Self-adjuvant Astragalus polysaccharide-based nanovaccines for enhanced tumor immunotherapy:a novel delivery system candidate for tumor vaccines

The study of tumor nanovaccines(NVs)has gained interest because they specifically recognize and eliminate tumor cells.However,the poor recognition and internalization by dendritic cells(DCs)and insufficient immunogenicity restricted the vaccine efficacy.Herein,we extracted two molecular-weight Astragalus polysaccharides(APS,12.19 k D;APSHMw,135.67 k D)from Radix Astragali and made them self-assemble with OVA257–264directly forming OVA/APS integrated nanocomplexes through the microfluidic method.The nanocomplexes were wrapped with a sheddable calcium phosphate layer to improve stability.APS in the formed nanocomplexes served as drug carriers and immune adjuvants for potent tumor immunotherapy.The optimal APS-NVs were approximately 160 nm with uniform size distribution and could remain stable in physiological saline solution.The FITC-OVA in APS-NVs could be effectively taken up by DCs,and APS-NVs could stimulate the maturation of DCs,improving the antigen cross-presentation efficiency in vitro.The possible mechanism was that APS can induce DC activation via multiple receptors such as dectin-1 and Toll-like receptors 2 and 4.Enhanced accumulation of APS-NVs both in draining and distal lymph nodes were observed following s.***.injection.Smaller APS-NVs could easily access the lymph nodes.Furthermore,APS-NVs could markedly promote antigen delivery efficiency to DCs and activate cytotoxic T cells.In addition,APS-NVs achieve a better antitumor effect in established B16-OVA melanoma tumors compared with the OVA+Alum treatment group.The antitumor mechanism correlated with the increase in cytotoxic T cells in the tumor region.Subsequently,the poor tumor inhibitory effect of APS-NVs on the nude mouse model of melanoma also confirmed the participation of antitumor adaptive immune response induced by NVs.Therefore,this study developed a promising APS-based tumor NV that is an efficient tumor immunotherapy without systemic side effects.

A designed Mg_(88.7)Ni_(6.3)Y_(5)hydrogen storage alloy containing 14H type LPSO(long-period stacking ordered)and ternary eutectic structure was prepared by regulating the alloy composition and casting.The hydrogen storage performance of the alloy was improved by adding nano-flower-like TiO_(2)@C catalyst.The decomposition of the LPSO structure during hydrogenation led to the formation of plenty of nanocrystals which provided abundant interphase boundaries and activation sites.The nanoscale TiO_(2)@C catalyst was uniformly dispersed on the surface of alloy particles,and the"hydrogen overflow''effect of TiO_(2)@C accelerated the dissociation and diffusion of hydrogen on the surface of the alloy particles.As a result,the in-situ endogenous nanocrystals of the LPSO structure decomposition and the externally added flower-like TiO_(2)@C catalyst uniformly dispersed on the surface of the nanoparticles played a synergistic catalytic role in improving the hydrogen storage performance of the Mg-based alloy.With the addition of the TiO_(2)@C catalyst,the beginning hydrogen desorption temperature was reduced to 200℃.Furthermore,the saturated hydrogen absorption capacity of the sample was 5.32 wt.%,and it reached 4.25 wt.%H_(2) in 1 min at 200℃and 30 bar.

In future optical transport networks,lightpath performance analysis is of great practical significance for fully automated management.In general,the quality of transmission(QoT)of lightpaths,measured by optical quality factor or optical signal-to-noise ratio,has a complex time-varying process,along with the interactions of the other lightpath state parameters(LSPs),such as transmit power,chromatic dispersion,polarization mode dispersion.Current studies are mostly focused on lightpath QoT estimation,but ignoring lightpath-level data analytics.In this case,our article proposes a novel lightpath performance analysis method considering recurrence plot(RP)and cross recurrence plot(CRP).Firstly,we give a detailed interpretation on the recurrence patterns of LSPs via a qualitative 2-D RP representation and its quantitative measure.It can potentially enable the accurate and fast lightpath failure detection with a low computational burden.On the other hand,CRP is devoted to modeling the relationships between lightpath QoT and LSPs,and the correlation degree is determined by a geometric mean of multiple indexes of cross recurrence quantification analysis.From the view of application,such CRP analysis can provide the effective knowledge sharing to guarantee more credible QoT prediction.Extensive experiments on a real-world optical network dataset have clearly demonstrated the effectiveness of our proposal.

Impacts of agri-food e-commerce on traditional wholesale industry:Evidence from China

Rapidly expanding studies investigate the effects of e-commerce on company operations in the retail market.However,the interaction between agri-food e-commerce(AEC)and the traditional agri-food wholesale industry(AWI)has not received enough attention in the existing literature.Based on the provincial panel data from 2013 to 2020 in China,this paper examines the effect of AEC on AWI,comprising three dimensions:digitalization(DIGITAL),agrifood e-commerce infrastructure and supporting services(AECI),and agri-food e-commerce economy(AECE).First,AWI and AEC are measured using an entropy-based combination of indicators.The results indicate that for China as a whole,AWI has remained practically unchanged,whereas AEC exhibits a significant rising trend.Second,the findings of the fixed-effect regression reveal that DIGITAL and AECE tend to raise AWI,whereas AECI negatively affects AWI.Third,threshold regression results indicate that AECI tends to diminish AWI with three-stage inhibitory intensity,which manifests as a first increase and then a drop in the inhibition degree.These results suggest that with the introduction of e-commerce for agricultural product circulation,digital development will have catfish effects that tend to stimulate the vitality of the conventional wholesale industry and promote technical progress.Furthermore,the traditional wholesale industry benefits financially from e-commerce even while it diverts part of the traditional wholesale circulation for agricultural products.

