Filamentous fungi are one of the major sources of natural products (NPs;which are also termed as secondary metabolites (SMs)) with diverse biological activities,which have been widely used in agriculture,industry,and *** are the second largest species in nature,and their biodiversity implies genomic diversity,which,in turn,predicts the structural diversity of *** general,NPs can be classified as polyketides (PKs),nonribosomal peptides (NRPs),hybrid PK-NRPs,or terpenoids according to the types of their biosynthetic skeleton *** enzymes include polyketide synthases (PKSs),nonribosomal peptide synthetases (NRPSs),hybrid polyketide synthasenonribosomal peptide synthetases (PKS-NRPSs),and terpene synthases or terpene cyclases (TCs).A hallmark in fungal NP biosynthesis is that the relevant genes are usually arranged close to each other on the chromosome in what is called a biosynthetic gene cluster (BGC).Over 2,200 fungal genome sequences are publicly available because of the rapid development of genome sequencing technology and the launch of 1000 Fungal Genome Project by the Joint Genome Institute (JGI).Bioinformatics analyses confirm that the fungal genome has an abundance of *** NP “dark matter” treasure may be present in the fungal genome as over 90% of BGCs are unexplored,silent,or cryptic (He et al.,2018;Keller,2019).These data provide us an opportunity to search for fungal NPs using gene/genome as the starting point.

Serving as the electrical to optical converter,the on-chip silicon light source is an indispensable component of silicon photonic technologies and has long been ***,we briefly review the history and recent progress of a few promising contenders for on-chip light sources in terms of operating wavelength,pump condition,power consumption,and fabrication ***,the performance of each contender is also assessed with respect to thermal stability,which is a crucial parameter to consider in complex optoelectronic integrated circuits(OEICs)and optical ***,III-V-based silicon(Si)lasers formed via bonding techniques demonstrate the best performance and display the best opportunity for commercial usage in the near ***,in the long term,direct hetero-epitaxial growth of III–V materials on Si seems more promising for low-cost,high-yield *** demonstration of high-performance quantum dot(QD)lasers monolithically grown on Si strongly forecasts its feasibility and enormous potential for on-chip *** superior temperature-insensitive characteristics of the QD laser promote this design in large-scale high-density *** Germanium(Ge)-on-Si laser is also competitive for large-scale monolithic integration in the *** with a III-V-based Si laser,the biggest potential advantage of a Ge-on-Si laser lies in its material and processing compatibility with Si ***,the versatility of Ge facilitates photon emission,modulation,and detection simultaneously with a simple process complexity and low cost.

Comparison and quantitative assessment of two regional soil erosion survey approaches

Regional soil erosion surveys are the first step of soil conservation *** and sampling ap-proaches are commonly used for soil erosion surveys at the regional *** the grid approach,the region is divided into grids(e.g.,1 km × 1 km),and the soil erosion rate of each grid was *** the sampling approach,a small portion of the area(typically 4%or less)was taken and then the soil erosion rate was *** on the above calculation,both approaches evaluated the soil erosion area(which is the area where the soil erosion rate is greater than the soil loss tolerance(t))and its distribution in the whole *** purpose of this paper is to compare the assessment results of the two approaches and analyse their practicality using Yunyang County,Chongqing,China(area is 3636 km^(2)).The soil erosion rate,percentage of soil erosion area(PSEA,which is the ratio of the area where the soil erosion rate is greater than the soil loss tolerance(t)to the area of the total study region)and its spatial distribution were compared between the two *** results showed that the grid approach overestimated the average soil erosion rate and PSEA overall for all 33 sample units and the whole region in comparison to the sampling *** sampling approach can yield a reasonable spatial distribution of the soil erosion rate in the whole *** addition,the soil erosion regions were more clustered than those using the grid approach,which can be more suitable for soil conservation ***,the sampling approach is an efficient and practical approach in regional soil erosion *** results can provide insights into regional soil erosion surveys.

Laccases, multicopper oxidoreductases, are mainly produced in white-rot fungi and are considered as ideal green catalysts in industrial and biotechnological applications. However, the development of laccases is limited due to the slow growth of natural laccase producing strains and the low expression levels of laccases. In this study, we designed three regulation strategies for laccase gene expression in the model fungus Aspergillus nidulans. By introducing various promoters in front of the laccase gene pslcc from the white-rot fungus Pycnoporus sanguineus, we found that the laccase gene with the original promoter had effective expression in A. nidulans. Using the previously identified transcription factor RsmA regulatory mechanism, the aflR promoter was inserted into the pslcc expression vectors, and the laccase production was 15-fold higher in the strain overexpressing of RsmA compared to the control strain. To improve the laccase yield, the dipeptidyl-peptidase DppV, aspartic protease PepA and mannosyltransferase Mnn9 were successfully deleted in the A. nidulans host. The laccase activities were increased approximately 8-fold and 13-fold in the double deletions strains of Δmnn9ΔpepA and ΔdppVΔpepA over the control strains, *** together, these results not only demonstrate an efficient system for heterologous protein production in the model fungus *** but also provide a general approach to applying regulatory methods to control gene expression.

