BACKGROUND Colon cancer(CC)occurrence and progression are considerably influenced by the tumor ***,the exact underlying regulatory mechanisms remain *** To investigate immune infiltration-related differentially expressed genes(DEGs)in CC and specifically explored the role and potential molecular mechanisms of complement factor I(CFI).METHODS Immune infiltration-associated DEGs were screened for CC using *** reverse transcription polymerase chain reaction was used to examine hub DEGs expression in the CC cell *** CFI-knockdown HT29 and HCT116 cell lines were constructed,and the diverse roles of CFI in vitro were assessed using CCK-8,5-ethynyl-2’-deoxyuridine,wound healing,and transwell *** and eosin staining and immunohistochemistry staining were employed to evaluate the influence of CFI on the tumorigenesis of CC xenograft models constructed using BALB/c male nude *** proteins associated with glycolysis and the Wnt pathway were measured using western *** Six key immune infiltration-related DEGs were screened,among which the expression of CFI,complement factor B,lymphoid enhancer binding factor 1,and SRY-related high-mobility-group box 4 was upregulated,whereas that of fatty acid-binding protein 1,and bone morphogenic protein-2 was ***,CFI could be used as a diagnostic biomarker for ***,CFI silencing inhibited CC cell proliferation,migration,invasion,and tumor ***,CFI knockdown downregulated the expression of key glycolysis-related proteins(glucose transporter type 1,hexokinase 2,lactate dehydrogenase A,and pyruvate kinase M2)and the Wnt pathway-related proteins(β-catenin and c-Myc).Further investigation indicated that CFI knockdown inhibited glycolysis in CC by blocking the Wnt/β-catenin/c-Myc *** The findings of the present study demonstrate that CFI plays a crucial role in CC development by influencing g

Dark Matter Search in Dwarf Irregular Galaxies with Ten Years of Data from the Ice Cube Neutrino Observatory

Dwarf irregular galaxies(dIrrs),as rotationally supported systems,have more reliable J-factor measurements than dwarf spheroidal galaxies and have received attention as targets for dark matter detection in recent *** this paper,we use 10 yr of IceCube muon-track data and an unbinned maximum-likelihood-ratio method to search for neutrino signals beyond the background from the directions of seven dIrrs,aiming to detect neutrinos produced by heavy annihilation dark *** do not detect any significant *** on such null results,we calculate the upper limits on the velocity-averaged annihilation cross section for 1 TeV–10 PeV dark *** limits,although weaker than the strictest constraints in the literature in this mass range,are also a good complement to the existing results considering the more reliable J-factor measurements of dIrrs.

Xinqiao Gully is located in the area of the 2008 Wenchuan M_(s)8.0 earthquake in Sichuan province,*** on the investigation of the 2023"6-26"Xinqiao Gully debris flow event,this study assessed the effectiveness of the debris flow control project and evaluated the debris flow *** field investigation and numerical simulation methods,the indicators of flow intensity reduction rate and storage capacity fullness were proposed to quantify the effectiveness of the engineering measures in the debris flow *** simulation results show that the debris flow control project reduced the flow intensity by41.05%to 64.61%.The storage capacity of the dam decreases gradually from upstream to the mouth of the gully,thus effectively intercepting and controlling the debris *** evaluating the debris flow of different recurrence intervals,further measures are recommended for managing debris flow events.

A novel alternative for pyrogen detection based on a transgenic cell line

Pyrogen,often as a contaminant,is a key indicator affecting the safety of almost all parenteral drugs(including biologicals,chemicals,traditional Chinese medicines and medical devices).It has become a goal to completely replace the in vivo rabbit pyrogen test by using the in vitro pyrogen test based on the promoted‘reduction,replacement and refinement’principle,which has been highly considered by regulatory agencies from different *** used NF-κB,a central signalling molecule mediating inflammatory responses,as a pyrogenic marker and the monocyte line THP-1 transfected with a luciferase reporter gene regulated by NF-κB as an in vitro model to detect pyrogens by measuring the intensity of a fluorescence ***,we show that this test can quantitatively and sensitively detect endotoxin(lipopolysaccharide from different strains)and nonendotoxin(lipoteichoic acid,zymosan,peptidoglycan,lectin and glucan),has good stability in terms of NF-κB activity and cell phenotypes at 39 cell passages and can be applied to detect pyrogens in biologicals(group A&C meningococcal polysaccharide vaccine;basiliximab;rabies vaccine(Vero cells)for human use,freeze-dried;Japanese encephalitis vaccine(Vero cells),inactivated;insulin aspart injection;human albumin;recombinant human erythropoietin injection(CHO Cell)).The within-laboratory reproducibility of the test in three independent laboratories was 85%,80%and 80%and the interlaboratory reproducibility among laboratories was 83.3%,95.6%and 86.7%.The sensitivity(true positive rate)and specificity(true negative rate)of the test were 89.9%and 90.9%,*** summary,the test provides a novel alternative for pyrogen detection.

