Application of single-cell omics in inflammatory bowel disease

Over the past decade,the advent of single cell RNA-sequencing has revolutionized the approach in cellular transcriptomics *** current technology offers an unbiased platform to understand how genotype correlates to ***-cell omics applications in gastrointestinal(GI)research namely inflammatory bowel disease(IBD)has become popular in the last few years with multiple publications as single-cell omics techniques can be applied directly to the target organ,the GI tract at the tissue *** examination of mucosal tissue and peripheral blood in IBD,the recent boom in single cell research has identified a myriad of key immune players from enterocytes to tissue resident memory T cells,and explored functional heterogeneity within cellular subsets previously *** we begin to unravel the complex mucosal immune system in states of health and disease like IBD,the power of exploration through single-cell omics can change our approach to translational *** novel techniques evolve through multiplexing single-cell omics and spatial transcriptomics come to the forefront,we can begin to fully comprehend the disease IBD and better design targets of *** addition,hopefully these techniques can ultimately begin to identify biomarkers of therapeutic response and answer clinically relevant questions in how to tailor individual therapy to patients through personalized medicine.

The critical role of calcium dyshomeostasis in the pathogenesis of Alzheimer’s disease(AD):AD is a progressive neurodegenerative disease characterized by cognitive decline,memory impairment,and behavioral *** an estimated 50 million people being affected worldwide,the incidence of AD is constantly increasing *** hallmark of AD is the accumulation of amyloid-beta protein(Aβ)in the form of amyloid plaques and hyperphosphorylated tau protein in the form of neurofibrillary ***,increasing evidence suggests that calcium ion(Ca2+)dysregulation also plays a crucial role in the pathogenesis of AD(Calvo-Rodriguez and Bacskai,2021).As a key second messenger,Ca2+regulates a wide range of cellular processes,including the release of neurotransmitters,gene expression,and cell ***^(2+)also regulates the activity of Calcium/calmodulin-dependent protein kinase II,which is critical for synaptic plasticity,learning,and memory(Kaushik et al.,2022).Alternation in the Ca^(2+)signal is an early event in the pathogenesis of AD,which can lead to synaptic dysfunction,neuronal loss,and cognitive impairment.

Transformations of the Pleiomorphic Plant MTOC during Sporogenesis in the Hepatic Marchantia polymorpha

Sporogenesis in the hepatic Marchantia polymorpha L. provides an outstanding example of the pleiomorphic nature of the plant microtubule organizing center （MTOC）. Microtubules are nucleated from γ-tubuUn in MTOCs that change form during mitosis and meiosis. Following entry of cells into the reproductive pathway of sporogenesis, successive rounds of mitosis give rise to packets of 4-16 sporocytes. Mitotic spindles are organized at discrete polar organizers （POs）, a type of MTOC that is unique to this group of early divergent land plants. An abrupt and radical transformation in microtubule organization occurs when sporocytes enter meiosis： POs are lost and γ-tubulin is closely associated with surfaces of two large elongated plastids that subsequently divide into four. Migration of the four plastid MTOCs into a tetrahedral arrangement establishes the future spore domains and the division polarity of meiosis. As is typical of many bryophytes, cones of microtubules from the four plastid MTOCs initiate a quadripolar microtubule system （QMS） in meiotic prophase. At this point a transformation in the organization of the MTOCs occurs. The γ-tubulin detaches from plastids and forms a diffuse spheroidal pole in each of the spore domains. The plastids, which are no longer MTOCs, continue to divide. The diffuse MTOCs continue to nucleate cones of microtubules during transformation of the QMS to a bipolar spindle. Following meiosis I, γ-tubulin is associated with nuclear envelopes, and the spindles of meiosis II are organized from diffuse MTOCs at the tetrad poles. At simultaneous cytokinesis, radial microtubule systems are organized at nuclear envelope MTOCs in each of the tetrad members.

