A hallmark of systemic lupus erythematosus(SLE)is the breaking of b-cell tolerance with the generation of high-affinity autoantibodies;however,the antibody-independent features of the b-cell compartment in SLE are less *** this study,we performed an extensive examination of b-cell subsets and their proinflammatory properties in a Chinese cohort of new-onset SLE *** observed that SLE patients exhibited an increased frequency of transitional b cells compared with healthy donors and rheumatoid arthritis *** from SLE patients potently promoted the survival of transitional b cells in a type I IFN-dependent manner,which can be recapitulated by direct IFN-α***,the effect of IFN-αon enhanced survival of transitional b cells was associated with NF-κb pathway activation and reduced expression of the pro-apoptotic molecule *** b cells from SLE patients harbored a higher capacity to produce proinflammatory cytokine IL-6,which was also linked to the overactivated type I IFN *** addition,the frequency of IL-6-producing transitional b cells was positively correlated with disease activity in SLE patients,and these cells were significantly reduced after short-term standard ***,the current study provides a direct link between type I IFN pathway overactivation and the abnormally high frequency and proinflammatory properties of transitional b cells in active SLE patients,which contributes to the understanding of the roles of type I IFNs and b cells in the pathogenesis of SLE.

Efforts to mitigate the COVID-19 crisis revealed that fast,accurate,and scalable testing is crucial for curbing the current impact and that of future *** propose an optical method for directly imaging unlabeled viral particles and using deep learning for detection and *** ultrasensitive interferometric method was used to image four virus types with nanoscale optical path-length *** these data with fluorescence images for ground truth,we trained semantic segmentation models based on U-Net,a particular type of convolutional neural *** trained network was applied to classify the viruses from the interferometric images only,containing simultaneously SARS-CoV-2,H1N1(influenza-A virus),HAdV(adenovirus),and ZIKV(Zika virus).Remarkably,due to the nanoscale sensitivity in the input data,the neural network was able to identify SARS-CoV-2 *** other viruses with 96%*** inference time for each image is 60 ms,on a common graphic-processing *** approach of directly imaging unlabeled viral particles may provide an extremely fast test,of less than a minute per *** the imaging instrument operates on regular glass slides,we envision this method as potentially testing on patient breath *** necessary high throughput can be achieved by translating concepts from digital pathology,where a microscope can scan hundreds of slides automatically.

Spectroscopic Characterization and Quantitative Estimation of Natural Weathering of Silicates in Sediments of Dikrong River, India

The sediments samples were collected from the Dikrong River at various sites to assess the weathering nature and mineral characterization. The Fourier Transform Infrared (FTIR) and X-ray fluorescence (XRF) spectroscopic techniques have been used to characterization of minerals in the sediment samples. The plagioclase index of alteration (PIA), chemical index of alteration (CIA) and index of compositional variation (ICV) are investigated for evaluating the weathering nature in the sediment. The obtained results show the presence of quartz, feldspar in different structure and kaolinite as major minerals. Carbonates and organic carbon are found as minor minerals. The correlations of SiO2 with major elements are authenticated the presence of bulk quartz grains and primary depositional environment. The presence of metamorphosed pyrophanite (MnTiO3) in the adjoined areas is reported. The presence of infrared absorption peaks in between 1611 - 1622 cm?1 in this study is indicative to the weathered metamorphic origin of the silicate minerals. The index of compositional variation indicates the presence of less clay minerals and more rock forming minerals such as plagioclase and alkali-feldspar. The obtained results exhibit the area belongs to the intermediate silicate weathering.

Physio-biochemical and Genetic Exploration for Submergence Tolerance in Rice (<i>Oryza sativa</i>L.) Landraces with Special References to <i>Sub</i>1 Loci

In the present study a group of four indigenous and less popular rice genotypes (Meghi, Panibhasha, Jabra and Sholey) reported by growers as submergence tolerant lines from flood prone areas of south bengal were explored through study of nodal anatomy, physio-biochemical screening under submergence and genotyping with submergence tolerance linked rice microsatellite loci (RM loci). To identify the different allelic forms of different Sub1 compnents (Sub1A, Sub1b and Sub1C) among the studied lines, the genomic DNA of individual genotypes was amplified with three ethylene response factor like genes from Sub1 loci, located on rice chromosome 9. From the different physio-biochemical experiments performed in this investigation, it has been shown that Meghi and Jabra are the two probable potent genotypes which share common properties of both submergence tolerant and deep water nature whereas rest two genotypes (Sholey and Panibhasha) behave like typical deep water rice. The submergence tolerance property of Meghi was also confirmed from submergence tolerance linked SSR based genotyping by sharing with FR13A for some common alleles as reflected in fingerprint derived dendrogram. The rest of the genotypes shared a number of alleles and were included in a separate cluster. The common behaviour of Meghi and FR13A under submergence was also confirmed from genetic study of Sub1 loci through sharing of some common alleles for three Sub1 components (Sub1A, Sub1b and Sub1C loci). One SSR loci (RM 285) was identified as a potent molecular marker for submergence tolerance breeding programme involving these two selected rice lines (Meghi and Jabra) as donor plant through marker assisted selection.

