The clinical role and underlying mechanisms of valproic acid(VPA)on bone homeostasis remain ***,we confirmed that VPA treatment was associated with decreased bone mass and bone mineral density(BMD)in both patients and *** effect was attributed to VPA-induced elevation in osteoclast formation and *** RNA-sequencing,we observed a significant rise in precursor miR-6359 expression in VPA-treated osteoclast precursors in vitro,and further,a marked upregulation of mature miR-6359(miR-6359)in vivo was demonstrated using quantitative real-time PCR(qRT-PCR)and miR-6359 fluorescent in situ hybridization(miR-6359-FIsH).specifically,the miR-6359 was predominantly increased in osteoclast precursors and macrophages but not in neutrophils,T lymphocytes,monocytes and bone marrow-derived mesenchymal stem cells(BMsCs)following VPA stimulation,which influenced osteoclast differentiation and bone-resorptive ***,VPA-induced miR-6359 enrichment in osteoclast precursors enhanced reactive oxygen species(ROs)production by silencing the sIRT3 protein expression,followed by activation of the MAPK signaling pathway,which enhanced osteoclast formation and activity,thereby accelerating bone ***,there are no medications that can effectively treat VPA-induced bone ***,we constructed engineered small extracellular vesicles(E-sEVs)targeting osteoclast precursors in bone and naturally carrying anti-miR-6359 by introducing of EXOmotif(CGGGAGC)in the 3’-end of the anti-miR-6359 *** confirmed that the E-sEVs exhibited decent bone/osteoclast precursor targeting and exerted protective therapeutic effects on VPA-induced bone loss,but not on ovariectomy(OVX)and glucocorticoid-induced osteoporotic models,deepening our understanding of the underlying mechanism and treatment strategies for VPA-induced bone loss.

The purpose of this contribution is to review our current understanding of the source and biochemistry of the circadian efferent input to the eyes of the American horseshoe crab Limuluspolyphemus and the impact of this input on the structure, physiology and biochemistry of Limulus eyes. special emphasis is given to the role of the biogenic amine octopamine and bio- chemical cascades it activates in the eyes. In addition to reviewing published data, we present new data showing that octopamine elevates cAMP levels in Limulus lateral eyes, and we partially characterize the pharmacology of the receptors involved in this response. We also present new data showing that octopamine regulates gene expression in Limulus lateral eyes by activating a cAMP cascade

Perinatal hypoxic-ischemic(HI)brain injury is a leading cause of morbidity and longstanding disability in newborns(Millar et al.,2017).Improved neonatal intensive care has increased survival in infants with pregnancy and birth related ***,many surviving neonates exhibit neurological abnormalities that can persist throughout life(Millar et al.,2017).Early neuroprotective strategies have the potential to improve neurological outcomes and attenuate developmental delay in ***,hypothermia is the only currently approved intervention for HI encephalopathy in fullterm infants,which is only partially protective(Millar et al.,2017).Findings in preterm and full-term infants suggest that elevations in proinflammatory cytokines are important in the pathogenesis of HI-related brain injury(Millar et al.,2017).The high mobility group box-1(HMGB1),a representative damage associatedmolecular pattern(DAMP)protein,has been reported to be implicated in a variety of brain related inflammatory diseases including traumatic brain injury,epilepsy,and stroke(Nishibori et al.,2019).Anti-HMGB1 therapies have gained increasing interest to treat inflammation related disorders in the brain(Nishibori et al.,2019).However,there is a paucity of information regarding the pathology of HMGB1 in HI-related brain injury during the perinatal *** current perspective discusses the potential contributions of HMGB1 to HI-related brain injury during the perinatal period and also addresses the potential of HMGB1 as a therapeutic target of the brain ***,this perspective emphasizes the potential for combinational therapeutics for hypothermia with anti-HMGB1 monoclonal antibodies(mAb)in perinatal HI brain injury.