Experimental Investigation on Solution Regeneration Performance and Coefficients in Full-Open System for Heat and Water Recovery of Flue Gas

The recovery of heat and water from low-grade flue gas is of considerable importance for energy conservation and environmental preservation.While the full-open absorption heat pump shows promise as a means of achieving heat and water recovery,the lack of research on heat and mass transfer performance of open-type solution evaporation regeneration represents a significant impediment to its design and operation.This paper experimentally investigates the regeneration performance of an open-type spaying tower equipped with ceramic structured packings.Two different regeneration modes are proposed,namely ambient air receiver mode and flue gas receiver mode,to utilize air or low-grade flue gas as a driving source.The impact of different input parameters on the regeneration characteristics,including heat transfer capacity,water removal rate,thermal efficiency,and humidity effectiveness,are demonstrated.The findings indicate that the enhancement of regeneration can be achieved through the increase of solution flow rate,solution temperature,and flue gas flow rate in both regeneration modes.However,high solution concentration and flue gas humidity ratio can weaken water removal rates and reduce thermal efficiency.For the regeneration of CaCl_(2)-H_(2)O with a concentration of55%,flue gas around 200℃with a humidity ratio below 44 g/kg can successfully drive the solution regeneration process.When the solution concentration or flue gas humidity ratio continues to rise,additional energy is necessary for regeneration.Furthermore,the coupled heat and mass transfer coefficients are fitted,which can contribute to the design and optimization of the open-type regenerator.

The function of exogenous alanine(Ala)in regulating biomass accumulation,lipid production,photosynthesis,and respiration in Chlorella pyrenoidosa was studied.Result shows that the supplementation of Ala increased C.pyrenoidosa biomass and lipid production in an 8-d batch culture.The concentration of 10 mmol/L of Ala was optimum and increased the microalgal cell biomass and lipid content by 39.3%and 21.4%,respectively,compared with that in the control(0-mmol/L Ala).Ala supplementation reduced photosynthetic activity while boosting respiratory activity and pyruvate levels,indicating that C.pyrenoidosa used exogenous Ala for biomass accumulation through the respiratory metabolic process.The accelerated respiratory metabolism due to Ala supplementation elevated the substrate pool and improved the lipogenic gene expression,promoting lipid production at last.This study provided a novel method for increasing biomass accumulation and lipid production and elucidated the role of Ala in regulating lipid production.

Affected by climate warming and anthropogenic disturbances, the thermo-mechanical stability of warm and ice-rich frozen ground along the Qinghai-Tibet engineering corridor(QTEC) is continuously decreased, which may delay the construction of major projects in the future. In this study, based on chemical stabilization of warm and icerich frozen ground, the soil-cement column(SCC) for ground improvement was recommended to reinforce the foundations in warm and ice-rich permafrost regions. To explore the validity of countermeasures mentioned above, both the original foundation and the composite foundation consisting of SCC with soil temperature of -1.0℃ were prepared in the laboratory, and then the plate loading tests were carried out. The laboratory investigations indicated that the bearing capacity of composite foundation consisting of SCC was higher than that of original foundation, and the total deformation of original foundation was greater than that of composite foundation, meaning that overall stability of foundation with warm and ice-rich frozen soil can be improved by SCC installation. Meanwhile, a numerical model considering the interface interaction between frozen soil and SCC was established for interpretating the bearing mechanism of composite foundation. The numerical investigations revealed that the SCC within composite foundation was responsible for the more applied load, and the applied load can be delivered to deeper zone in depth due to the SCC installation, which was favorable for improving the bearing characteristic of composite foundation. The investigations provide the valuable guideline for the choice of engineering supporting techniques to major projects within the QTEC.

Based on hourly rain gauge data during May–September of 2016–20,we analyze the spatiotemporal distributions of total rainfall(TR)and short-duration heavy rainfall(SDHR;hourly rainfall≥20 mm)and their diurnal variations over the middle reaches of the Yangtze River basin.For all three types of terrain(i.e.,mountain,foothill,and plain),the amount of TR and SDHR both maximize in June/July,and the contribution of SDHR to TR(CST)peaks in August(amount:23%;frequency:1.74%).Foothill rainfall is characterized by a high TR amount and a high CST(in amount);mountain rainfall is characterized by a high TR frequency but a small CST(in amount);and plain rainfall shows a low TR amount and frequency,but a high CST(in amount).Overall,stations with high TR(amount and frequency)are mainly located over the mountains and in the foothills,while those with high SDHR(amount and frequency)are mainly concentrated in the foothills and plains close to mountainous areas.For all three types of terrain,the diurnal variations of both TR and SDHR exhibit a double peak(weak early morning and strong late afternoon)and a phase shift from the early-morning peak to the late-afternoon peak from May to August.Around the late-afternoon peak,the amount of TR and SDHR in the foothills is larger than over the mountains and plains.The TR intensity in the foothills increases significantly from midnight to afternoon,suggesting that thermal instability may play an important role in this process.

Massive machine type communication aims to support the connection of massive devices,which is still an important scenario in 6G.In this paper,a novel cluster-based massive access method is proposed for massive multiple input multiple output systems.By exploiting the angular domain characteristics,devices are separated into multiple clusters with a learned cluster-specific dictionary,which enhances the identification of active devices.For detected active devices whose data recovery fails,power domain nonorthogonal multiple access with successive interference cancellation is employed to recover their data via re-transmission.Simulation results show that the proposed scheme and algorithm achieve improved performance on active user detection and data recovery.
