Compost Input Effect on Dryland Wheat and Forage Yields and Soil Quality

Organic agricultural systems rely on organic amendments to achieve crop fertility requirements, and weed control must be achieved without synthetic herbicides. Our obj.ctive was to determine the crop yield and soil quality as affected by a transition from grass to dryland organic agriculture in the Central Great Plains of North America. This study evaluated three beef feedlot compost(BFC)treatments in 2010–2015 following biennial application rates: 0(control), 22.9, and 108.7 t ha^(-1) on two dryland organic cropping systems: a wheat(Triticum aestivum)-fallow(WF) rotation harvested for grain and a triticale(Triticosecale)/pea(Pisum sativum)-fallow(T/P-F) rotation harvested for forage. The triticale + pea biomass responded positively to the 108.7-t ha^(-1) BFC treatment,but not the 22.9-t ha^(-1) BFC treatment. The wheat biomass was not affected by BFC addition, but biomass N content *** feedlot compost input did not increase wheat grain yields, but had a positive effect on wheat grain Zn content. Soil total C and N contents increased with the rate of 108.7 t ha^(-1) BFC after three applications, but not with 22.9 t ha^(-1) BFC. Soil enzyme activities associated with N and C cycling responded positively to the 108.7-t ha^(-1) BFC treatment. Saturated salts were high in the soil receiving 108.7 t ha^(-1) of BFC, but did not affect crop yields. These results showed that BFC was effective in enhancing forage yields, wheat grain quality, and soil C and N, as well as specific microbial enzymes important for nutrient cycling. However, the large rates of BFC necessary to elicit these positive responses did not increase grain yields, and resulted in an excessive buildup of soil P.

YIKI is an antimalarial phytomedicine used by a traditional healer to treat malaria in Bobo-Dioulasso. However, there is no scientific evidence to support its use by local populations. The aim of this study was to identify the medicinal practices of the healer holder of YIKI and assess the clinical evidence of its phytomedicine in the uncomplicated malaria treatment. Ethnomedical survey based on a semi-structured and open questionnaire was conducted from October to December 2019 with the healer. Malaria knowledge and diagnosis methods, patient treatment and monitoring, and recipe formulation steps were surveyed. Moreover, thick and thin blood smears were taken, haemoglobin levels and temperature of consenting patients were measured before treatment, mid-treatment and at the end of treatment. The survey revealed that the healer has a good knowledge of malaria symptoms and his diagnosis is based on observation and physical examination of patients. The healer’s malaria diagnosis was rudimentary and had accuracy problems, with only 62.79% of malaria cases confirmed by microscopy. The formulation of YIKI and its use to treat malaria follow a standard process for plant harvesting, powder quantities and posology, but do not use any reproducible parameters for dose adj.stment. Forty-three patients diagnosed and treated by the healer participated in the study. Laboratory results revealed 27 Plasmodium falciparum infection cases, including 2 with parasitaemia ≥ 200,000 p/µl blood. 25 patients were selected for therapeutic evidence assessment. There was a 48% elimination of parasites, a 28% parasitaemia decrease without complete cure, with gametocytogenesis in some patients, and a 24% parasitaemia increase. Haemoglobin and temperature results suggested that YIKI was not cytotoxic and reduced fever. Encouraging preliminary results have been obtained, but in view of the low number of patients, further YIKI efficacy and toxicity studies will be necessary for patient safety.

We present detailed characterization of laser-driven fusion and neutron production(-10^(5)/second) using 8 mj. 40 fs laser pulses on a thin(j.309 organic scintillator with pulse-shape discrimination, a ~3He proportional counter and a set of 36 bubble detectors. Time-of-flight analysis of the scintillator data shows the energy of the produced neutrons to be consistent with 2.45 MeV. Particle-in-cell simulations using the WarpX code support significant neutron production from D-D fusion events in the laser±target interaction region. This high-repetition-rate laser-driven neutron source could provide a low-cost, on-demand test bed for radiation hardening and imaging applications.

Periprosthetic j.int infection(Pj.)is a rare but terrible complication in hip and knee arthroplasty,and the use of topical vancomycin powder(VP)has been investigated as a tool to potentially reduce its ***,there remains no consensus on its ***,the aim of this review is to provide an overview on the application of topical vancomycin in orthopaedic surgery focusing on the recent evidence and results in total j.int *** systematic reviews and meta-analyses on topical VP in hip and knee arthroplasty have been recently published reporting sometimes conflicting *** from all being limited by the quality of the included studies(mostly level III and IV),confounding variables are often included potentially leading to biased *** taken into consideration the exclusive use of VP in isolation,the available data,although very limited,suggest that it does not reduce the infection rate in routine primary hip and knee ***,we still cannot advise for a routinary application.A properly powered randomized-controlled trial would be necessary to clarify the role of VP in hip and knee *** on the analysis of the current evidence,the use of topical VP appears to be safe when used locally in terms of systemic adverse reactions,hence,if proven to be effective,it could bring great benefits due to its low cost and accessibility.

