Study on the preservation effects of the amputated forelimb by machine perfusion at physiological temperature

Purpose:Ischemia and hypoxia are the main factors limiting limb replantation and transplantation.Static cold storage(SCS),a common preservation method for tissues and organs,can only prolong limb ischemia time to 4-6 h.The normothermic machine perfusion(NMP)is a promising method for the preservation of tissues and organs,which can extend the preservation time in vitro by providing continuous oxygen and nutrients.This study aimed to evaluate the difference in the efficacy of the 2 limb preservation methods.Methods:The 6 forelimbs from beagle dogs were divided into 2 groups.In the SCS group(n=3),the limbs were preserved in a sterile refrigerator at 4°C for 24 h,and in the NMP group(n=3),the perfusate prepared with autologous blood was used for the oxygenated machine perfusion at physiological temperature for 24 h,and the solution was changed every 6 h.The effects of limb storage were evaluated by weight gain,perfusate biochemical analysis,enzyme-linked immunosorbent assay,and histological analysis.All statistical analyses and graphs were performed using GraphPad Prism 9.0 one-way or two-way analysis of variance.The p value of less than 0.05 was considered to indicate statistical significance.Results:In the NMP group,the weight gained percentage was 11.72%±4.06%;the hypoxia-inducible factor-1αcontents showed no significant changes;the shape of muscle fibers was normal;the gap between muscle fibers slightly increased,showing the intercellular distance of(30.19±2.83)μm;and the vascularα-smooth muscle actin(α-SMA)contents were lower than those in the normal blood vessels.The creatine kinase level in the perfusate of the NMP group increased from the beginning of perfusion,decreased after each perfusate change,and remained stable at the end of perfusion showing a peak level of 4097.6 U/L.The lactate dehydrogenase level of the NMP group increased near the end of perfusion and reached the peak level of 374.4 U/L.In the SCS group,the percentage of weight gain was 0.18%±0.10%,and the contents of hypoxia-inducible factor-1αin

Different percutaneous transhepatic biliary stent placements and catheter drainage in the treatment of middle and low malignant biliary obstruction

BACKGROUND For cases of middle and low biliary obstruction with left and right hepatic duct dilatation,the type of approach and whether different approaches affect the difficulty of puncture operation and intraoperative and postoperative complications have not been discussed in detail.AIM To compare the efficacy of different percutaneous transhepatic biliary stent placements and catheter drainage in treating middle and low biliary obstruction.METHODS A retrospective analysis was performed on the medical records of 424 patients with middle and low biliary obstruction who underwent percutaneous liver puncture biliary stent placement and catheter drainage at the Department of Interventional Radiology,Shaanxi Provincial People’s Hospital between March 2016 and March 2022.Based on the puncture path,patients were categorized into two groups:Subxiphoid left hepatic lobe approach group(Group A,224 cases)and right intercostal,right hepatic lobe approach group(Group B,200 cases).Liver function improvement,postoperative biliary bleeding incidence,postoperative pain duration,and abdominal effusion leakage around the drainage tube were compared between the two groups at 3 d and 1 wk after the surgery.Patient survival time was recorded during follow-up.RESULTS All 424 surgeries were successful without adverse events.Group A comprised 224 cases,and Group B had 200 cases.There was no statistically significant difference in basic data between Group A and Group B(P>0.05).No significant difference in postoperative biliary bleeding incidence was observed between the groups(P>0.05).The decreased rates for total bilirubin(Group A:69.23±4.50,Group B:63.79±5.65),direct bilirubin(Group A:79.30±11.19,Group B:63.62±5.64),and alkaline phosphatase(Group A:60.51±12.23,Group B:42.68±23.56)in the 1st wk after surgery were significantly faster in Group A than in Group B.The decreased rate of gamma-glutamyl transpeptidase was also significantly faster in Group A at both 3 d(Group A:40.56±10.32,Group B:32.22±5.12)and 1 wk(Group A:73.19±7.05,Group

Coherent link between a Ti:sapphire comb and a 1.5μm laser via nonlinear interaction in photonic crystal fiber

Optical clock networks have distinct advantages for the dissemination of time/frequency,geodesy,and fundamental research.To realize such a network,the telecom band and optical atomic clocks have to be coherently bridged.Since the telecom band and optical atomic clocks reside in a distinct spectral region,second-harmonic generation is usually introduced to bridge the large frequency gap.In this paper,we introduce a new method to coherently link a 1550 nm continuous wave laser with a Ti:sapphire mode-locked laser-based optical frequency comb.By coupling the 1550 nm continuous wave laser light and the Ti:sapphire comb light together into a photonic crystal fiber,nonlinear interaction takes place,and new comblike frequency components related to the 1550 nm laser frequency are generated in the visible region.Consequently,we can detect beat notes between two combs in the visible region with a signal-to-noise ratio of more than 40 dB in a resolution bandwidth of300 kHz.With this signal,we realize an optical frequency divider for converting the frequency of optical clocks in the visible region to the telecom band at 1.55μm.An out-of-loop measurement shows that the additional noise and uncertainty induced in optical frequency conversion are 5×10^(-18)at 1 s averaging time and 2.2×10^(-19),respectively,which are limited by the uncompensated light path fluctuation but fulfill precision measurement using state-of-the-art optical clocks.

