Dipeptidyl peptidase-4 inhibitors and the risk of infection: A systematic review and meta-analysis of cardiovascular outcome trials

BACKGROUND Since adverse events during treatment affect adherence and subsequent glycemic control,understanding the safety profile of oral anti-diabetic drugs is imperative for type 2 diabetes mellitus(T2DM)therapy.AIM To evaluate the risk of infection in patients with T2DM treated with dipeptidyl-peptidase 4(DPP-4)inhibitors.METHODS Electronic databases were searched.The selection criteria included randomized controlled trials focused on cardiovascular outcomes.In these studies,the effects of DPP-4 inhibitors were directly compared to those of either other active anti-diabetic treatments or placebo.Six trials involving 53616 patients were deemed eligible.We calculated aggregate relative risks employing both random-effects and fixed-effects approaches,contingent upon the context.RESULTS The application of DPP-4 inhibitors showed no significant link to the overall infection risk[0.98(0.95,1.02)]or the risk of serious infections[0.96(0.85,1.08)],additionally,no significant associations were found with opportunistic infections[0.69(0.46,1.04)],site-specific infections[respiratory infection 0.99(0.96,1.03),urinary tract infections 1.02(0.95,1.10),abdominal and gastrointestinal infections 1.02(0.83,1.25),skin structure and soft tissue infections 0.81(0.60,1.09),bone infections 0.96(0.68,1.36),and bloodstream infections 0.97(0.80,1.18)].CONCLUSION This meta-analysis of data from cardiovascular outcome trials revealed no heightened infection risk in patients undergoing DPP-4 inhibitor therapy compared to control cohorts.

Objective:In B-cell acute lymphoblastic leukemia(B-ALL),current intensive chemotherapies for adult patients fail to achieve durable responses in more than 50%of cases,underscoring the urgent need for new therapeutic regimens for this patient population.The present study aimed to determine whether HZX-02-059,a novel dual-target inhibitor targeting both phosphatidylinositol-3-phosphate 5-kinase(PIKfyve)and tubulin,is lethal to B-ALL cells and is a potential therapeutic for B-ALL patients.Methods:Cell proliferation,vacuolization,apoptosis,cell cycle,and in-vivo tumor growth were evaluated.In addition,Genome-wide RNA-sequencing studies were conducted to elucidate the mechanisms of action underlying the anti-leukemia activity of HZX-02-059 in B-ALL.Results:HZX-02-059 was found to inhibit cell proliferation,induce vacuolization,promote apoptosis,block the cell cycle,and reduce in-vivo tumor growth.Downregulation of the p53 pathway and suppression of the phosphoinositide 3-kinase(PI3K)/AKT pathway and the downstream transcription factors c-Myc and NF-κB were responsible for these observations.Conclusion:Overall,these findings suggest that HZX-02-059 is a promising agent for the treatment of B-ALL patients resistant to conventional therapies.

Effect of saline water ionic strength on phosphorus recovery from synthetic swine wastewater

Declining worldwide phosphate rock reserves has driven a growing interest in exploration of alternative phosphate supplies. This study involved phosphorus recovery from swine wastewater through precipitation of struvite, a valuable slow-release fertiliser. The economic feasibility of this process is highly dependent on the cost of magnesium source. Two different magnesium sources were used for phosphorus recovery: pure magnesium chloride and nanofiltration(NF) saline water retentate. The paper focuses on the impact of ionic strength on phosphorus recovery performance that has not been reported elsewhere. Experimental design with five numerical variables(Mg/P molar ratio, pH, PO_(4)^(3-)-P, NH_(4)^(+)-N, and Ca^(2+) levels) and one categorical variable(type of magnesium source) was used to evaluate the effect of ionic strength on phosphorus removal and struvite purity. The experimental data were analysed using analysis of variance(ANOVA) and principal component analysis(PCA). Results indicated that a magnesium source obtained from NF retentate was as effective as MgCl_(2) for struvite precipitation. It was also revealed that ionic strength had a more positive effect on struvite purity than on phosphorus removal. Within the range of parameters studied in this research, high ionic strength, high p H and wastewater with high phosphate, high ammonium and low calcium contents were found to be the most favourable conditions for struvite precipitation. Findings from this study will be beneficial to determine the feasibility of using high ionic strength saline water, such as NF seawater retentate, as a magnesium source for phosphorus recovery from wastewater that is rich in ammoniumnitrogen and phosphate.

