Parameter calibration and experimental verification of discrete element simulation model for Protaetia brevitarsis larvae bioconversion mixture

To improve the survival rate of larvae during material separation after biotransformation of existing residual film mixtures of Protaetia brevitarsis larvae,this paper adopts the method of combining physical test and EDEM simulation test,and selects Hertz Mindlin with JKR contact model to calibrate the discrete element simulation contact parameters of the Protaetia brevitarsis larvae and the frass ***,the cylinder lifting method was used to determine the actual repose angle of the mixture of larvae and *** collision recovery coefficients between larvae-frass and steel,static friction coefficient,kinetic friction coefficient and the collision recovery coefficient between larvae were measured through physical tests such as the inclined plane *** Plackett-burman test was then used to screen out the factors that have a significant impact on the repose angle:Poisson’s ratio of frass,frass-frass rolling friction coefficient,frass JKR surface energy,frass-larvae JKR surface *** optimal value intervals of four significant factors were determined based on the steepest climb test,based on the box-behnken response surface analysis test,the second-order regression model between the repose angle and four significant factors was determined,and variance and interaction effects were *** with the actual repose angle as the goal,the significant factors were optimized and the optimal parameter combination of the four significant factors was *** simulation test of material repose angle and screening was carried out with the optimal parameter combination,and compared with the physical *** was found that the maximum relative errors of the two tests were 1.48%and 3.79%respectively,indicating that the calibrated parameter values are true and reliable,It can provide a reference for the discrete element simulation of the transportation and separation of the Protaetia brevitarsis larvae-frass mixture.

A novel live attenuated vaccine candidate protects chickens against subtype b avian metapneumovirus

Avian metapneumovirus(aMPV) is a highly contagious pathogen that causes acute upper respiratory tract diseases in chickens and turkeys, resulting in serious economic losses. Subtype b aMPV has recently become the dominant epidemic strain in China. We developed an attenuated aMPV subtype b strain by serial passaging in Vero cells and evaluated its safety and efficacy as a vaccine candidate. The safety test showed that after the 30th passage, the LN16-A strain was fully attenuated, as clinical signs of infection and histological lesions were absent after *** LN16-A strain did not revert to a virulent strain after five serial passages in chickens. The genomic sequence of LN16-A differed from that of the parent wild-type LN16(wtLN16) strain and had nine amino acid mutations. In chickens, a single immunization with LN16-A induced robust humoral and cellular immune responses, including the abundant production of neutralizing antibodies, CD4^(+) T lymphocytes, and the Th1(IFN-γ) and Th2(IL-4 and IL-6)cytokines. We also confirmed that LN16-A provided 100% protection against subtype b aMPV and significantly reduced viral shedding and turbinate inflammation. Our findings suggest that the LN16-A strain is a promising live attenuated vaccine candidate that can prevent infection with subtype b aMPV.

AP-1 and SP1 trans-activate the expression of hepatic CYP1A1 and CYP2A6 in the bioactivation of AFb1 in chicken

Dietary exposure to aflatoxin b1(AFb1)is harmful to the health and performance of domestic *** hepatic cytochrome P450s(CYPs),CYP1A1 and CYP2A6,are the primary enzymes responsible for the bioactivation of AFb1to the highly toxic exo-AFb1-8,9-epoxide(AFbO)in ***,the transcriptional regulation mechanism of these CYP genes in the liver of chicks in AFb1metabolism remains ***-luciferase reporter assay,bioinformatics and site-directed mutation results indicated that specificity protein 1(SP1)and activator protein-1(AP-1)motifs were located in the core region-1,063/-948,-606/-541 of the CYP1A1 promoter as well as-636/-595,-503/-462,-147/-1 of the CYP2A6 ***,overexpression and decoy oligodeoxynucleotide technologies demonstrated that SP1 and AP-1 were pivotal transcriptional activators regulating the promoter activity of CYP1A1 and ***,bioactivation of AFb1to AFbO could be increased by upregulation of CYP1A1 and CYP2A6 expression,which was trans-activated owing to the upregulation of AP-1,rather than SP1,stimulated by AFb1-induced reactive oxygen ***,nano-selenium could reduce ROS,downregulate AP-1 expression and then decrease the expression of CYP1A1 and CYP2A6,thus alleviating the toxicity of *** conclusion,AP-1 and SP1 played important roles in the transactivation of CYP1A1 and CYP2A6 expression and further bioactivated AFb1to AFbO in chicken liver,which could provide novel targets for the remediation of aflatoxicosis in chicks.