Comparative study of galectin-3 and B-type natriuretic peptide as biomarkers for the diagnosis of heart failure

Background Heart failure （HF） is a common disease with complex pathophysiological causes. The diagnosis of HF commonly relies on comprehensive analyses of medical history and symptoms, and results from echocardiography and biochemical tests. Galectin-3, a rela-tively new biomarker in HF, was approved by the US Food and Drug Administration in 2010 as a marker in the stratification of risk for HF. We assessed galectin-3 as a biomarker for HF diagnosis in patients with preserved ejection fraction （pEF） and compared its performance with that of B-type natriuretic peptide （BNP）. Methods Thirty-five pEF patients with HF （HFpEF group） and 43 pEF patients without HF （control group） were enrolled. Plasma levels of galectin-3 and BNP in HFpEF and control subjects were determined. Sensitivity, specificity, pre dictive values, and accuracy of galectin-3 and BNP as markers for HF diagnosis were calculated and compared. Results Levels of galec- tin-3 and BNP were 23.09 ±6.97 ng/mL and 270.46 ± 330.41 pg/mL in the HFpEF group, and 16.74 ± 2.75 ng/mL and 59.94 ± 29.93 pg/mL in the control group, respectively. Differences in levels of galectin-3 and BNP between the two groups were significant （P 〈 0.01）. As a bio- marker for HF diagnosis in study subjects, galectin-3 showed sensitivity and specificity of 94.3% and 65.1%, respectively, at a cutoff value of 17.8 ug/mL. BNP showed sensitivity and specificity of 77.1% and 90.7%, respectively, at a cutoff value of 100 pg/mL. Galectin-3 was a significantly more sensitive （P 〈 0.05） but less specific （P 〈 0.01） biomarker compared with BNP. Differences in positive predictive value, negative predictive value, and accuracy between galectin-3 and BNP markers were not significant （P 〉 0.05）. Areas under the receiver operating characteristic curve （95% confidence interval） were 0.891 （0.808-0.974） and 0.896 （0.809-0.984） for galectin-3 and BNP, respec- tively, with no significant difference between the two values （P 〉 0.05）. We develop a new method to prepare micropowders with a combination of jet-milling and electrostatic dispersion *** dispersiveness of the powder can be obviously improved by charging the particles during the process of *** carbonate(CaCO_(3))powders with high dispersion are prepared by using this method from two different initial particle sizes(10.93 and 25.43𝜈m).The experimental studies and theoretical analysis about the effects of preparation parameter on dispersiveness of the powder are investigated,showing that the jet-milling/electrostatic dispersion(𝐾J/E𝐹)is a considerably effective way to prepare micropowder in ambient *** is found that the strength of electrostatic field and particle radius of the raw powder strongly affect the *** average particle size of both powders decreases with the increase in charging voltage while the reduction of particle size is more obvious in the powder with larger initial particle size.

Polymer/metal composite segmental Janus nanoparticles （NPs） are synthesized by sequential growth against poly（4-vinylpyridine）（P4VP） crosslinked cP4VP-PS Janus NPs. A Janus cluster of poly（4-vinylpyridine）-block-polystyrene （P4VP-b-PS） diblock copolymer is self-organized after absorption onto a silica patchy sphere via hydrogen bonding. Selective crosslinking of P4VP leads to the formation of robust cP4VP-PS Janus NPs. Within the cP4VP domain, functional species such as metals are preferentially grown by in situ reduction. Other thiol-capped polymers, for example, thiol-capped poly（N-isopropylacrylamide）（PNIPAM-SH）, can be conjugated onto the opposite side to form polymer/metal triple segmental Janus NPs. The hyperthermia effect ofAu NP of PNIPAM-Au@cP4VP-PS by near infrared （NIR） irradiation can trigger a fast transition from amphiphilic to hydrophobic of the Janus NPs at low surrounding temperature. De-stabilization of the emulsion is NIR triggered although the system temperature is below LCST （-32 ℃）.