Ectomycorrhizal and Saprotrophic Fungal Communities Vary Across mm-Scale Soil Microsites Differing in Phosphatase Activity

To understand nutrient cycling in soils, soil processes and microorganisms need be better characterized. To determine whether specific trophic groups of fungi are associated with soil enzyme activity, we used soil imprinting to guide mm-scale sampling from microsites with high and low phosphatase activities in birch/Douglas-fir stands. Study 1 involved sampling one root window per site at 12 sites of different ages(stands);study 2 was conducted at one of the stem-exclusion stands, at which 5 root windows had been installed. Total fungal and ectomycorrhizal(EM) fungal terminal-restriction fragment length polymorphism(TRFLP)fingerprints differed between high-and low-phosphatase activity microsites at 8 of 12 root windows across 12 sites. Where differences were detected, fewer EM fungi were detected in high-than low-phosphatase activity microsites. Using 5 root windows at one site,next-generation sequencing detected similar fungal communities across microsites, but the ratio of saprotrophic to EM fungal reads was higher in high-phosphatase activity microsites in the two windows that had low EM fungal richness. In windows with differences in fungal communities, both studies indicated that EM fungi were less successful than saprotrophic fungi in colonizing fine-scale,organic matter-rich microsites. Fine-scale sampling linked with in situ detection of enzyme activity revealed relationships between soil fungal communities and phosphatase activity that could not be observed at the scales employed by conventional approaches, thereby contributing to the understanding of fine-scale phosphorus cycling in forest soils.

Correlation between plasmid DNA damage induced by PM_(10) and trace metals in inhalable particulate matters in Beij.ng air

Based on the study of Beij.ng PM10 bioreactivity with the newly developed plasmid DNA assay method, and analysis for trace elements of PM10, the cause of plasmid DNA damage by PM10 was investigated. The study showed that plasmid DNA oxidative damages by PM10 are of difference in different seasons at various areas. The concentrations of TM50 of PM10 in whole samples respectively collected at urban and comparison sites during winter were 900 μg mL?1 and 74 μg mL?1, while those in their corresponding soluble fractions were 540 μg mL?1 and 86 μg mL?1. In contrast, TM50 contents of PM10 from summer whole samples at urban areas and comparison sites were 116 μg mL?1 and 210 μg mL?1, whereas those in their soluble fractions were 180 μg mL?1 and 306 μg mL?1. The difference of bioreactivity of Beij.ng PM10 resulted from the variation of trace elements. The oxidative damage of plasmid DNA caused by Pb, Zn, As in PM10 (whole sample) was relatively strong. TM50 and Mn, V, Zn display stronger correlation in the soluble fraction. It implies that Zn could be the maj.r trace element in Beij.ng PM10 which contributes to oxidative damage to plasmid DNA.

Origin of high particle number concentrations reaching the St. Louis, Midwest Supersite

Ultrafine particles are associated with adverse health effects. Total Particle Number Concentration（TNC） of fine particles were measured during 2002 at the St. Louis — Midwest supersite. The time series showed overall low level with frequent large peaks. The time series was analyzed alongside criteria pollutant measurements and meteorological observations. Multiple regression analysis was used to identify further contributing factors and to determine the association of different pollutants with TNC levels. This showed the strong contribution of sulfur dioxide（SO2） and nitrogen oxides（NO x） to high TNC levels. The analysis also suggested that increased dispersion resulting from faster winds and higher mixing heights led to higher TNC levels. Overall, the results show that there were intense particle nucleation events in a SO2 rich plume reaching the site which contributed around 29% of TNC. A further 40% was associated with primary emissions from mobile sources. By separating the remaining TNC by time of day and clear sky conditions,we suggest that most likely 8% of TNC are due to regional nucleation events and 23% are associated with the general urban background.

Experimental investigation of flow pattern and sediment deposition in rectangular shallow reservoirs

This paper reports the experimental investigation of flow pattern, preferential regions of deposition and trap efficiency as a function of the length of rectangular shallow reservoirs. Four flow patterns were identified (from longer to shorter reservoirs): an asymmetric flow with two reattachment points, an asymmetric flow with one reattachment point, an unstable flow, and a symmetric flow without any reattachment point. Using dye visualizations, the median value and the temporal variability of the reattachment lengths were precisely measured for the asymmetric flows. For each stable flow, sediment tests with plastic particles were carried out. The regions of deposition on the bed of the reservoir were clearly a function of the flow pattern. The transition from an asymmetric flow pattern to a symmetric flow pattern was responsible for an abrupt decrease of the trap efficiency; a number of regression laws were discussed to take it into account.

In Vivo Phosphorylation Site Mapping and Functional Characterization of Arabidopsis Phototropin 1

Phototropins （phot1 and phot2） are blue-light receptor kinases controlling a range of responses that optimize the photosynthetic efficiency of plants. Light sensing is mediated by two flavin-binding motifs, known as LOVl and LOV2, located within the N-terminal region of the protein. Photoexcitation via LOV2 leads to activation of the C-terminal kinase domain and consequently receptor autophosphorylation. However, knowledge of the in-vivo phosphorylation sites for Arabidopsis phototropins is lacking and has impeded progress in elucidating the functional significance of receptor phos- phorylation. We have purified photl from Arabidopsis and identified the in-vivo sites of receptor phosphorylation by liquid chromatography tandem mass spectrometry. Arabidopsis-derived photl binds flavin mononucleotide as chromophore and is phosphorylated at four maj.r sites located upstream of LOV2 （Ser^58, Ser^85, Ser^350, and Ser^410）, three of which are induced by blue light. Nevertheless, structure-function analysis indicates that the biological activity of photl can be attributed to a modular unit comprising the LOV2-kinase region of the protein. Thus, peptide regions upstream of LOV2, including the sites of receptor phosphorylation identified here, do not appear to be important for receptor signaling. By contrast, these regions may be necessary for maximizing stomatal performance and possibly light-induced relocalization of photl.
