Bitter melon fruit is susceptible to yellowing,softening,and rotting under room-temperature storage conditions,resulting in reduced commercial value.Nitric oxide(NO)is an important signaling molecule and plays a crucial role in regulating the fruit postharvest quality.In this study,we investigated the effects of NO treatment on changes in sensory and firmness of bitter melon fruit during postharvest storage.Moreover,transcriptomic,metabolomic,and proteomic analyses were performed to elucidate the regulatory mechanisms through which No treatment delays the ripening and senescence of bitter melon fruit.Our results show that differentially expressed genes(DEGs)were involved in fruit texture(CSLE,β-Gal,and PME),plant hormone signal transduction(ACS,JAR4,and AUX28),and fruit flavor and aroma(SUS2,LOX,and GDH2).In addition,proteins differentially abundant were associated with fruit texture(PLY,PME,and PGA)and plant hormone signal transduction(PBL15,JAR1,and PYL9).Moreover,No significantly increased the abundance of key enzymes involved in the phenylpropanoid biosynthetic pathway,thus enhancing the disease resistance and alleviating softening of bitter melon fruit.Finally,differential metabolites mainly included phenolic acids,terpenoids,and flavonoids.These results provide a theoretical basis for further studies on the physiological changes associated with postharvest ripening and senescence of bitter melon fruit.

在细胞尺度上定量分析导管特征,对于揭示植物对环境的适应策略具有重要意义。直接打磨样品并结合体视显微镜成像技术是目前研究木质部解剖结构(针叶树管胞和阔叶树导管)的主要方法之一,但仍不可避免损伤木质部细胞,限制了对木质部细胞真实解剖结构的认识。本研究选取了加拿大魁北克南部的糖枫树、北美白桦、美洲白蜡树、美洲铁木、大齿杨、苦味山核桃、北美红橡7个阔叶树种,应用计算机显微断层扫描技术(μCT)和体视显微镜法分别测量各树种的导管直径和面积,采用线性模型对两种方法的测量结果进行数据拟合,探讨采用μCT量化阔叶树导管大小的可行性。结果表明:对所测定的7个树种,应用两种方法测量导管大小的结果高度相似(R~2=0.98)。环孔材树种应用两种方法测量的导管直径结果拟合优度(苦味山核桃R~2=0.98,美洲白蜡树R~2=0.96,北美红橡R~2=0.99)高于散孔材树种(北美白桦R~2=0.88,美洲铁木R~2=0.73,糖枫树R~2=0.68,大齿杨R~2=0.88)。应用2种方法测量小导管(直径≤200μm,R~2=0.94)的结果拟合优度高于大导管(直径>200μm,R~2=0.92)。μCT技术为量化阔叶树种木质部导管提供了一种无损检测新途径。

Estimating the number of Chinese cancer patients eligible for and benefit from immune checkpoint inhibitors

The total number of cancer patients who are eligible for and will benefit from immune checkpoint inhibitors(ICIs)in China has not been quantified.This cross-sectional study was conducted to estimate the number of Chinese cancer patients with eligibility and response to ICIs based on the 2015 Chinese cancer statistics and the immune checkpoint inhibitor clinical practice guideline of the Chinese Society of Clinical Oncology.A total of 11 ICIs were recommended for 17 cancer types.The estimated number of eligible patients annually was 1290156(55.18%),which included 888738 males(60.05%)and 400468 females(46.67%).The estimated number of responders annually was 448972(19.20%),which included 309023 males(20.88%)and 139764 females(16.29%).Gastric cancer(n=291000,12.45%),non-small-cell lung cancer(n=289629,12.39%),and hepatocellular carcinoma(n=277100,11.85%)were the top three cancer types with the highest number of eligible patients.Non-small-cell lung cancer(n=180022,7.70%),hepatocellular carcinoma(n=75648,3.24%),and small-cell lung cancer(n=64362,2.75%)were the top three cancer types with the highest number of responders.In conclusion,ICIs provide considerable benefit in Chinese cancer patients under optimal estimation.

UV-C照射对鲜切豇豆的保鲜效果研究

为探究短波紫外线(UV-C)照射对鲜切豇豆的保鲜效果,在预试验结果的基础上,分别采用0、0.5 kJ/m^(2)的UV-C处理豇豆,然后将其置于4℃条件下贮藏,定期测定相关指标。结果表明:采用0.5 kJ/m^(2)UV-C照射可以维持鲜切豇豆较高的VC、可溶性固形物、可溶性蛋白和叶绿素含量,减少鲜切豇豆丙二醛(MDA)的积累,同时还能保持鲜切豇豆较高的总酚、类黄酮含量,并提高抗氧化酶的活性,包括过氧化物酶(POD)、过氧化氢酶(CAT)、抗坏血酸过氧化物酶(APX),一定程度上稳定多酚氧化酶(PPO)的活性。综合来说,0.5 kJ/m^(2)UV-C处理可用于鲜切豇豆的贮藏保鲜。

Efficacy and safety of gemcitabine plus S-1 vs. gemcitabine plus nab-paclitaxel in treatment-na?ve advanced pancreatic ductal adenocarcinoma