Nd-Fe-B permanent magnets play a crucial role in energy conversion and electronic devices.The essential magnetic properties of Nd-Fe-B magnets,particularly coercivity and remanent magnetization,are significantly infuenced by the phase characteristics and microstructure.In this work,Nd-Fe-B magnets were manufactured using vacuum induction melting(VIM),laser directed energy deposition(LDED)and laser powder bed fusion(LPBF)technologies.Themicrostructure evolution and phase selection of Nd-Fe-B magnets were then clarified in detail.The results indicated that the solidification velocity(V)and cooling rate(R)are key factors in the phase selection.In terms of the VIM-casting Nd-Fe-B magnet,a large volume fraction of theα-Fe soft magnetic phase(39.7 vol.%)and Nd2Fe17Bxmetastable phase(34.7 vol.%)areformed due to the low R(2.3×10-1?C s-1),whereas only a minor fraction of the Nd2Fe14B hard magnetic phase(5.15 vol.%)is presented.For the LDED-processed Nd-Fe-B deposit,although the Nd2Fe14B hard magnetic phase also had a low value(3.4 vol.%)as the values of V(both the VIM-casting and LDED-processed Nd-Fe-B deposits exhibited poor magnetic properties.In contrast,employing the high values of V(>10-2m s-1)and R(1.45×106?C s-1)in the LPBF process resulted in the substantial formation of the Nd2Fe14B hard magnetic phase(55.8 vol.%)directly from the liquid,while theα-Fe soft magnetic phase and Nd2Fe17Bxmetastable phase precipitation are suppressed in the LPBF-processed Nd-Fe-B magnet.Additionally,crystallographic texture analysis reveals that the LPBF-processedNd-Fe-B magnets exhibit isotropic magnetic characteristics.Consequently,the LPBF-processed Nd-Fe-B deposit,exhibiting a coercivity of 656 k A m-1,remanence of 0.79 T and maximum energy product of 71.5 k J m-3,achieved an acceptable magnetic performance,comparable to other additive manufacturing

The BEL1-like transcription factor GhBLH5-A05 participates in cotton response to drought stress

Drought stress impairs crop growth and development.BEL1-like family transcription factors may be involved in plant response to drought stress,but little is known of the molecular mechanism by which these proteins regulate plant response and defense to drought stress.Here we show that the BEL1-like transcription factor GhBLH5-A05 functions in cotton(Gossypium hirsutum)response and defense to drought stress.Expression of GhBLH5-A05 in cotton was induced by drought stress.Overexpression of GhBLH5-A05 in both Arabidopsis and cotton increased drought tolerance,whereas silencing GhBLH5-A05 in cotton resulted in elevated sensitivity to drought stress.GhBLH5-A05 binds to cis elements in the promoters of GhRD20-A09 and GhDREB2C-D05 to activate the expression of these genes.GhBLH5-A05 interacted with the KNOX transcription factor GhKNAT6-A03.Co-expression of GhBLH5-A05 and GhKNAT6-A03 increased the transcription of GhRD20-A09 and GhDREB2C-D05.We conclude that GhBLH5-A05 acts as a regulatory factor with GhKNAT6-A03 functioning in cotton response to drought stress by activating the expression of the drought-responsive genes GhRD20-A09 and GhDREB2C-D05.

A Distributed Self-Tallying Electronic Voting System Using the Smart Contract

For electronic voting(e-voting) with a trusted authority, the ballots may be discarded or tampered, so it is attractive to eliminate the dependence on the trusted party. An e-voting protocol, where the final voting result can be calculated by any entity, is known as self-tallying e-voting protocol. To the best of our knowledge, addressing both abortive issue and adaptive issue simultaneously is still an open problem in self-tallying e-voting protocols.Combining Ethereum blockchain with cryptographic technologies, we present a decentralized self-tallying e-voting protocol. We solve the above problem efficiently by utilizing optimized Group Encryption Scheme and standard Exponential El Gamal Cryptosystem. We use zero-knowledge proof and homomorphic encryption to protect votes' secrecy and achieve self-tallying. All ballots can be verified by anyone and the final voting result can be calculated by any entity. In addition, using the paradigm of score voting and “1-out-of-k” proof, our e-voting system is suitable for a wide range of application scenarios. Experiments show that our protocol is more competitive and more suitable for large-scale voting.