Multiple mixed state variable incremental integration for reconstructing extreme multistability in a novel memristive hyperchaotic jerk system with multiple cubic nonlinearity

Memristor-based chaotic systems with infinite equilibria are interesting because they generate extreme *** initial state-dependent dynamics can be explained in a reduced-dimension model by converting the incremental integration of the state variables into system ***,this approach cannot solve memristive systems in the presence of nonlinear terms other than the memristor *** addition,the converted state variables may suffer from a degree of *** allow simpler mechanistic analysis and physical implementation of extreme multistability phenomena,this paper uses a multiple mixed state variable incremental integration(MMSVII)method,which successfully reconstructs a four-dimensional hyperchaotic jerk system with multiple cubic nonlinearities except for the memristor term in a three-dimensional model using a clever linear state variable mapping that eliminates the divergence of the state ***,the simulation circuit of the reduced-dimension system is constructed using Multisim simulation software and the simulation results are consistent with the MATLAb numerical simulation *** results show that the method of MMSVII proposed in this paper is useful for analyzing extreme multistable systems with multiple higher-order nonlinear terms.

Association of DNA methylation/demethylation with the functional outcome of stroke in a hyperinflammatory state

Inflammation is closely related to stroke prognosis, and high inflammation status leads to poor functional outcome in stroke. DNA methylation is involved in the pathogenesis and prognosis of stroke. However, the effect of DNA methylation on stroke at high levels of inflammation is unclear. In this study, we constructed a hyperinflammatory cerebral ischemia mouse model and investigated the effect of hypomethylation and hypermethylation on the functional outcome. We constructed a mouse model of transient middle cerebral artery occlusion and treated the mice with lipopolysaccharide to induce a hyperinflammatory state. To investigate the effect of DNA methylation on stroke, we used small molecule inhibitors to restrain the function of key DNA methylation and demethylation enzymes. 2,3,5-Triphenyltetrazolium chloride staining, neurological function scores, neurobehavioral tests, enzyme-linked immunosorbent assay, quantitative reverse transcription PCR and western blot assay were used to evaluate the effects after stroke in mice. We assessed changes in the global methylation status by measuring DNA 5-mc and DNA 5-hmc levels in peripheral blood after the use of the inhibitor. In the group treated with the DNA methylation inhibitor, brain tissue 2,3,5-triphenyltetrazolium chloride staining showed an increase in infarct volume, which was accompanied by a decrease in neurological scores and worsening of neurobehavioral performance. The levels of inflammatory factors interleukin 6 and interleukin-1 beta in ischemic brain tissue and plasma were elevated, indicating increased inflammation. Related inflammatory pathway exploration showed significant overactivation of nuclear factor kappa b. These results suggested that inhibiting DNA methylation led to poor functional outcome in mice with high inflammation following stroke. Further, the effects were reversed by inhibition of DNA demethylation. Our findings suggest that DNA methylation regulates the inflammatory response in stroke

Noncollinear antiferromagnet Mn_(3)Sn has shown remarkable efficiency in charge-spin conversion,a novel magnetic spin Hall effect,and a stable topological antiferromagnetic state,which has resulted in great interest from researchers in the field of spin-orbit *** research has primarily focused on the spin-orbit torque effect of epitaxially grown noncollinear antiferromagnet Mn_(3)Sn ***,this method is not suitable for large-scale industrial *** this study,amorphous Mn_(3)Sn films and Mn_(3)Sn/Py heterostructures were prepared using magnetron sputtering on silicon *** spin-torque ferromagnetic resonance measurement demonstrated that only the conventional spin-orbit torque effect generated by in-plane polarized spin currents existed in the Mn_(3)Sn/Py heterostructure,with a spin-orbit torque efficiency of ***,we prepared the perpendicular magnetized Mn_(3)Sn/CoTb heterostructure based on amorphous Mn_(3)Sn film,where the spin-orbit torque driven perpendicular magnetization switching was achieved with a lower critical switching current density(3.9×10^(7)A/cm^(2))compared to Ta/CoTb *** research reveals the spin-orbit torque effect of amorphous Mn_(3)Sn films and establishes a foundation for further advancement in the practical application of Mn_(3)Sn materials in spintronic devices.

We design two new layered indium halide compounds LaOInF_(2)and LaOInCl_(2)by means of first-principles calculations and evolutionary crystal structure *** find both compounds crystallize in a tetragonal structure with P4/nmm space group and have indirect band gaps of 2.58 eV and 3.21 eV,*** substituting O with F,both of them become metallic and superconducting at low *** F-doping leads to strong electron-phonon coupling in the low-energy acoustic phonon modes which is mainly responsible for the induced *** total electron-phonon coupling strength are 1.86 and 1.48,while the superconducting transition temperature(T_(c))are about 7.2 K and 6.5 K with 10%and 5%F doping for LaOInF_(2)and LaOInCl_(2),respectively.
