Background and Aims:Age-related mosaic chromosomal alterations(mCAs)detected from genotyping of blood-de-rived DNA are structural somatic variants that indicate clonal *** study aimed to investigate whether mCAs contribute to the risk of cirrhosis and modify the effect of a polygenic risk score(PRS)on cirrhosis risk ***:mCA call sets of individuals with European ancestry were obtained from the UK *** PRS was constructed based on 12 susceptible single-nucleotide poly-morphisms for *** proportional hazard models were applied to evaluate the associations between mCAs and cirrhosis ***:Among 448,645 individuals with a median follow-up of 12.5 years,we identified 2,681 cas-es of cirrhosis,1,775 cases of compensated cirrhosis,and 1,706 cases of decompensated *** to non-carriers,individuals with copy-neutral loss of heterozygosity mCAs had a significantly increased risk of cirrhosis(hazard ratio(HR)1.42,95%confidence interval(CI)1.12-1.81).This risk was higher in patients with expanded cell fractions of mCAs(cell fractions≥10%*** fractions<10%),especially for the risk of decompensated cirrhosis(HR 2.03[95%CI 1.09-3.78]vs.1.14[0.80-1.64]).In comparison to non-carriers of mCAs with low genetic risk,individuals with expanded copy-neutral loss of heterozygosity and high genetic risk showed the highest cirrhosis risk(HR5.39[95%CI 2.41-12.07]).Conclusions:The presence of mCAs is associated with increased susceptibility to cirrhosis risk and could be combined with PRS for personalized cirrhosis risk stratification.

Introduction:Severe fever with thrombocytopenia syndrome(SFTS)is an emerging infectious disease caused by the SFTS virus,which has a high mortality *** the number of SFTS cases is essential for early outbreak warning and can offer valuable insights for establishing prevention and control ***:In this study,data on monthly SFTS cases in Hubei Province,China,from 2013 to 2020 were *** time series models based on seasonal auto-regressive integrated moving average(SARIMA),Prophet,eXtreme Gradient Boosting(XGBoost),and long short-term memory(LSTM)were developed using these historical data to predict SFTS *** established models were evaluated and compared using mean absolute error(MAE)and root mean squared error(RMSE).Results:Four models were developed and performed well in predicting the trend of SFTS *** XGBoost model outperformed the others,yielding the closest fit to the actual case numbers and exhibiting the smallest MAE(2.54)and RMSE(2.89)in capturing the seasonal trend and predicting the monthly number of SFTS cases in Hubei ***:The developed XGBoost model represents a promising and valuable tool for SFTS prediction and early warning in Hubei Province,China.

Atorvastatin, etanercept and the nephrogenic cardiac sympathetic remodeling in chronic renal failure rats

BACKGROUND Chronic renal failure(CRF) patients are predisposed to arrhythmias, while the detailed mechanisms are unclear. We hypothesized the chronic inflammatory state of CRF patients may lead to cardiac sympathetic remodeling, increasing the incidence of ventricular arrhythmia(VA) and sudden cardiac death. And explored the role of atorvastatin and etanercept in this *** A total of 48 rats were randomly divided into sham operation group(Sham group), CRF group, CRF + atorvastatin group(CRF + statin group), and CRF + etanercept group(CRF + rhTNFR-Fcgroup). Sympathetic nerve remodeling was assessed by immunofluorescence of growth-associated protein 43(GAP-43) and tyrosine hydroxylase positive area fraction. Electrophysiological testing was performed to assess the incidence of VA by assessing the ventricular effective refractory period and ventricular fibrillation threshold. The levels of tumor necrosis factor-alpha(TNF-α) and interleukin-1beta were determined by Western blotting and enzyme-linked immunosorbent *** Echocardiogram showed that compared with the Sham group, left ventricular end-systolic diameter and ventricular weight/body weight ratio were significantly higher in the CRF group. Hematoxylin-eosin and Masson staining indicated that myocardial fibers were broken, disordered, and fibrotic in the CRF group. Given the extremely high inter-patient heterogeneity of acute myeloid leukemia(AML),the identification of biomarkers for prognostic assessment and therapeutic guidance is *** surface markers(CSMs)have been shown to play an important role in AML leukemogenesis and *** the current study,we evaluated the prognostic potential of all human CSMs in 130 AML patients from The Cancer Genome Atlas(TCGA)based on differential gene expression analysis and univariable Cox proportional hazards regression *** using multi-model analysis,including Adaptive LASSO regression,LASSO regression,and Elastic Net,we constructed a 9-CSMs prognostic model for risk stratification of the AML *** predictive value of the 9-CSMs risk score was further validated at the transcriptome and proteome *** Cox regression analysis showed that the risk score was an independent prognostic factor for the AML *** AML patients with high 9-CSMs risk scores had a shorter overall and event-free survival time than those with low ***,single-cell RNA-sequencing analysis indicated that patients with high 9-CSMs risk scores exhibited chemotherapy ***,PI3K inhibitors were identified as potential treatments for these high-risk *** conclusion,we constructed a 9-CSMs prognostic model that served as an independent prognostic factor for the survival of AML patients and held the potential for guiding drug therapy.

