Involvement of carbon monoxide produced by heme oxygenase in ABA-induced stomatal closure in Vicia faba and its proposed signal transduction pathway

Carbon monoxide (CO) has recently proven to be an important bioactive or signaling molecule in mammalian cells. Its effects are mainly mediated by nitric oxide (NO) and cyclic GMP (cGMP). In Vicia faba leaves, CO production and heme oxygenase (HO) activity, an important CO synthetic enzyme, are first reported to increase in response to ABA treatment, which could result in stomatal closure. Inter- estingly, ABA-induced stomatal closure in V. faba guard cells is partially blocked when the synthetic CO inhibitor ZnPP, or the CO/NO scavenger Hb is added. Furthermore, we show that, exogenously applied CO donor, hematin, and CO aqueous solution not only result in the enhancement of CO release, but also time-dependently induce stomatal closure, and the latter is mimicked by the application of an NO donor SNP. The above-mentioned stomatal closure effects are differentially reversed by the addition of tungstate, a potent inhibitor of NO synthetic enzyme nitrate reductase (NR), the specific NO scavenger cPTIO, ZnPP, or Hb. During treatment for 4 h, SNP, 0.01% CO aqueous solution or hematin significantly triggers NO synthesis, whereas cPTIO, or tungstate approximately fully inhibits NO fluorescence. Ad- ditionally, application of the GC inhibitor ODQ blocks CO-induced stomatal closure. This inhibition could be reversed when 8-Br-cGMP is added. Thus, the above results suggest that CO produced by HO is involved in ABA-induced stomatal closure, and NO and cGMP may function as downstream interme- diates in the CO signaling responsible for stomatal closure.

Microstructure and mechanical properties of a compound reinforced Mg95Y2.5Zn2.5 alloy with long period stacking ordered phase and W phase

The microstructure evolution of Mg100-2xYxZnx （x=2, 2.5, 3, 3.5） alloys was investigated. Results show that the Mg100-2xYxZnx alloys are composed of a-Mg, long period stacking ordered （LPSO） phase and eutectic structure phase （W phase）, and the Mg95Y2.5Zn2.5 alloy has the best comprehensive mechanical properties. Subsequently, the microstructure evolution of the optimized alloy Mg95Y2.5Zn2.5 during solidification and heat treatment processes was analyzed and discussed by means of OM, SEM, TEM, XRD and DTA. After heat treatment, the lamellar phase 14H-LPSO precipitated in a-Mg and W phase transforms into particle phase （MgyZn2）. Due to the compound reinforcement effect of the particle phase and LPSO phase （18R＋14H）, the mechanical properties of the alloy are enhanced. The tensile strength and elongation of the Mg95Y2.5Zn2.5 alloy is improved by 9.1% and 31.3% to 215 MPa and 10.5%, respectively, after solid-solution treatment.

为探明农业技术进步对农业碳排放强度的影响效应及作用机制,首先对其影响效应及作用机制做出理论分析并设置三个假设,然后基于2009-2021年30个省区市面板数据,运用双固定效应模型、中介效应模型进行实证分析。结果显示:2009-2021年我国农业碳排放强度从0.443(10^(4)t/10^(8)元)持续下降到0.234(10^(4)t/10^(8)元),下降了47.2%。农业技术进步对农业碳排放强度的影响系数为-0.152,表明农业技术进步率每提高1%将使农业碳排放强度减少0.152%。研究表明:第一,农业技术进步对农业碳排放强度具有显著的负向影响;第二,农业技术进步通过降低农业化学化水平从而降低农业碳排放强度;第三,农业技术进步通过提高农业产值水平从而降低农业碳排放强度。本文验证农业技术进步能降低农业碳排放强度,完善了农业碳排放强度影响因素体系,促进绿色农业技术进步,同时为农业科技创新方向提供降低农业化学化水平和提高农业产值两个参考。

Three-dimensional(3D) Printing Technology Assisted by Minimally Invasive Surgery for Pubic Rami Fractures

Proteo-transcriptomic profiles reveal key regulatory pathways and functions of LDHA in the ovulation of domestic chickens(Gallus gallus)