New discovery rarely runs smooth： an update on progranulin/TNFR interactions

Progranulin （PGRN） is a growth factor implicated in various pathophysiological processes, including wound healing, inflammation, tumorigenesis, and neurodegeneration. It was previously reported that PGRN binds to tumor necrosis factor receptors （TNFR） and has thera- peutic effects in inflammatory arthritis （Tang et. al, in Science 332：478-484, 2011）; however, Chen et al. reported their inability to demonstrate the PGRN-TNFR interactions under their own conditions （Chert et. al, in J Neurosci 33：9202-9213, 2013）. A letter-to-editor was then published by the original group in response to the Chen et al. paper that discussed the reasons for the latter＇s inability to recapitulate the interactions. In addition, the group published follow-up studies that further reinforced and dissected the interactions of PGRN- TNFR. Recently, the dispute about the legitimacy of PGRN-TNFR interactions appears to be finally settled with independent confirmations of these interactions in various conditions by numerous laboratories. This review presents a chronological update on the story of PGRN-TNFR interactions, highlighting the independent confirmations of these interactions in various diseases and conditions.

The immunology of human cytomegalovirus latency： could latent infection be cleared by novel immunotherapeutic strategies？

While the host immune response following primary human cytomegalovirus （HCMV） infection is generally effective at stopping virus replication and dissemination, virus is never cleared by the host and like all herpesviruses, persists for life. At least in part, this persistence is known to be facilitated by the ability of HCMV to establish latency in myeloid cells in which infection is essentially silent with, importantly, a total lack of new virus production. However, although the viral transcription programme during latency is much suppressed, a number of viral genes are expressed during latent infection at the protein level and many of these have been shown to have profound effects on the latent cell and its environment. Intriguingly, many of these latency-associated genes are also expressed during lyric infection. Therefore, why the same potent host immune responses generated during lytic infection to these viral gene products are not recognized during latency, thereby allowing clearance of latently infected cells, is far from clear. Reactivation from latency is also a major cause of HCMV-mediated disease, particularly in the immune compromised and immune naive, and is also likely to be a major source of virus in chronic subclinical HCMV infection which has been suggested to be associated with long-term diseases such as atherosclerosis and some neoplasias. Consequently, understanding latency and why latenUy infected cells appear to be immunoprivileged is crucial for an understanding of the pathogenesis of HCMV and may help to design strategies to eliminate latent virus reservoirs, at least in certain clinical settings.

Effects of seed type and bruchid genotype on the performance and oviposition behavior of Zabrotes subfasciatus （Coleoptera： Bruchidae）

The effect of different bean varieties on the performance of the bruchid beetle Zabrotes subfasciatus Boheman （Coleoptera： Bruchidae）, was determined by using wild and cultivated seeds of the genus Phaseolus. Results showed that the quality of the host plant affected the performance and the oviposition behavior of female beetles. Overall, bruchid performance was higher on cultivated seeds than on wild seeds. It was also found that the oviposition behavior and the performance of their offspring differed between females that originated from wild versus cultivated seeds. We also demonstrated the importance of a genetic component in bruchid performance： longevity, fecundity, larval development, adult size, and the sex ratio of the progeny varied according to their matemal lineage （‘family＇）. For example, on the same host type, some females laid twice as many eggs as females from other‘families＇, and the sex ratio among the offspring correlated with that of their mother. Thus, the performance and behavior of Z. subfasciatus are not only affected by environmental factors such as the quality of the seeds on which they develop, but also have a genetic basis which can counterbalance a less suitable quality of the host plant. For a crop pest such as Z. subfasciatus, its ability to survive and adapt on host plants of differing quality may be an important attribute to consider for pest management.

Forecasting tropical cyclone track and intensity is a great challenge for the meteorological community,and safeguarding the life and property of people living near the coast is an important *** major reason for challenging forecasts is the lack of observations over the vast *** tropical cyclone Mulan between 8 and 10 August 2022 over the northern part of the South China Sea,the meteorological authority and research institutes of Chinese mainland collaborated with the meteorological service in Hong Kong on conducting the first-ever ground–space–sky observing system experiment on tropical cyclone *** enhanced targeted observations collected during the experiment include Geostationary Interferometric Infrared Sounder,round-trip radiosondes,and aircraft-launched *** paper describes the campaign,technical details of the meteorological models used,and impact of the additional targeted observation data on the tropical cyclone ***,similar enhanced observation campaigns could be conducted in the future,not only in the northern part of the South China Sea,but also in other ocean basins.