Aim:To investigate the effects of adenoviral gene transfer of IκBα mutant (IκBαM),a novel inhibitor of nuclear factor κB(NF-κB),on apoptosis,phenotype and func-tion of human monocyte-derived dendritic cells (DC).Methods:Monocytes,cocultured with granulocyte/macrophage colony-stimulating factor (GM-CSF;900ng/mL) and interleukin (IL)-4 (300ng/mL) for 5d,followed by stimulation withlipopolysaccharide (LPS;100ng/mL) for 2d differentiated into mature ***-cytes were either left untransfected or were transfected with AdIκBαM or *** transcription and expression of the IκBαM gene,and the inhibitory effect ofIκBαM on tumor necrosis factor (TNF)-α-induced NF-κB activation in mature DCwere detected by polymerase chain reaction (PCR),reverse transcription-poly-merase chain reaction (RT-PCR),Western blot analysis,and electrophoretic mobil-ity shift assays,*** phenotype,apoptosis,IL-12 secretion level ofDC,and ability to stimulate the proliferation of T cells were determined by flowcytometry,enzyme-linked immunosorbent assay and mixed leukocyte ***:PCR and RT-PCR were used to detect a unique 801 bp band in AdIκBαM-transfected mature DC,and also a dose-and time-dependent expression of theIκBαM gene,which peaked at a multiplicity of infection of 100pfu/cell and at ***,AdIκBαM significantly suppressed the TNF-α-induced NF-κBactivation,augmented apoptosis,downregulated CD80,CD83,and CD86 surfacemolecules,IL-12 secretion levels and the ability to stimulate the proliferation of Tcells in mature ***:AdIκBαM effectively transfected and potentlyinhibited NF-κB activation in monocyte-derived mature *** ofthe IκBαM gene in mature DC may contribute to T-cell immunosuppression throughinduction of DC apoptosis and downregulation of B7 molecules,providing a po-tential strategy for future DC-based immunotherapy of asthma.

Tumor-resident microbiota in breast cancer promotes cancer initiation and malignant ***,targeting microbiota to improve the effects of breast cancer therapy has not been investigated in ***,we evaluated the microbiota composition of breast tumors and found that enterotoxigenic Bacteroides fragilis(ETBF)was highly enriched in the tumors of patients who did not respond to taxane-based neoadjuvant ***,albeit at low biomass,secreted the toxic protein BFT-1 to promote breast cancer cell stemness and *** studies showed that BFT-1 directly bound to NOD1 and stabilized NOD1 ***1 was highly expressed on ALDH+breast cancer stem cells(BCSCs)and cooperated with GAK to phosphorylate NUMB and promote its lysosomal degradation,thereby activating the NOTCH1-HEY1 signaling pathway to increase ***1 inhibition and ETBF clearance increase the chemosensitivity of breast cancer by impairing BCSCs.