Background In poultry, the smooth transition of follicles from the preovulatory-to-postovulatory phase impacts egg production in hens and can benefit the poultry industry. However, the regulatory mechanism underlying follicular ovulation in avians is a complex biological process that remains *** Critical biochemical events involved in ovulation in domestic chickens(Gallus gallus) were evaluated by transcriptomics, proteomics, and in vitro assays. Comparative transcriptome analyses of the largest preovulatory follicle(F1) and postovulatory follicle(POF1) in continuous laying(CL) and intermittent laying(IL) chickens indicated the greatest difference between CL_F1 and IL_F1, with 950 differentially expressed genes(DEGs), and the smallest difference between CL_POF1 and IL_POF1, with 14 DEGs. Additionally, data-independent acquisition proteomics revealed 252 differentially abundant proteins between CL_F1 and IL_F1. Perivitelline membrane synthesis, steroid biosynthesis, lysosomes, and oxidative phosphorylation were identified as pivotal pathways contributing to ovulation regulation. In particular, the regulation of zona pellucida sperm-binding protein 3, plasminogen activator, cathepsin A, and lactate dehydrogenase A(LDHA) was shown to be essential for ovulation. Furthermore, the inhibition of LDHA decreased cell viability and promoted apoptosis of ovarian follicles in *** This study reveals several important biochemical events involved in the process of ovulation, as well as crucial role of LDHA. Establishment of a high-resolution 2-D reference map of human spermatozoal proteins from 12 fertile sperm-bank donors

Aim： To extend the analysis of the proteome of human spermatozoa and establish a 2-D gel electrophoresis （2-DE） reference map of human spermatozoal proteins in a pH range of 3.5-9.0. Methods： In order to reveal more protein spots, immobilized pH gradient strips （24 cm） of broad range of pH 3-10 and the narrower range of pH 6-9, as well as different overlapping narrow range pH immobilized pH gradient （IPG） strips, including 3.5-4.5, 4.0-5.0, 4.5-5.5, 5.0-6.0 and 5.5-6.7, were used. After 2-DE, several visually identical spots between the different pH range 2-D gel pairs were cut from the gels and confirmed by mass spectrometry and used as landmarks for computer analysis. Results： The 2-D reference map with pH value from 3.5 to 9.0 was synthesized by using the ImageMaster analysis software. The overlapping spots were excluded, so that every spot was counted only once. A total of 3 872 different protein spots were identified from the reference map, an approximately 3-fold increase compared to the broad range pH 3-10 IPG strip （1 306 spots）. Conclusion： The present 2-D pattern is a high resolution 2-D reference map for human fertile spermatozoal protein spots. A comprehensive knowledge of the protein composition of human spermatozoa is very meaningful in studying dysregulation of male fertility.

Tumor necrosis factorα(TNFα)exhibits diverse biological functions;however,its regulatory roles in myogenesis are not fully *** the present study,we explored the function of TNFαin myoblast proliferation,differentiation,migration,and myotube fusion in primary myoblasts and C2C12 *** this end,we constructed TNFαmuscle-conditional knockout(TNFα-CKO)mice and compared them with flox mice to assess the effects of TNFαknockout on skeletal *** indicated that TNFα-CKO mice displayed phenotypes such as accelerated muscle development,enhanced regenerative capacity,and improved exercise endurance compared to flox mice,with no significant differences observed in major visceral organs or skeletal *** label-free proteomic analysis,we found that TNFα-CKO altered the distribution of several muscle development-related proteins,such as Hira,Casz1,Casp7,Arhgap10,Gas1,Diaph1,Map3k20,Cfl2,and Igf2,in the nucleus and *** set enrichment analysis(GSEA)further revealed that TNFαdeficiency resulted in positive enrichment in oxidative phosphorylation and MyoD targets and negative enrichment in JAK-STAT *** findings suggest that TNFα-CKO positively regulates muscle growth and development,possibly via these newly identified targets and pathways.

Simulation and evaluation study of atmospheric aerosol nonsphericity as a function of particle size

Aerosol nonsphericity causes great uncertainty in radiative forcing assessments and climate *** considerable studies have attempted to quantify this uncertainty,the relationship between aerosol nonsphericity and particle size is usually not considered,thus reducing the accuracy of the *** this study,a coupled inversion algorithm combining an improved stochastic particle swarm optimization algorithm and angular light scattering is used for the nonparametric estimation of aerosol nonsphericity variation with particle size,and the optimal sample selection method is employed to screen the *** on the verification of inversion accuracy,the variation of aerosol aspect ratio with particle size based on the ellipsoidal model in global regions has been obtained from Aerosol Robotic Network(AERONET)data,and the effect of nonsphericity on radiative forcing and dry deposition has been *** results show that the aspect ratio increases with particle size in all regions,with the maximum ranging from 1.4 to 1.8 in the desert,reflecting the differences in aerosol composition at different particle *** radiation calculations,considering aerosol nonsphericity makes the aerosol cooling effect weaker and surface radiative fluxes increase,but hardly changes the aerosol absorption,with maximum differences of 9.22%and 22.12%at the bottom and top of the atmosphere,***,the differences in radiative forcing between aspect ratios as a function of particle size and not varying with particle size are not significant,averaging less than 2%.Besides,the aspect ratio not varying with particle size underestimates the deposition velocity of small particles and overestimates that of large particles compared to that as a function of particle size,with maximum differences of 7%and 4%,respectively.
