptoms among National Collegiate Athletic Association （NCAA） division I collegiate athletes while also assessing various risk factors that may increase an athlete＇s vulnerability to depression. Using a sample of 950 NCAA division I athletes, it was found that 33.2% of athletes experienced symptoms of depression, contradicting findings from previous studies that have suggested a prevalence rate lower than the general college population. Female athletes （P = 0.00）, underclassmen （P = 0.01）, recently injured athletes （P = 0.05）, and in-season athletes （P = 0.05）, were all found to experience higher rates of depression symptoms than other athletes. It was found that 25.7% of athletes did not know how or where to access mental health treatment at their university, and 44.5% had received no mental health education from their athletic department. The results from this study suggest that depression is a more significant issue in college athletics than previously thought, and they highlight the need for continued improvements to be made in both the understanding of mental health issues in college athletics and the services that are provided to athletes.