Development of 400-μW cryogen-free dilution refrigerators for quantum experiments

We have successfully developed cryogen-free dilution refrigerators with medium cooling power that can be applied to quantum experiments. Breakthroughs have been made in some key technologies and components of heat switches and dilution units. Our prototype has been running continuously and stably for more than 100 hours below 10 m K, with a minimum temperature of 7.6 m K and a cooling power of 450 μW at 100 m K. At the same time, we have also made progress in the application of dilution refrigerators, such as quantum computing, low-temperature detector, and magnet integration. These indicators and test results indicate good prospects for application in physics, astronomy, and quantum information.

工业过程运行状态评估方法是对过程当前运行状态进行合理的评价,为工业过程的安全、高效运行提供有益的指导.带钢轧制过程具有流程长且系统层级多等特点,而传统的运行状态评估往往采用集中式的评估方法,难以对轧制过程全流程的运行状态进行合理的评估.针对此问题,提出一种融合分层分块信息的轧制过程运行状态评估方法.采用一种多层级分块的评估策略,将全流程分为若干个层级和子块,提高评估结果的可解释性.提出一种联合核主成分分析(kernel principal component analysis,KPCA)和t-分布随机邻域嵌入算法(t-distributed stochastic neighbor embedding,t-SNE)的特征提取方法,并行地提取全局和局部的特征信息.进而,针对传统支持向量机(support vector machine,SVM)输出结果为硬判型输出,将SVM的输出结果映射为后验概率,并通过D-S证据理论融合多个层级的运行状态评估结果,从而实现决策层面的信息融合,提高评估结果的准确性.最后,将所提出方法应用于实际带钢轧制过程,与各类传统的方法相比评估准确率提高近18%.

河岸缓冲带植被配置对径流水中磷的截留效果

[目的]研究不同配置缓冲带对径流水中磷素截留效果的影响,为选择适宜太湖河岸缓冲带和有效治理富营养化提供参考。[方法]以7块大小为20 m×40 m的太湖人工林河岸缓冲带为研究对象,比较不同宽度(15、30、40 m)、不同植被类型(‘南林95’杨、中山杉林、‘南林95’杨-中山杉混交林)、不同林分密度(400、1000、1600株·hm^(−2))的河岸缓冲带对径流水中总磷和可溶性磷的截留效果的差异。[结果](1)冬季缓冲带对径流水中总磷和可溶性磷的拦截效果明显优于春季缓冲带;缓冲带对径流水中可溶性磷的截留率显著高于总磷。(2)不同的河岸缓冲带对宽度的需求不同。对于中山杉林、混交林、400和1000株·hm^(−2)的杨树林而言,15 m宽的河岸缓冲带基本能满足截磷需求;1600株·hm^(−2)的杨树林需要40 m以上的河岸缓冲带宽度才能满足对磷的截留要求(3)对于中山杉林和杨树林缓冲带而言,林分密度为400株·hm^(−2)的缓冲带对径流水中总磷和可溶性磷的截留效果最好。(4)中山杉林缓冲带对径流水中磷素的截留效果优于中山杉-杨树混交林和杨树林缓冲带,中山杉可作为太湖流域河岸缓冲带构建的首选植物。[结论]15 m宽400株·hm^(−2)的中山杉林缓冲带对径流水中磷素的截留能达到最大效果。

系统评价参芍胶囊联合常规西药治疗冠心病心绞痛的有效性和安全性。计算机检索中国知网(CNKI)、万方(Wanfang)、维普(VIP)、中国生物医学文献服务系统(SinoMed)、EMbase、PubMed、Cochrane Library等7个数据库,从建库至2023年12月关于参芍胶囊治疗冠心病心绞痛的随机对照试验(RCT)。根据纳入、排除标准,对文献进行筛选、资料提取,采用Cochrane偏倚风险评估工具2.0(RoB 2.0)对纳入文献进行质量评价,RevMan 5.4和Stata/SE 15.1软件进行Meta分析,GRADE系统进行证据质量评价,TSA 0.***.5.10 beta软件进行试验序贯分析(TSA)。共纳入RCTs 12篇,样本量1 128例,其中试验组567例,对照组561例。Meta分析显示,参芍胶囊+常规西药vs常规西药可提高临床疗效(RR=1.20,95%CI[1.15,1.26],P<0.000 01)、心电图疗效(RR=1.16,95%CI [1.04,1.30],P=0.01),减少心绞痛周发作频率(MD=-2.85,95%CI[-5.27,-0.43],P=0.02)、心绞痛日发作频率(MD=-0.30,95%CI[-0.57,-0.03],P=0.03)、心绞痛发作持续时间(RR=-2.28,95%CI[-3.44,-1.12],P=0.000 1)。不良反应发生方面,2组差异无统计学意义(RR=1.33,95%CI[0.71,2.51],P=0.37)。TSA显示,临床疗效累计的研究穿过了传统界值,但未穿过TSA界值,表明结果可能存在假阳性。GRADE系统评价显示,除了临床疗效为低级证据,其余均为极低级。结果表明,参芍胶囊+常规西药在提高临床疗效、心电图疗效,降低心绞痛发作频率、心绞痛发作持续时间可能具有一定优势,但因存在的局限性,尚需设计更为严谨、高质量的RCT加以验证其疗效和安全性。
