Intracellular organelle networks： Understanding their organization and communication through systems-level modeling and analysis

baCKGROUND： Membrane-bound intracellular organelles are biochemically distinct compartments used by eukaryotic cells for serving specialized physiological functions and organizing their internal environment. Recent studies revealed surprisingly extensive communication between these organelles and highlighted the network nature of their organization and communication. Since organization and communication of the organelles are carried out at the systems level through their networks, systems-level studies are essential for understanding the underlying mechanisms. METHODS： We reviewed recent studies that used systems-level quantitative modeling and analysis to understand organization and communication of intracellular organelle networks. RESULTS： We first review modeling and analysis studies on how fusion/fission and degradation/biogenesis, two essential and closely related classes of activities of individual organelles, collectively mediate the dynamic organization of their networks. We then turn to another important aspect of the dynamic organization of the organelle networks, namely how organelles are physically connected within their networks, a property referred to as the topology of the networks in mathematics, and summarize some of their distinct properties. Lastly, we briefly review modeling and analysis studies that aim to understand communication between different organelle networks, focusing on cellular calcium homeostasis as an example. We conclude with a brief discussion of future directions for research in this area. CONCLUSIONS： Together, the reviewed studies provide critical insights into how diverse activities of individual organelles collectively mediate the organization and communication of their networks. They demonstrate the essential role of systems- level modeling and analysis in understanding complex behavior of such networks.

Electrocoagulation is progressively becoming an ecologically friendly water treatment method owing to its lack of secondary pollution,high active ingredient concentration,high treatment effectiveness,simple equipment,and simplicity of automated control ***,electrocoagulation is offered as a method for treating wastewater containing azo dyes using a revolutionary flexible electronic fabric that can be mass-producible at a reasonable price.A computer-controlled machine embroiders 316L stainless steel fiber(316L SSF)onto an insulating fabric to manufacture a flexible electronic device of cathode and anode with a monopolar arrangement on the fabric *** methyl orange(MO)solution to simulate azo dye wastewater,the decolorization rate of 500 ml MO reached 99.25% under the conditions of 50 mg·L^(-1)initial mass concentration,120 min electrolysis time,15 mA·g^(-1)current density,1 cm electrode spacing,0.1 mol·L^(-1)NaCl,pH 7.6,200 r·min^(-1)rotational speed of the stirrer,and 22-25℃ room *** addition,it is feasible to embroider flexible electronic fabrics with varied sizes and numbers of electrodes based on the amount of treated sewage to increase the degradation rate,which has significant practical application value.

Mass balance Modeling for Electric arc Furnace and Ladle Furnace System in Steelmaking Facility in Turkey

In the electric arc furnace （EAF） steel production processes, scrap steel is principally used as a raw material instead of iron ore. In the steelmaking process with EAF, scrap is first melted in the furnace and then the desired chemical composition of the steel can be obtained in a special furnace such as ladle furnace （LF）. This kind of furnace process is used for the secondary refining of alloy steel. LF furnace offers strong heating fluxes and enables precise temperature control, thereby allowing for the addition of desired amounts of various alloying elements. It also provides outstanding desulfurization at high-temperature treatment by reducing molten steel fluxes and removing deoxidation products. Elemental analysis with mass balance modeling is important to know the precise amount of required alloys for the LF input with respect to scrap composition. In present study, chemical reactions with mass conservation law in EAF and LF were modeled altogether as a whole system and chemical compositions of the final steel alloy output can be obtained precisely according to different scrap compositions, alloying elements ratios, and other input amounts. Besides, it was found that the mass efficiency for iron element in the system is 95.93%. These efficiencies are calculated for all input elements as 8. 45% for C, 30.31% for Si, 46.36% for Mn, 30.64% for P, 41.96% for S, and 69.79% for Cr, etc. These efficiencies provide valuable ideas about the amount of the input materials that are vanished or combusted for 100 kg of each of the input materials in the EAF and LF system.