Objective:Gemcitabine plus nab-paclitaxel(GnP)is the standard first-line therapy for advanced pancreatic ductal adenocarcinoma(PDAC).S-1,an oral fluoropyrimidine derivative,as compared with gemcitabine,is non-inferior in terms of overall survival(OS)and is associated with lower hematologic toxicity.Accordingly,S-1 is a convenient oral alternative treatment for advanced PDAC.This study was aimed at comparing the efficacy and safety of gemcitabine plus S-1(GS)vs.GnP as first-line chemotherapy for advanced PDAC.Methods:Patients with advanced PDAC who received first-line GS or GnP at the Peking Union Medical College Hospital between March 2011 and November 2022 were evaluated.Results:A total of 300 patients were assessed,of whom 84 received GS and 216 received GnP.The chemotherapy completion rate was higher with GS than GnP(50.0%vs.30.3%,P=0.0028).The objective response rate(ORR)was slightly higher(14.3%vs.9.7%,P=0.35),and the median OS was significantly longer(17.9 months vs.13.3 months,P=0.0078),in the GS group than the GnP group.However,the median progression-free survival(PFS)did not significantly differ between groups.Leukopenia risk was significantly lower in the GS group than the GnP group(14.9%vs.28.1%,P=0.049).Conclusions:As first-line chemotherapy for advanced PDAC,the GS regimen led to a significantly longer OS than the GnP regimen.The PFS,ORR,and incidence of severe adverse events were comparable between the GS and GnP groups.

Objective:Different SSTR2 antagonists have been developed.This study aims to evaluate the impact of different peptides and chelators on the diagnostic performance of SSTR2 antagonists in well-differentiated NETs.Methods:In this prospective study,participants were equally randomized into two arms:Arm A,participants would undergo a whole-body 68Ga-NODAGA-LM3 PET/CT scan on the 1st day and 68Ga-DOTA-LM3 PET/CT scan on the 2nd day;Arm B,participants would undergo a whole-body 68Ga-NODAGA-LM3 PET/CT scan on the 1st day and 68Ga-NODAGA-JR11 PET/CT scan on the 2nd day.Biodistribution in normal organs,lesion detection ability,and tumor uptakes were compared within each Arm.Results:A total of 40 participants(age,49.5±13.4,21 men),20 in each arm,were recruited in the study.In Arm A,68Ga-DOTA-LM3 showed lower background.However,the lesion detection ability(overall lesion detected,445 versus 548,P=0.005)and the lesion uptake(overall lesions SUVmax,19.8±17.2 versus 35.3±28.8,P<0.001)was significantly lower than those of 68Ga-NODAGA-LM3.In Arm B,both 68Ga-NODAGA-LM3 and 68Ga-NODAGA-JR11 showed similar biodistribution and lesion uptake(SUVmax,28.5±23.8 versus 25.0±20.0,P<0.001)despite minor differences.The lesion detection ability was the same between these two tracers(overall lesion detected,503 versus 503).Conclusions:The diagnostic performance of SSTR2 antagonists was sensitive to chelators.Both 68Ga-NODAGA-LM3 and 68Ga-NODAGA-JR11 outperformed 68Ga-DOTA-LM3 with higher lesion uptake and detection ability,of which 68Ga-NODAGA-LM3 had marginally but significantly higher lesion uptake.

Tomato is one of the most important vegetable crops in the world and is a model plant used to study the ripening of climacteric fleshy fruit.During the ripening process of tomato fruit,flavor and aroma metabolites,color,texture and plant hormones undergo significant changes.However,low temperatures delayed the ripening process of tomato fruit,inhibiting flavor compounds and ethylene production.Metabolomics and transcriptomics analyses of tomato fruit stored under low temperature(LT,5°C)and room temperature(RT,25°C)were carried out to investigate the effects of storage temperature on the physiological changes in tomato fruit after harvest.The results of transcriptomics changes revealed that the differentially expressed genes(DEGs)involved in tomato fruit ripening,including several kinds of transcription factors(TFs)(TCP,WRKY,MYB and bZIP),enzymes involved in cell wall metabolism[beta-galactosidase(β-GAL),pectinesterase(PE)and pectate lyase(PL),cellulose and cellulose synthase(CESA)],enzymes associated with fruit flavor and aroma[acetyltransferase(AT),malic enzyme(ME),lipoxygenase(LOX),aldehyde dehydrogenase(ALDH),alcohol dehydrogenase(ADH)and hexokinase(HK)],genes associated with heat stress protein 70 and genes involved in the production of plant hormones such as Ethylene responsive factor 1(ERF1),Auxin/indoleacetic acids protein(AUX/IAA),gibberellin regulated protein.Based on the above results,we constructed a regulatory network model of the effects of different temperatures during the fruit ripening process.According to the analysis of the metabolomics results,it was found that the contents of many metabolites in tomato fruit were greatly affected by storage temperature,including,organic acids(L-tartaric acid,a-hydroxyisobutyric acid and 4-acetamidobutyric acid),sugars(melezitose,beta-Dlactose,D-sedoheptulose 7-phosphate,2-deoxyribose 1-phosphate and raffinose)and phenols(coniferin,curcumin and feruloylputrescine).This study revealed the effects of storage temperature on postharvest tomato fruit and provided a