Dynamic tracking of exsolved PdPt alloy/perovskite catalyst for efficient lean methane oxidation

Supported Pd based catalysts are considered as the efficient candidates for low-carbon alkane oxidation for their outstanding capability to break C-H bond. Whereas, the irreversible deactivation of Pd based catalysts was still frequently observed. Herein, we reinforced the extruded Pd nanoparticles with quantitive Pt to assemble the evenly distributed Pd Pt nanoalloy onto ferrite perovskite(Pd Pt-LCF) matrix with strengthened robustness of metal/oxide support interface. We further co-achieved the enhanced performance, anti-overoxidation as well as resistance of vapor-poisoning in durability measurement. The operando X-ray photoelectron spectroscopy(O-XPS) combined with various morphology characterizations confirms that the accumulation of surface deep-oxidation species of Pd^(4+) is the culprit for fast activity loss in exsolved Pd system, especially at high temperature of 400 ℃. Conversely, it could be completely suppressed by in-situ alloying Pd with equal amount of Pt, which helps maintain the metastable Pd^(2+)/Pd shell and metallic solid-solution core structure. The density function theory(DFT) calculations further buttress that the dissociation of C–H was facilitated on alloy/perovskite interface which is, on the contrary, resistant toward O–H bond cleavage, as compared to Pd/perovskite. Our work suggests that the modification of exsolved metal/oxide catalytic interface could further enrich the toolkit of heterogeneous catalyst design.

Study on tungsten sintering temperature field based on electromagnetic simulation and computational fluid dynamics

In order to study the influence of various sintering variables more intuitively and clearly,electromagnetic field analysis and computational fl uid dynamics are used to analyze the infl uence of gas flow,heating power and other process parameters on the temperature range of medium-frequency sintering in this paper.The results indicate that the uniformity of temperature fi eld is determined by the gas flow rate and heating power.When the heating power is 50 kW,the stable temperature drops about 10 K for every 1 m^(3)·h^(−1)increase of hydrogen flow.The peak value of the maximum temperature diff erence on the tungsten rods gradually increases with the increase of the hydrogen flow rate,and it appears slightly later.The temperature distribution of tungsten rods in furnace is similar with diff erent hydrogen flow rate.At the same time,the closer to the furnace wall,the higher the temperature of the tungsten rod.The temperature diff erence between tungsten rods mainly occurred within 1 h after the beginning of heating.Increasing the heating power can improve the stable temperature,but has little eff ect on the heating time.Increasing heating power can increase the peak value of the maximum temperature diff erence on the tungsten rods,but has little eff ect on the occurrence time of the peak value of the maximum temperature diff erence on the tungsten rods.Increasing the heating power will increase the temperature diff erence of a single tungsten rod,but the temperature diff erence of a single tungsten rod never exceeds 65 K.This study can be used to guide the process optimization design.Combined with numerical simulation calculation,the sintering parameters with low energy consumption and high temperature uniformity can be quickly and eff ectively found,thus improving the microstructure and properties of sintered tungsten products.

Efficient adsorption separation of methane from C_(2)-C_(3) hydrocarbons in a Co(Ⅱ)-nodes metal-organic framework

Methane(CH_(4))as a substitute for other mineral fuels plays a crucial role in reducing energy consumption and preventing environmental pollution.The present study employs a solvothermal method to fabricate a porous framework Co-metal-organic framework(Co-MOF)containing two distinct secondary building units(SBUs):an anionic[Co_(2)(μ_(2)-OH)(COO)_(4)(H_(2)O)]and a neutral[CoN_(2)(COO)_(2)].Notably,within the anionic SBUs,the coordinated water molecules induce the generation of divergent unsaturated Co(Ⅱ)centers in the unidirectional porous channels,thereby creating open metal sites.The adsorption performance of Co-MOF towards pure component gases was systematically investigated.The results demonstrated that Co-MOF exhibits superior adsorption capacity for C_(2)-C_(3) hydrocarbons compared to CH_(4),which offers the potential for efficient adsorption and separation of CH_(4) from C_(2)-C_(3) hydrocarbons.The gas selectivity separation ratios of Co-MOF for C_(2)H_(6)/CH_(4) and C_(3)H_8/CH_(4) were calculated using the ideal adsorbed solution theory method at 273/298 K and 0.1 MPa.The results revealed that Co-MOF achieved remarkable equilibrium separation selectivity for CH_(4) and C_(2)-C_(3) hydrocarbon gases among non-modified MOFs,signifying the potential of the synthesized Co-MOF for efficient recovery and purification of CH_(4) from C_(2)-C_(3) hydrocarbons.Breakthrough experiments further demonstrate the ability of Co-MOF to purify methane from C_(2)-C_(3) hydrocarbons in practical gas separation scenarios.Additionally,molecular simulation calculations further substantiate the propensity of anionic SBUs to interact with C_(2)-C_(3) hydrocarbon compounds.This study provides a novel paradigm for the development of porous MOF materials in the application of gas mixture separation.

