Adhesive small bowel obstruction(ASBO)is the most frequently encountered surgical disorder of the small *** to 80%of ASBO cases resolve spontaneously and do not require invasive *** is important to identify such patients that will benefit from conservative treatment in order to prevent unnecessarily exposing them to the risks associated with surgical intervention,such as morbidity and further adhesion *** the remaining ASBO patients,timely surgical intervention is necessary to prevent small bowel strangulation,which may cause intestinal ischemia and bowel *** early identification of these patients is key to decreasing ASBO-related morbidity and mortality,the non-specific signs and laboratory findings upon clinic presentation limit timely diagnosis and implementation of appropriate clinical *** the clinical presentation findings with those from other diagnostic imaging modalities,such as abdominal X-ray,computed tomography-scan and water-soluble contrast studies,will improve diagnosis of ASBO and help clinicians to better evaluate the potential of conservative management as a safe strategy for a particular ***,patients who present with moderate findings by all these approaches continue to represent a challenge.A new diagnostic strategy is urgently needed to further improve our ability to identify early signs of strangulated bowel,and this diagnostic modality should be able to indicate when surgical management is required.A number of potential serum markers have been proposed for this purpose,including intestinal fatty acid binding protein andα-glutathione S ***-going research is attempting to clearly define their diagnostic utility and to optimize their potential role in determining which patients should be managed surgically.

Bone loss from Wnt inhibition mitigated by concurrent alendronate therapy

Dysregulated Wnt signaling is associated with the pathogenesis of cancers, fibrosis, and vascular diseases. Inhibition of Wnt signaling has shown efficacy in various pre-clinical models of these disorders. One of the key challenges in developing targeted anti-cancer drugs is to balance efficacy with on-target toxicity. Given the crucial role Wnts play in the differentiation of osteoblasts and osteoclasts, acute inhibition of Wnt signaling is likely to affect bone homeostasis. In this study, we evaluated the skeletal effect of small molecule inhibitor of an o-acyl transferase porcupine（PORCN） that prevents Wnt signaling by blocking the secretion of all Wnts. Micro-computed tomography and histomorphometric evaluation revealed that the bones of mice treated with two structurally distinct PORCN inhibitors LGK974 and ETC-1922159（ETC-159） had loss-of-bone volume and density within 4 weeks of exposure. This decreased bone mass was associated with a significant increase in adipocytes within the bone marrow. Notably,simultaneous administration of a clinically approved anti-resorptive, alendronate, a member of the bisphosphonate family,mitigated loss-of-bone mass seen upon ETC-159 treatment by regulating activity of osteoclasts and blocking accumulation of bone marrow adipocytes. Our results support the addition of bone protective agents when treating patients with PORCN *** of bone toxicity can extend the therapeutic utility of Wnt pathway inhibitors.

Background:LDL receptor(LDLr)and ABC-transporter A1(ABCA1)are two distinct players in cellular cholesterol *** recognizes apoprotein B100 and apoE in lipoproteins to mediate the uptake of cholesterol,while ABCA1 promotes excess cholesterol efttux to apoprotein ***:The present study aims to investigate whether LDLr deficiency influences the atheroprotective effects of leukocyte ABCA1 in *** and Results:LDLr-/-mice were transplanted with bone marrow from LDLr/ABCA1mice,LDLr/ABCA1/-mice,and their respective *** leukocyte ABCA1 deficiency reduced plasma cholesterol levels,after 8weeks of Western-type diet feeding,animals transplanted with LDLr/ABCA1and LDLr/ABCA1bone marrow developed 1.6-fold(p<0.01)and 1.4-fold(p<0.05)larger lesions,compared to their respective *** clearly indicates that the atheroprotection of leukocyte ABCA1 is not affected by ***-way ANOVA analysis also demonstrated that deletion of the leukocyte LDLr reduced lesion development(p<0.001),especially in the absence of leukocyte ***,leukocyte LDLr deficiency did not affect monocytosis,recruitment of moncocytes and macrophage foam cell ***,the deletion of leukocyte ABCA1 did reduce lymphocytosis and recruitment of T cells into the adventitia of lesions,associated with smaller lesion size in LDLr/ABCA1transplanted mice,indicating the potential contribution of lymphocytosis and recruitment ofT cells to ***:Leukocyte LDLr deficiency does not affect the atheroprotective effects of leukocyte ***,suppression of lymphocytosis and recruitment of T cells in the adventitia could contribute to the atheroprotection of Leukocyte LDLr deficiency and ABCA1.

Determination of the Toxicological Risk of Urban Waste from the City of Uvira Dumped into the North-Western Coast in Lake Tanganyika (Democratic Republic of Congo)

This study focuses on determining the toxicological risks of urban waste from the city of Uvira, discharged into Lake Tanganyika, on the surrounding population. Volatile organic compounds were measured in a variety of solid waste matrices, including inorganic micropollutants in wastewater and fish. The concentrations of Hg and Pb in the lake were found to be 1.21 and 1.42 μg/L respectively and between 0.83 to 18.36 μg/L of Hg and 8.25 to 670 μg/L of Pb, at the collector outlet. The presence of trace metallic elements, such as Cr, Co, Ni, Cu, Zn, As, Sb, Hg and Pb, were detected at high concentrations compared to the WHO standard. An ecotoxicology experiment herein on wastewater samples showed lethal pollutant concentrations of the order of 0.0055 mL/mL which killed at least 50% of fish (LC50), confirming the toxicity of the wastewater. These potentially harmful effluents also contain volatile organic compounds originating in high concentration from the pharmaceutical discharges of the general Uvira hospital, in particular: toluene, ethylbenzene, m-xylene/p-xylene, o-xylene and chloroform in higher concentrations compared to the norm. Other components such as benzene, bromodichloroethane and 1,1-dichloroethane were found to be present, but at a concentration below 0.05 ppb. A variety of trace organics can be suspected to be present as well.