Quantum-confined CsPbBr3 perovskites are promising blue emitters for ultra-high-definition displays,but their soft lattice caused by highly ionic nature has a limited stability.Here,we endow CsPbBr3 nanoplatelets(NPLs)with atomic crystal-like structural rigidity through proper surface engineering,by using strongly bound N-dodecylbenzene sulfonic acid(DBSA).A stable,rigid crystal structure,as well as uniform,orderly-arranged surface of these NPLs is achieved by optimizing intermediate reaction stage,by switching from molecular clusters to mono-octahedra,while interaction with DBSA resulted in formation of a CsxO monolayer shell capping the NPL surface.As a result,both structural and optical stability of the CsPbBr3 NPLs is enhanced by strong covalent bonding of DBSA,which inhibits undesired phase transitions and decomposition of the perovskite phase potentially caused by ligand desorption.Moreover,rather small amount of DBSA ligands at the NPL surface results in a short inter-NPL spacing in their closely-packed films,which facilitates efficient charge injection and transport.Blue photoluminescence of the produced CsPbBr3 NPLs is bright(nearly unity emission quantum yield)and peaks at 457 nm with an extremely narrow bandwidth of 3.7 nm at 80 K,while the bandwidth of the electroluminescence(peaked at 460 nm)also reaches a record-narrow value of 15 nm at room temperature.This value corresponds to the CIE coordinates of(0.141,0.062),which meets Rec.2020 standards for ultra-high-definition displays.

Synthesis of betanin by expression of the core betalain biosynthetic pathway in carrot

Betalain has received increased attention because of its high nutritional value and crucial physiological functions.Based on the elucidation of its core biosynthetic pathway,betalain can be produced in additional plants by metabolic engineering.Synthesis of betalain in carrot(Daucus carota L.)can improve its nutritional quality and economic value by extracting betalain from the fleshy root,non-edible part,and processing residue of carrot.In this study,two different constructs,namely,pYB:mCD(AomelOS,BvCYP76AD1S,and BvDODA1S)and p YB:CDD(BvCYP76AD1S,BvDODA1S,and MjcDOPA5GTS),were introduced into carrot for betanin synthesis by Agrobacterium-mediated transformation.Betanin can be synthetized in both transgenic calli,and p YB:m CD-transgenic callus can be used to produce betacyanin by suspension culture.However,pYB:mCD-transgenic seedlings can synthetize betanin only by tyrosine feeding.The p YB:CDD-transgenic lines can synthetize betanin in whole plants.The betanin content in fleshy root of pYB:CDD-transgenic carrot was(63.4±9)μg·g^(-1)fresh weight according to quantitative analysis.These betanin-producing carrot plant materials can be used to synthesize betanin for industrial application or consumption as dietary sources.

Targeting the Warburg effect:A revisited perspective from molecular mechanisms to traditional and innovative therapeutic strategies in cancer

Cancer reprogramming is an important facilitator of cancer development and survival,with tumor cells exhibiting a preference for aerobic glycolysis beyond oxidative phosphorylation,even under sufficient oxygen supply condition.This metabolic alteration,known as the Warburg effect,serves as a significant indicator of malignant tumor transformation.The Warburg effect primarily impacts cancer occurrence by influencing the aerobic glycolysis pathway in cancer cells.Key enzymes involved in this process include glucose transporters(GLUTs),HKs,PFKs,LDHs,and PKM2.Moreover,the expression of transcriptional regulatory factors and proteins,such as FOXM1,p53,NF-κB,HIF1a,and c-Myc,can also influence cancer progression.Furthermore,lncRNAs,miRNAs,and circular RNAs play a vital role in directly regulating the Warburg effect.Additionally,gene mutations,tumor microenvironment remodeling,and immune system interactions are closely associated with the Warburg effect.Notably,the development of drugs targeting the Warburg effect has exhibited promising potential in tumor treatment.This comprehensive review presents novel directions and approaches for the early diagnosis and treatment of cancer patients by conducting in-depth research and summarizing the bright prospects of targeting the Warburg effect in cancer.

SDH-FCOS:An Efficient Neural Network for Defect Detection in Urban Underground Pipelines

Urban underground pipelines are an important infrastructure in cities,and timely investigation of problems in underground pipelines can help ensure the normal operation of cities.Owing to the growing demand for defect detection in urban underground pipelines,this study developed an improved defect detection method for urban underground pipelines based on fully convolutional one-stage object detector(FCOS),called spatial pyramid pooling-fast(SPPF)feature fusion and dual detection heads based on FCOS(SDH-FCOS)model.This study improved the feature fusion component of the model network based on FCOS,introduced an SPPF network structure behind the last output feature layer of the backbone network,fused the local and global features,added a top-down path to accelerate the circulation of shallowinformation,and enriched the semantic information acquired by shallow features.The ability of the model to detect objects with multiple morphologies was strengthened by introducing dual detection heads.The experimental results using an open dataset of underground pipes show that the proposed SDH-FCOS model can recognize underground pipe defects more accurately;the average accuracy was improved by 2.7% compared with the original FCOS model,reducing the leakage rate to a large extent and achieving real-time detection.Also,our model achieved a good trade-off between accuracy and speed compared with other mainstream methods.This proved the effectiveness of the proposed model.
