TALEN-Mediated Homologous Recombination Produces Site-Directed DNA Base Change and Herbicide-Resistant Rice

Over the last decades,much endeavor has been made to advance genome editing technology due to its promising role in both basic and synthetic *** breakthrough has been made in recent years with the advent of sequence-specific endonucleases,especially zinc finger nucleases（ZFNs）,transcription activator-like effector nucleases（TALENs） and clustered regularly interspaced short palindromic repeats（CRISPRs） guided nucleases（e.g.,Cas9）.In higher eukaryotic organisms,site-directed mutagenesis usually can be achieved through non-homologous end-joining（NHEJ） repair to the DNA double-strand breaks（DSBs） caused by the exogenously applied ***,site-specific gene replacement or genuine genome editing through homologous recombination（HR） repair to DSBs remains a *** a proof of concept gene replacement through TALEN-based HR in rice（Oryza sativa）,we successfully produced double point mutations in rice acetolactate synthase gene（OsALS） and generated herbicide resistant rice lines by using TALENs and donor DNA carrying the desired *** ballistic delivery into rice calli of TALEN construct and donor DNA,nine HR events with different genotypes of OsALS were obtained in T_0 generation at the efficiency of 1.4%—6.3%from three *** HRmediated gene edits were heritable to the progeny of T_1 *** edited T_1 plants were as morphologically normal as the control plants while displayed strong herbicide *** results demonstrate the feasibility of TALEN-mediated genome editing in rice and provide useful information for further genome editing by other nuclease-based genome editing platforms.

Synthesis and properties of electrodeposited Ni-CeO_2 nano-composite coatings

Current machinery requires metallic materials to have better surface properties. Based on an orthogonal experimental design and analysis method, the CeO2-rein- forced nickel nano-composite coatings were prepared by direct current electrodeposition in a nickel sulfate bath containing CeO2 nanoparticles. Statistical results indicate that current density is the most significant variable in the electrodeposition processing, while temperature is the least important factor. The microstructure of Ni and Ni-CeO2 nano-composite coatings was characterized by scanning electron microscopy （SEM） equipped with energy-disper- sive spectroscopy （EDS）, and X-ray diffraction （XRD）. The microhardness of the Ni coating is enhanced by the incorporation of CeO2 nanoparticles. Potentiodynamic polarization and electrochemical impedance spectroscopy （EIS） were used to characterize the corrosion behavior of Ni and Ni-CeO2 coatings. These studies show that Ni- CeOz coating has better corrosion resistance compared to Ni coating.

Electrostatic capacitors based on dielectrics with high energy density and efficiency are desired for modern electrical systems owing to their intrinsic fast charging-discharging speed and excellent *** longstanding bottleneck is their relatively small energy ***,we report enhanced energy density and efficiency in the Aurivillius Pb_(2)Bi_(4)Ti_(5)O_(18)films by controlling the post-annealing *** results demonstrate that the fabrication atmosphere has significant effects on the film texture and *** the increase of the oxygen pressure of annealing atmosphere,the Pb_(2)Bi_(4)Ti_(5)O_(18)films show a preferred growth orientation of(00l)and fewer defects,which leads to a higher polarization and breakdown field for the film annealed in air atmosphere and thus help to achieve an ultrahigh energy density of59.4 J·cm^(-3)and an improved efficiency of 81.2%.Moreover,the film also exhibits excellent cycling reliability and good thermal *** Pb_(2)Bi_(4)Ti_(5)O_(18)films show a significant potential application for dielectric capacitors.

Optimisation Method for Determination of Crack Tip Position Based on Gauss-Newton Iterative Technique

