Leafhopper salivary carboxylesterase suppresses jA-Ile synthesis to facilitate initial arbovirus transmission in rice phloem

Plant jasmonoyl-L-isoleucine(jA-Ile)is a major defense signal against insect feeding,but whether or how insect salivary effectors suppress jA-Ile synthesis and thus facilitate viral transmission in the plant phloem remains *** carboxylesterases(CarEs)are the third major family of detoxification ***,we identify a new leafhopper CarE,CarE10,that is specifically expressed in salivary glands and is secreted into the rice phloem as a saliva *** CarE10 directly binds to rice jasmonate resistant 1(jAR1)and promotes its degradation by the proteasome ***,the direct association of CarE10 with jAR1 clearly impairs jAR1 enzyme activity for conversion of jA to jA-Ile in an in vitro jAIle synthesis system.A devastating rice reovirus activates and promotes the co-secretion of virions and CarE10 via virus-induced vesicles into the saliva-storing salivary cavities of the leafhopper vector and ultimately into the rice phloem to establish initial ***,a virus-mediated increase in CarE10 secretion or overexpression of CarE10 in transgenic rice plants causes reduced levels of jAR1 and thus suppresses jA-Ile synthesis,promoting host attractiveness to insect vectors and facilitating initial viral *** findings provide insight into how the insect salivary protein CarE10 suppresses host jA-Ile synthesis to promote initial virus transmission in the rice phloem.

when benefiting other beneficiaries,cushion plants may reciprocally receive feedback *** feedback effects on different sex morphs,however,remains *** this study,taking the gynodioecious Arenaria polytrichiodes as a model species,we aimed to assess the sex-specific facilitation intensity of cushion plant by measuring the beneficiary cover ratio,and to assess the potential costs in cushion reproductive functions by measuring the flower and fruit cover *** total beneficiary cover ratio was similar between females and *** produced much less flowers but more fruits than *** results suggested that females and hermaphrodites possess similar facilitation intensity,and female cushion *** may allocate more resources saved from pollen production to seed production,while hermaphrodites possibly allocate more resources to pollen production hence reducing seed *** surface areas covered by beneficiaries produced less flowers and fruits than areas without *** addition,strong negative correlations between beneficiary cover and flower cover were detected for both females and hermaphrodites,but the correlation strength were similar for these two sex ***,the correlation between beneficiary cover and fruit cover was only significantly negative for females,suggesting that beneficiary plants negatively affect fruit reproduction of females while have neutral effects on *** the results suggest that to facilitate other beneficiaries can induce reproductive costs on cushion ***,with females possibly suffering greater cost than *** differentiation in reproductive costs between sex morphs,in long-term perspective,may imply sex imbalance in population dynamics.

Platelets are small, versatile blood cells that are critical for hemostasis/thrombosis. Local platelet accumulation is a known contributor to proinflammation in various disease states. However, the anti-inflammatory/immunosuppressive potential of platelets has been poorly explored. Here, we uncovered, unexpectedly, desialylated platelets (dPLTs) down-regulated immune responses against both platelet-associated and -independent antigen challenges. Utilizing multispectral photoacoustic tomography, we tracked dPLT trafficking to gut vasculature and an exclusive Kupffer cell-mediated dPLT clearance in the liver, a process that we identified to be synergistically dependent on platelet glycoprotein Ibα and hepatic Ashwell–Morell receptor. Mechanistically, Kupffer cell clearance of dPLT potentiated a systemic immunosuppressive state with increased anti-inflammatory cytokines and circulating CD4+ regulatory T cells, abolishable by Kupffer cell depletion. Last, in a clinically relevant model of hemophilia A, presensitization with dPLT attenuated anti-factor VIII antibody production after factor VIII ( infusion. As platelet desialylation commonly occurs in daily-aged and activated platelets, these findings open new avenues toward understanding immune homeostasis and potentiate the therapeutic potential of dPLT and engineered dPLT transfusions in controlling autoimmune and alloimmune diseases.

Electrocatalytic water splitting coupled with sustainable energies is identified as an environmentally friendly and renewable strategy to generate high-quality hydrogen for the fuel ***,the main challenge is to develop high performance,low cost and chemically stable electrocatalysts to decline the energy barriers and enhance the sluggish kinetics of hydrogen evolution reaction(HER).Herein,a three-dimensional hierarchically ordered macroporous Ru-CoP@NC electrocatalyst(3DOM Ru-CoP@NC)derived from ordered macro-microporous metal-organic frameworks has been prepared using the precursor@template and double-solvent *** prepared 3DOM Ru-CoP@NC catalyst exhibits an overpotential of 15 mV(j=10 mA·cm^(-2))and a reaction Tafel slope of 38 mV·dec^(-1)in alkaline electrolyte,which are superior to commercial Pt@C ***,the overpotential and reaction Tafel slope of this catalyst in acidic media are 45 mV and 50 mV·dec^(-1),*** outstanding HER activities of 3DOM Ru-CoP@NC catalysts are ascribed to the 3D highly interconnectedreticular nanospaces that can increase effective reaction active *** N dope d carbon framework improves the electronic properties and ***,the strong interaction of Ru and CoP nanoparticles also boosts the HER *** results indicate that 3DOM Ru-CoP@NC catalysts with high catalytic activities have a broad application prospect in the future.

Dynamic fluorescent materials play a crucial role in secure inks for data encryption;however,they are still plagued by issues such as photodegradation,poor latency,and susceptibility to unauthorized ***,we propose a photochemically modulated dynamic fluorescent encryption system based on^(1)O_(2)sensitization of fluorescent composites,comprising a^(1)O_(2)-sensitive fluorophore(F2)and non-emissive *** UV irradiation,in-situ generated^(1)O_(2)from the polymer effectively binds with F2 to form endoperoxides(F2EPO),resulting in a significant redshift in emission,up to 150 ***^(1)O_(2)concentration is closely related to the irradiation time,rendering different fluorescent colors in a time-gated ***,the emission of F2EPO can be regulated by polymer chemical structure,molecular weight,and crosslinking *** on these merits,we develop a dynamic data encryption method with various non-emissive polymers as the data storage media,UV light irradiation as the data encoder,and F2 as the data *** light irradiation of diverse polymer solutions generates^(1)O_(2)at different concentrations,effectively encoding the data,which remains invisible under both UV and natural *** addition of F2 to these irradiated polymer solutions produces different redshifted fluorescence,enabling secure data *** to the non-emissive nature of the polymers,time-gated readout fashion,excellent latency of^(1)O_(2),and subtle interactions between^(1)O_(2)and F2,this data encryption is nearly *** work offers an advantage data encryption approach beyond the reach of conventional fluorophores.

Variable projection algorithms with sparse constraint for separable nonlinear models

Separable nonlinear models are widely used in various fields such as time series analysis, system modeling, and machine learning, due to their flexible structures and ability to capture nonlinear behavior of data. However, identifying the parameters of these models is challenging, especially when sparse models with better interpretability are desired by practitioners. Previous theoretical and practical studies have shown that variable projection (VP) is an efficient method for identifying separable nonlinear models, but these are based on \(L_2\) penalty of model parameters, which cannot be directly extended to deal with sparse constraint. Based on the exploration of the structural characteristics of separable models, this paper proposes gradient-based and trust-region-based variable projection algorithms, which mainly solve two key problems: how to eliminate linear parameters under sparse constraint;and how to deal with the coupling relationship between linear and nonlinear parameters in the model. Finally, numerical experiments on synthetic data and real time series data are conducted to verify the effectiveness of the proposed algorithms.