background & Aims:-blockade of immune checkpoints to reinvigorate the tumor infiltrating CD8 T cells, especially exhausted CD8 T cells, through PD1-PDL1 pathway is one of the most promising immunotherapies. Here we investigated the characteristics of tumor infiltrating CD8 exhausted T cells of hepatocellular carcinoma(HCC) patients and its clinical significance for ***: We collected fresh HCC samples(T) with adjacent tumor-free tissues(P) and blood(b) specimens from 41 patients who underwent surgical resection at Zhongshan Hospital. CD8 T cells cytokine secretion and situ expression pattern was detected. The clinical prognosis was evaluated by Cox regression and Kaplan-Meier analysis. Results: Upon the expression level of PD1 and TIM3, tumor infiltration CD8 T cells were classified to three distinct subpopulations: PD1-negative(PD1-), PD1-intermediate(PD1 Int) and PD1-high(PD1 ). Compared with PD1-and PD1, PD1 CD8 T cells were significantly abundant in T than P and b, and correlated more aggressive clinical features. They expressed multi-inhibitory receptors including TIM3, CTLA4 and LAG3, and showed up-regulated expression of exhaustion related transcription factors, such as Eomes and blimp-1. Moreover, pro-inflammatary cytokines were decreased while anti-inflammatary cytokine IL-10 was up-regulated. Intriguingly, PD1 and TIM3PD1 CD8 T cells were correlated with poor prognosis and more adjacent to CD68PDL1 macrophage than PD1CD8 T cells in spatial ***: PD1 was heterogeneously expressed in tumor infiltrating CD8 T cells in HCC. PD1 CD8 T cells were highly expressed in tumor and correlated with exhausted phenotypes. In spatial, CD68PDL1 macrophage might interact with PD1 and TIM3PD1 CD8 T cells and cause CD8 T cells exhausted besides tumor cells. PD1 and TIM3PD1 could be a potential biomarker to stratify patients for optimal therapy.

Dosimetric Analysis of Three Different Radiotherapy Techniques in Patients with breast Cancer and Their Impact on Organs at Risk (OAR)

Aim: Dosimetric analysis of three different Radiotherapy techniques in patients with breast Cancer and their impact on OAR’s. Materials and Methods: 12 patients of Carcinoma breast who received breast radiotherapy were selected for analysis. Computed tomography (CT) simulation image data sets were retrieved. Planning target Volume (PTV), heart and ipsilateral lung were contoured for planning and analysis of doses. Three different plans using conventional bi-tangential fields were prepared, Plan 1 with SAD full beam with wedge (SAD-FbW), Plan 2 with SSD half beam with wedge (SSD-HbW) and plan 3 with SSD half beam without wedge techniques (SSD-HbO) in CMS Xio TPS. Statistical analysis was done using SPSS version 16.0. Results: The PTV coverage was significantly better in SAD-FbW techniques when compared with the other two techniques i.e. SSD-HbW (mean = 92.33, SD = 4.69, p = 0.005) and SSD-HbO (mean = 75.05, SD = 11.92, p = 0.002). The mean heart doses were significantly better in SAD-FbW compared to SSD-HbW (mean = 3.75, SD = 2.27, p = 0.017) but in SSD-HbO technique mean heart doses were better than SAD-FbW technique (mean = 3.27, SD = 1.94, p = 0.004). Similarly, the left lung V20 values were significantly better in SSD-HbO technique than SAD-FbW technique (mean = 11.75, SD = 4.34, p = 0.004) but there was statistically insignificant difference between the SAD-FbW and SSD-HbW techniques. The treatment monitor units were significantly less in SAD-FbW compared to SSD-HbW but SSD-HbO has less MU compared to SAD-FbW technique. Conclusion: SAD Full beam with wedge technique is practically better than SSD Half beam technique with or without wedge without compromising PTV coverage considering other dosimetric parameters. We also recommend that centers treating patient with half beam SSD techniques should shift immediately to SAD techniques.

背景与目的:诱导共刺激分子(inducible costimulator,ICOS)属于b7-CD28免疫球蛋白超家族成员,表达于活化T细胞表面,在T细胞的激活和免疫应答中发挥重要作用;但目前对于ICOS及ICOS^+T细胞在肝细胞癌(hepatocellular carcinoma,HCC)肿瘤组织中的表达及其意义尚不清楚。该研究旨在测定ICOS、ICOS^+T细胞在HCC患者肿瘤组织中的表达情况并探讨其预后价值。方法:应用组织芯片(tissue microarrays,TMAs)和多重免疫组织化学技术(multiplex immunohistochemistry,mIHC)检测2006-2007年在复旦大学附属中山医院接受手术治疗的358例原发性HCC患者肿瘤组织和癌旁组织中ICOS^+细胞、ICOS^+CD4^+及ICOS^+CD8^+T细胞的浸润密度和ICOS^+细胞占CD4^+、CD8^+T细胞的百分比。应用KaplanMeier法和多因素COX回归模型分析上述指标对患者预后的影响。结果:与癌旁组织相比,ICOS^+细胞和ICOS^+CD4^+T细胞在肿瘤组织中浸润数量显著增加(P<0.000 1和P=0.009 1),而ICOS^+CD8^+T细胞则呈相反的降低趋势(P=0.033);在肿瘤中,ICOS^+CD4^+T细胞的浸润程度显著高于ICOS^+CD8^+T细胞。此外,ICOS^+CD4^+和ICOS^+CD8^+T细胞占各自T细胞亚群的百分比在肿瘤组织中均显著增加(P均<0.000 1)。预后分析发现,肿瘤组织中ICOS^+细胞、ICOS^+CD4^+及ICOS^+CD8^+T细胞浸润高的患者总生存期(overall survival,OS)更长,多因素分析证实上述指标是HCC的独立预后良好因素。结论:ICOS在HCC患者的肿瘤组织中高表达,且ICOS^+细胞、ICOS^+CD4^+及ICOS^+CD8^+T细胞是OS的独立预后良好因素,提示上述指标可作为新的HCC预后免疫标志物。