In the digital image correlation research of fatigue crack growth rate,the accuracy of the crack tip position determines the accuracy of the calculation of the stress intensity factor,thereby affecting the life *** paper proposes a Gauss-Newton iteration method for solving the crack tip *** conventional linear fitting method provides an iterative initial solution for this method,and the preconditioned conjugate gradient method is used to solve the ill-conditioned matrix.A noise-added artificial displacement field is used to verify the feasibility of the method,which shows that all parameters can be solved with satisfactory *** actual stress intensity factor solution case shows that the stress intensity factor value obtained by the method in this paper is very close to the finite element result,and the relative error between the two is only−0.621%;The Williams coefficient obtained by this method can also better define the contour of the plastic zone at the crack tip,and the maximum relative error with the test plastic zone area is−11.29%.The relative error between the contour of the plastic zone defined by the conventional method and the area of the experimental plastic zone reached a maximum of 26.05%.The crack tip coordinates,stress intensity factors,and plastic zone contour changes in the loading and unloading phases are *** results show that the crack tip change during the loading process is faster than the change during the unloading process;the stress intensity factor during the unloading process under the same load condition is larger than that during the loading process;under the same load,the theoretical plastic zone during the unloading process is higher than that during the loading process.

Extended social force model with a dynamic navigation field for bidirectional pedestrian flow

An extended social force model with a dynamic navigation field is proposed to study bidirectional pedestrian movement. The dynamic navigation field is introduced to describe the desired direction of pedestrian motion resulting from the decision-making processes of pedestrians. The macroscopic fundamental diagrams obtained using the extended model are validated against camera-based observations. Numerical results show that this extended model can reproduce collective phenomena in pedestrian traffic, such as dynamic multilane flow and stable separate-lane flow. Pedestrians＇ path choice behavior significantly affects the probability of congestion and the number of self-organized lanes.

Additive manufacturing of Cu-AI-Mn shape memory alloy with enhanced superelasticity

The Cu-based shape memory alloy(SMA)with highly oriented columnar crystals is an ideal candidate for the commercial application,especially the ones obtained through rapid cooling via additive manufacturing *** this work,Cu_(71)Al_(18)Mn_(11)(at%)shape memory alloy with strongtexture columnar grains was successfully prepared by selective laser melting(SLM).An L27(313)orthogonal array was designed to systematically investigate the effects of laser power,scanning speed,scanning spacing,layer thickness and their interactions on the forming quality of Cu_(71)Al_(18)Mn_(11)***_(71)Al_(18)Mn_(11)alloys with density of 7.3204 g·cm^(-3)and relative density of 99.18%were successfully prepared when the laser power,scanning speed,scanning distance and layer thickness were 240 W,1000 mm·s^(-1),0.11 mm and 25μm,the transformation onset temperature(Ms),martensite phase transformation termination temperature(Mf),austenite phase transformation onset temperature(AS)and austenite phase transformation termination temperature(Af)are-21.84,-26.04,-15.75 and-12.36℃,*** compression strength and fracture strain along the building direction(BD)were significantly superior to the scanning direction(SD),while the superelasticity of compression along the SD reached 2.50%,which was better than that of2.32%along *** mechanical property and superelasticity anisotropy due to the formation of columnar grains and texture were *** study shows that SLM is a proposed method for the preparation of Cu-Al-Mn SMAs with high superelasticity,which provides a new strategy for enhancing the shape memory alloy superelasticity.

Shear testing on rock tunnel models under constant normal stress conditions

Large shear deformation problems are frequently encountered in geotechnical *** expose the shear failure mechanism of rock tunnels,compression-shear tests for rock models with circular tunnel were carried out,including single tunnel and adjacent double *** failure process is recorded by the external video and miniature cameras around the tunnel,accompanied by real-time acoustic emission *** experiments indicate that the shearing processes of rock tunnel can be divided into four steps:(i)cracks appeared around tunnels,(ii)shear cracks and spalling ejection developed,(iii)floor warping occurred,and(iv)shear cracks ran through the tunnel ***,the roughness of the sheared fracture surface decreased with the increase in normal *** numerical simulation indicates that there are tensile stress concentrations and compressive stress concentrations around the tunnel during the shearing process,while the compressive stress concentration areas are under high risk of failure and the existence of adjacent tunnels will increase the degree of stress concentration.