An integrated north–south paleo-Dadu-Anning River: New insights from bulk major and trace element analyses of the Xigeda Formation

The Xianshuihe-Anninghe fault extends SE–S and constitutes the southeastern margin of the Tibetan Plateau.However,the Dadu River which is associated with the fault does not flow following the path,but makes a 90ºturn within a distance of 1 km at Shimian,heading east,and joins the Yangtze River,finally flowing into the East China Sea.Adjacent to the abrupt turn,a low and wide pass near the Daqiao reservoir at Mianning separates the N–S course of the Dadu River from the headwater of the Anning River which then flows south into the Yunnan Province along the Anninghe fault.Therefore,many previous studies assumed southward flow of the paleo-Dadu River from the Shimian to the Anning River.However,evidences for the capture of the integrated N–S paleo-Dadu-Anning River,its timing,and causes are still insufficient.This study explored the paleo-drainage pattern of the Dadu and Anning Rivers based on bulk mineral and geochemical analyses of the large quantities of fluvial/lacustrine sediments along the trunk of the Dadu and Anning Rivers.Similar with sands in the modern Dadu River,the Xigeda sediments also exhibit a granitoid affinity with the bulk major mineral compositions of quartz(>50%),anorthite(about 10%),orthoclase(about 5%),muscovite(about 5%),and clinochlore(about 4%).Correspondingly,bulk major elements show high SiO_(2),with all samples>60%,and some of them>70%,low TiO_(2)(≤0.75%),P_(2)O_(5)(≤0.55%),FeO*(≤5%),and relatively high CaO(1.02%–8.51%),Na_(2)O(1.60%–2.52%),and K_(2)O(2.17%–2.71%),with a uniform REE patterns.Therefore,synthesizing all these results indicate that these lacustrine sediments have similar material sources,which are mainly derived from its course in the Songpan-Ganzi flysch block,implying that the paleo-Dadu originally flowed southward into the Anning River and provided materials to the Xigeda ancient lake.The rearrangement of the paleo-Dadu River appears to be closely related to the locally focused uplift driven by strong activities of the XianshuiheXiaojiang fault system.

The multiple PDZ domain crumbs cell polarity complex component gene(MPDZ;MIM:603785),is highly expressed in the brain across the whole lifespan.It encodes the multiple PDZ domain protein,which is a member of the NMDAR signaling complex that may play a role in the control of AMPAR potentiation and synaptic plasticity in excitatory synapses."Previously,MPDZ variants have been demonstrated to be associated with autosomal recessive congenital hydrocephalus-2(HYC2;MIM:615219)which is commonly complicated by brain abnormalities and developmental delay.Seizures were reported in only one case.The association between MPDz and epilepsy requires clarification.

A four-band index for both liquid and solid water and its applications in the Aral Sea Basin

Various investigations have been conducted to analyze the water-coverage area of the Aral Sea and the Aral Sea Basin(ASB). However, the investigations incorporated considerable uncertainty and the used water indices had misclassification problem, which made different research groups present different results. Thus we first ascertain the boundaries of the ASB, the Syr and Amu river basins as well as their upper, middle and lower reaches. Then a four-band index for both liquid and solid water(ILSW) is proposed to address the misclassification problems of the classic water indices. ILSW is calculated by using the reflectance values of the green, red, near infrared, and thermal infrared bands, which combines the normalized difference water index(NDWI) and land surface temperature(LST) together. Validation results show that the ILSW water index has the highest accuracy by far in the Aral Sea Basin. Our results indicate that annual average decline of the water-coverage area was 963 km^(2) in the southern Aral Sea, whereas the northern Aral Sea has experienced little change. In the meanwhile, permanent ice and snow in upper reach of ASB has retreated considerably. Annual retreating rates of the permanent ice and snow were respectively 6233and 3841 km^(2) in upper reaches of Amu river basin(UARB) and Syr river basin(USRB). One of major reasons is that climate has become warmer in ASB. The climate change has caused serious water deficit problem. The water deficit had an increasing trend since the 1990s and its increasing rates was 3.778 billion m^(3) yearly on average. The total water deficit was 76.967 billion m^(3) on average in the whole area of ASB in the 2010s. However, up reaches of Syr river basin(USRB), a component area of ASB, had water surplus of 25.461 billion m^(3). These conclusions are useful for setting out a sustainable development strategy in ASB.

Genomic rearrangements play a significant role in disease, evolution, and tumorigenesis(Mani and Chinnaiyan,2010). While single nucleotide polymorphisms(SNPs) and small insertions/deletions contribute substantially to genetic variation, discerning the independent impact of structural variations poses a challenge. Additionally, experimental induction and control of structural variations present difficulties that hinder further investigation and exploitation.