Numerical modelling of non-ideal detonation in ANFO explosives applying Wood-Kirkwood theory coupled with EXPLO5 thermochemical code

Ammonium nitrate and fuel oil(ANFO)based explosive is a classic example of non-ideal high *** detonation is characterized by a strong dependence of detonation parameters on explosive charge diameter,presence and characteristics of confinement,as well as incomplete consumption of explosive at the sonic *** this work we propose a detonation model based on the Wood-Kirkwood(WK)theory coupled with the thermochemical code EXPLO5 and supplemented with reaction rate *** objective is to analyze the validity of the model for highly non-ideal ANFO explosives,with emphasis on effect of reaction rate *** was found that both single-step and two-step pressure-based models can be calibrated to reproduce experimental detonation velocity-charge radius data of ANFO at radii significantly above the failure radius(*** D/D_(id)>~0.6).single-step pressure-based model,with the pressure exponent equal to 1.4,proved to be the most accurate,even in the vicinity of the failure *** impact of the rate models is most evident on temporal(and spatial)distribution of flow parameters in detonation driving zone,especially when it comes to the conversion and width of detonation driving zone.

An Approach to Quantify the Heat Wave strength and Price a Heat Derivative for Risk Hedging

Mitigating the heat stress via a derivative policy is a vital financial option for agricultural producers and other business sectors to strategically adapt to the climate change scenario. This study has provided an approach to identifying heat stress events and pricing the heat stress weather derivative due to persistent days of high surface air temperature （sAT）. Cooling degree days （CDD） are used as the weather index for trade. In this study, a call-option model was used as an example for calculating the price of the index. Two heat stress indices were developed to describe the severity and physical impact of heat waves. The daily Global Historical Climatology Network （GHCN-D） sAT data from 1901 to 2007 from the southern California, UsA, were used. A major California heat wave that occurred 20-25 October 1965 was studied. The derivative price was calculated based on the call-option model for both long-term station data and the interpolated grid point data at a regular 0.1~ x0.1~ latitude-longitude grid. The resulting comparison indicates that （a） the interpolated data can be used as reliable proxy to price the CDD and （b） a normal distribution model cannot always be used to reliably calculate the CDD price. In conclusion, the data, models, and procedures described in this study have potential application in hedging agricultural and other risks.

Odorants could elicit repair processes in melanized neuronal and skin cells

The expression of ectopic olfactory receptors （ORs） in melanized cells, such as the human brain nigrostri- atal dopaminergic neurons and skin melanocytes, is here pointed out. ORs are recognized to regulate skin melanogenesis, whereas OR expression in the dopaminergic neurons, characterized by accumulation of pigment neuromelanin, is downregulated in Parkinson＇s disease. Furthermore, the correlation between the pigmentation process and the dopamine pathway through ct-synuclein expression is also highlighted. Purposely, these ORs are suggested as therapeutic target for neurodegenerative diseases related to the pig- mentation disorders. Based on this evidence, a possible way of turning odorants into drugs, acting on three specific olfactory receptors, OR51E2, OR2AT4 and VN1R1, is thus introduced. Various odorous molecules are shown to interact with these ORs and their therapeutic potential against melanogenic and neurodegen- erative dysfunctions, including melanoma and Parkinson＇s disease, is suggested. Finally, a direct functional link between olfactory and endocrine systems in human brain through VNIR1 is proposed, helping to counteract female susceptibility to Parkinson＇s disease in quiescent life.

该文主要论述了实质条件句和直述条件句的区别,并试图解决在逻辑分析此类句子时所面临的问题。用分析实质条件句的逻辑方法分析自然语言的直述条件句,其存在的问题主要表现在两个方面:1)实质条件句的推理模式明显不适合直述条件句(例如逆否推理和否定前件推理),2)将条件句的否定简单等同于前件与后件否定的合并,这往往不能成立。如果考虑到直述条件句的其它方面,或许能够较成功地解决目前存在的问题。首先,将“条件性”当作 if 的规约含义;其次,if 从句是从属于后件的根修饰语,然而不像多数副词性修饰成分,它们不传达新信息。条件句的不对称性,导致逆否推理在多数情况下难以成立或不可能成立。假前件不足以让条件性含义保留下来,因此否定前件推理似乎不能成立。

Three research papers were published recently to compare the virological properties of the Omicron variant with the earlier variants of concern(VOC).1,2,3 The sARs-CoV-2 B.1.1.529 variant,Omicron,was first detected in south Africa in October 2021 and has spread rapidly;within three months,it appeared in more than 87 countries.4 During the surveillance of the epidemiological data and mutations of earlier COVID-19 variants and Omicron,significant differences were revealed mainly in the reproduction rate and hospitalization.