Volume transport in the East Taiwan Channel in response to diff erent tracks of typhoons as revealed by HYCOM data

Northward infl ow through the East Taiwan Channel is vital in modulation of water exchange processes off northeastern Taiwan,*** addition to the eff ects of the Kuroshio Current and westward-propagating oceanic mesoscale eddies,the seas off eastern Taiwan,China,are frequently infl uenced by *** on extrema of East Taiwan Channel volume transport(ETCVT)that appear within days of typhoon infl uence,this study investigated 124 historical cyclones including 91 typhoons that passed over the study sea area off eastern Taiwan,*** on 25-year HYbrid Coordinate Ocean Model(HYCOM)data,71%of short-term(within 10 d)ETCVT absolute values with>5 Sv occurred under the infl uence of typhoons crossing the study sea area,and the maximum short-term ETCVT extrema induced by typhoons were 12.5 and-10.9 *** ETCVT extrema induced by typhoons showed reasonable positive correlation with typhoon wind *** importantly,the ETCVT extrema diff ered in response to diff erent typhoon *** types of typhoon were identifi ed based on their track and impact on *** typhoon cases were examined to elucidate the specifi cs of each typical *** on historical best track data and HYCOM data,it was established that Type I typhoons caused the ETCVT to exhibit a negative extremum followed by a positive *** Type I,II,and III typhoons tended to result in typical ETCVT fl uctuations in the surface mixing layer above the depth of 50-100 m,while Type II typhoons were more likely to induce ETCVT fl uctuations in the subsurface *** fi ndings of this study enhance understanding of ETCVT extrema that occur following typhoon passage,which is valuable for short-term physical-biogeochemical studies both in the study region and in areas downstream owing to the large net volume transport changes induced by typhoons.

Modified Model of Crack Tip Stress Field Considering Dislocation Slip Accumulation and Crack Tip Blunting

This study uses the digital image correlation technique to measure the crack tip displacement field at various crack lengths in U71MnG rail steel,and the interpolated continuous displacement field was obtained by fitting with a back propagation(BP)neural *** slip and stacking of dislocations affect crack initiation and growth,leading to changes in the crack tip field and the fatigue characteristics of crack *** Christopher-James-Patterson(CJP)model describes the elastic stress field around a growing fatigue crack that experiences plasticity-induced *** the present work,this model is modified by including the effect of the dislocation field on the plastic zone of the crack tip and hence on the elastic field by introducing a plastic flow factorρ,which represents the amount of blunting of the crack *** Levenberg-Marquardt(L-M)nonlinear least squares method was used to solve for the stress intensity *** verify the accuracy of this modified CJP model,the theoretical and experimental plastic zone errors before and after modification were compared,and the variation trends of the stress intensity factors and the plastic flow factorρwere *** results show that the CJP model,with the introduction ofρ,exhibits a good blunting *** the low plasticity state,the modified model can accurately describe the experimental plastic zone,and the modified stress intensity factors are more accurate,which proves the effectiveness of dislocation *** plastic flow correction provides a more accurate crack tip field model and improves the CJP crack growth relationship.

Aqueous-solution-driven HfGdO_x gate dielectrics for low-voltage-operated α-InGaZnO transistors and inverter circuits

In this work,a non-toxic and environmentally friendly aqueous-solution-based method has been adopted to prepare gadolinium-doped hafnium oxide(HfO2) gate dielectric thin *** adjusting the gadolinium(Gd) doping concentration,the oxygen vacancy content,band offset,interface trap density,and dielectric constant of HfGdOx(HGO) thin films have been *** have confirmed that HGO thin films with Gd doping ratio of 15 at.% have demonstrated appropriate dielectric constant of 27.1 and lower leakage current density of 5.8×10-9 A *** indium-gallium-zinc oxide(α-IGZO) thin film transistors(TFTs) based on HGO thin film(Gd:15 at.%) as gate dielectric layer have exhibited excellent electrical performance,such as larger saturated carrier mobility(μsat) of 20.1 cm2 V-1 S-1,high on/off current ratio(Ion/Ioff) of ~108,smaller sub-threshold swing(SS) of 0.07 V decade-1,and a negligible threshold voltage shift(ΔVTH) of 0.08 V under positive bias stress(PBS) for 7200 *** confirm its potential application in logic circuit,a resistor-loaded inverter based on HGO/α-IGZO TFTs has been constructed.A high voltage gain of 19.8 and stable full swing characteristics have been *** a result,it can be concluded that aqueous-solution-driven HGO dielectrics have potential application in high resolution flat panel displays and ultra-large-scale integrated logic circuits.