A direct position determination method with combined TDOA and FDOA based on particle filter

The localization of a stationary transmitter using moving receivers is considered. The original Direct Position Determination （DPD） methods, with combined Time Difference of arrival （TDOA） and Frequency Difference of arrival （FDOA）, do not perform well under low Signal-to-Noise Ratio （SNR）, and worse still, the computation cost is difficult to accept when the computational capabilities are limited. To get better positioning performance, we present a new DPD algorithm that proves to be more computationally efficient and more precise for weak signals than the conventional approach. The algorithm partitions the signal received with the same receiver into multiple non-overlapping short-time signal segments, and then uses the TDOA, the FDOA and the coherency among the short-time signals to locate the target. The fast maximum likelihood estimation, one iterative method based on particle filter, is designed to solve the problem of high computation load. A secondary but important result is a derivation of closed-form expressions of the Cramer-Rao Lower Bound （CRLB）. The simulation results show that the algorithm proposed in this paper outperforms the traditional DPD algorithms with more accurate results and higher computational efficiency, and especially at low SNR, it is more close to the CRLB.

Optimization of antioxidant activity by response surface methodology in hydrolysates of jellyfish(Rhopilema esculentum) umbrella collagen

To optimize the hydrolysis conditions to prepare hydrolysates of jellyfish umbrella collagen with the highest hydroxyl radical scavenging activity, collagen extracted from jellyfish umbrella was hydrolyzed with trypsin, and response surface methodology （RSM） was applied. The optimum conditions obtained from experiments were pH 7.75, temperature （T） 48.77 ℃, and enzyme-to-substrate ratio （[E]/[S]） 3.50%. The analysis of variance in RSM showed that pH and [E]/[S] were important factors that significantly affected the process （P〈0.05 and P〈0.01, respectively）. The hydrolysates of jellyfish umbrella collagen were fractionated by high performance liquid chromatography （HPLC）, and three fractions （HF-1〉3000 Da, 1000 Da〈HF-2〈3000 Da, and HF-3〈1000 Da） were collected. The HF-2 fraction had the highest hydroxyl radical scavenging activity with the highest yield compared with the other two fractions. Furthermore, HF-2 also showed the strongest Cu^2＋-chelating ability and the best tyrosinase-inhibitory activity.

Velocity field of wave-induced local fluid flow in double-porosity media

Under the excitation of elastic waves,local fluid flow in a complex porous medium is a major cause for wave dispersion and *** the local fluid flow process is simulated with wave propagation equations in the double-porosity medium,two porous skeletons are usually assumed,namely,host and *** them,the volume ratio of inclusion skeletons is *** previous studies have ignored the consideration of local fluid flow velocity field in inclusions,and therefore they can not completely describe the physical process of local flow oscillation and should not be applied to the situation where the fluid kinetic energy in inclusions cannot be *** this paper,we analyze the local fluid flow velocity fields inside and outside the inclusion,rewrite the kinetic energy function and dissipation function based on the double-porosity medium model containing spherical inclusions,and derive the reformulated Biot-Rayleigh(BR)equations of elastic wave propagation based on Hamilton’s *** present simulation examples with different rock and fluid *** between BR equations and reformulated BR equations show that there are significant differences in wave response ***,we compare the reformulated BR equations with the previous theories and experimental data,and the results show that the theoretical results of this paper are correct and effective.

Nonlinear Mathematical Modeling and Sensitivity Analysis of Hydraulic Drive Unit

The previous sensitivity analysis researches are not accurate enough and also have the limited reference value, because those mathematical models are relatively simple and the change of the load and the initial displacement changes of the piston are ignored, even experiment verification is not conducted. Therefore, in view of deficiencies above, a nonlinear mathematical model is established in this paper, including dynamic characteristics of servo valve, nonlinear characteristics of pressure-flow, initial displacement of servo cylinder piston and friction nonlinearity. The transfer function block diagram is built for the hydraulic drive unit closed loop position control, as well as the state equations. Through deriving the time-varying coefficient items matrix and time-varying free items matrix of sensitivity equations respectively, the expression of sensitivity equations based on the nonlinear mathematical model are obtained. According to structure parameters of hydraulic drive unit, working parameters, fluid transmission characteristics and measured friction-velocity curves, the simulation analysis of hydraulic drive unit is completed on the MATLAB/Simulink simulation platform with the displacement step 2 mm, 5 mm and 10 mm, respectively. The simulation results indicate that the developed nonlinear mathematical model is sufficient by comparing the characteristic curves of experimental step response and simulation step response under different constant load. Then, the sensitivity function time-history curves of seventeen parameters are obtained, basing on each state vector time-history curve of step response characteristic. The maximum value of displacement variation percentage and the sum of displacement variation absolute values in the sampling time are both taken as sensitivity indexes.
