Network pharmacology combined with molecular docking revealed the potential targets of Coridius chinensis in prostate cancer treatment

bACKGROUND Prostate cancer(PCa)has high morbidity and mortality rates in elderly *** a history of thousands of years,traditional Chinese medicine derived from insects could be an important source for developing cancer-targeted drugs to prevent tumorigenesis,enhance therapeutic effects,and reduce the risk of recurrence and *** studies have shown that Coridius chinensis(Cc)has anticancer *** To elucidate the mechanism of action of Cc against PCa via network pharma-cology and molecular *** Potential targets for Cc and PCa were predicted using ChemDraw 19.0 software,the PharmMapper database and the GeneCards ***,the STRING database was used to construct the protein–protein interaction *** Ontology(GO),Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment and molecular docking analyses were subsequently conducted to identify the key targets,active ingredients and pathways *** GO and KEGG analyses indicated that the PI3K-Akt signalling pathway was the critical pathway(P valuebeen identified in *** active compounds(asponguanosines A,asponguanine b,asponguanine C,aspong-pyrazine A,N-acetyldopamine,aspongadenine b and aspongpyrazine b)were selected for molecular docking with 9 potential targets,and the results revealed that aspongpyrazine A and asponguanosine A are the main components by which Cc affects PCa(affinitybonding),but more studies are *** We used network pharmacology to predict the bioactive components and important targets of Cc for the treatment of PCa,supporting the development of Cc as a natural anticancer agent.

Soybean is a photoperiod-sensitive short-day crop whose reproductive period and yield are markedly affected by day-length *** weight is one of the key traits determining the soybean yield;how-ever,the prominent genes that control the final seed weight of soybean and the mechanisms underlying the photoperiod's effect on this trait remain poorly *** this study,we identify SwW19 as a major locus controlling soybean seed weight by QTL mapping and determine Dt1,an orthologous gene of Arabidopsis TFL1 that is known to govern the soybean growth habit,as the causal gene of the SW19 *** showed that Dt1 is highly expressed in developing seeds and regulates photoperiod-dependent seed weight in *** analyses revealed that the Dt1 protein physically interacts with the sucrose transporter GmSWEET10a to negatively regulate the import of sucrose from seed coat to the embryo,thus modulating seed weight under long ***,Dt1 does not function in seed development under short days due to its very low ***,we discovered a novel natural allelic variant of Dt1(H4 haplotype)that decouples its pleiotropic effects on seed size and growth habit;i.e.,this variant remains functional in seed development but fails to regulate the stem growth habit of ***,our findings provide new insights into how soybean seed development responds to photoperiod at different latitudes,offering an ideal genetic component for improving soybean's yield by manipulating its seed weightandgrowth habit.

Simulation of deuterium pellet ablation and deposition in the EAST tokamak with HPI2 code

Pellet injection is a primary method for fueling the plasma in magnetic confinement *** that goal the knowledges of pellet ablation and deposition profiles are *** the present study,the pellet fueling code HPI2 was used to predict the ablation and deposition profiles of deuterium pellets injected into a typical H-mode discharge on the EAST *** ablation and deposition profiles were evaluated for various pellet injection locations,with the aim at optimizing the pellet injection to obtain a deep fueling *** this study,we investigate the effect of the injection angle on the deposition depth of the pellet at different velocities and *** ablation and deposition of the injected pellet are mainly studied at each injection position for three different injection angles:0°,45°,and 60°.The pellet injection on the high field side(HFS)can achieve a more ideal deposition depth than on the low field side(LFS).Among these angles,horizontal injection on the middle plane is relatively better on either the HFS or the *** the injection location is 0.468 m below the middle plane on the HFS or 0.40 m above the middle plane of the LFS,it can achieve a similar deposition depth to the one of its corresponding *** the pre-cooling effect is taken into account,the deposition depth is predicted to increase only slightly when the pellet is launched from the *** findings of this study will serve as a reference for the update of pellet injection systems for the EAST tokamak.

ZIF-90-loaded DNAzyme-responsive PROTAC for the self-powered degradation of NF-κb

Transcription factors(TFs) play vital roles in regulating gene expressions. Dysregulated or mutated TFs are implicated in a wide array of diseases, highlighting their potential as drug targets. However, TFs are considered undruggable by conventional inhibitor-based modalities. The recently emerged proteolysis targeting chimeras(PROTACs) have exhibited great potential in tackling TFs. In particular, DNA-ligand chimeras that arm E3 ligase-recruiting ligands to TF-binding double-strand DNA represent a promising strategy for the targeted proteolysis of TFs. Here, we report a DNAzyme-inducible PROTAC(DzTAC)that selectively degrades NF-κb in a zinc ion-dependent manner. We further applied zinc-imidazolate metal–organic framework-90(ZIF-90) to encapsulate DzTAC(ZIF-90@DzTAC), facilitating its delivery into cancer cells while self-supplying Zn2+via adenosine triphosphate(ATP)-triggered ZIF-90 dissociation. Moreover, ZIF-90@DzTAC can enhance the treatment efficacy of doxorubicin-based chemotherapy in tumor-bearing mice. The developed DzTAC provides specific modulation over TF activity and opens an avenue for more functional nucleic acid-based control over targeted protein degradation.

Coxsackievirus b3(CVb3)is the pathogen causing hand,foot and mouth disease(HFMD),which manifests across a spectrum of clinical severity from mild to ***,CVb3-infected mouse models mainly demonstrate viral myocarditis and pancreatitis,failing to replicate human HFMD *** several enteroviruses have been evaluated in Syrian hamsters and rhesus monkeys,there is no comprehensive data on *** this study,we have first tested the susceptibility of Syrian hamsters to CVb3 infection via different *** results showed that Syrian hamsters were successfully infected with CVb3 by intraperitoneal injection or nasal drip,leading to nasopharyngeal colonization,acute severe pathological injury,and typical HFMD ***,the nasal drip group exhibited a longer viral excretion cycle and more severe pathological *** the subsequent study,rhesus monkeys infected with CVb3 through nasal drips also presented signs of HFMD symptoms,viral excretion,serum antibody conversion,viral nucleic acids and antigens,and the specific organ damages,particularly in the ***,there were no significant differences in myocardial enzyme levels,and the clinical symptoms resembled those often associated with common,mild *** summary,the study successfully developed severe Syrian hamsters and mild rhesus monkey models for CVb3-induced *** models could serve as a basis for understanding the disease pathogenesis,conducting pre-trial prevention and evaluation,and implementing post-exposure intervention.
