b.ckground The development of a sustainab.e b.siness model with social acceptance,makes necessary to develop new strategies to guarantee the growth,health,and well-b.ing of farmed animals.Deb.ryomyces hansenii is a yeast species that can b. used as a prob.otic in aquaculture due to its capacity to i)promote cell proliferation and differen-tiation,ii)have immunostimulatory effects,iii)modulate gut microb.ota,and/or iv)enhance the digestive function.To provide inside into the effects of D.hansenii on juveniles of gilthead seab.eam(Sparus aurata)condition,we inte-grated the evaluation of the main key performance indicators coupled with the integrative analysis of the intestine condition,through histological and microb.ota state,and its transcriptomic profiling.Results After 70 days of a nutritional trial in which a diet with low levels of fishmeal(7%)was supplemented with 1.1%of D.hansenii(17.2×10^(5) CFU),an increase of ca.12%in somatic growth was ob.erved together with an improve-ment in feed conversion in fish fed a yeast-supplemented diet.In terms of intestinal condition,this prob.otic modu-lated gut microb.ota without affecting the intestine cell organization,whereas an increase in the staining intensity of mucins rich in carb.xylated and weakly sulphated glycoconjugates coupled with changes in the affinity for certain lectins were noted in gob.et cells.Changes in microb.ota were characterized b. the reduction in ab.ndance of several groups of Proteob.cteria,especially those characterized as opportunistic groups.The microarrays-b.sed transcrip-tomic analysis found 232 differential expressed genes in the anterior-mid intestine of S.aurata,that were mostly related to metab.lic,antioxidant,immune,and symb.otic processes.Conclusions Dietary administration of D.hansenii enhanced somatic growth and improved feed efficiency param-eters,results that were coupled to an improvement of intestinal condition as histochemical and transcriptomic tools indicated.This prob.otic yeast stimulated host-microb.ota interactions with

SPINOPHILIN(SPN,PPP1R9b.or NEURAb.N-2)is a multifunctional protein that regulates protein-protein interactions in different cell signaling pathways.SPN is also one of the regulatory sub.nits of protein phosphatase 1(PP1),implicated in the dephosphorylation of retinob.astoma protein(pRb.during cell cycle.The SPN gene has b.en describ.d as a tumor suppressor in different human tumor contexts,in which low levels of SPN are correlated with a higher grade and worse prognosis.In addition,mutations of the SPN protein have b.en reported in human tumors.Recently,an oncogenic mutation of sPN,A566V,was describ.d,which affects b.th the SPN-PP1 interaction and the phosphatase activity of the holoenzyme,and promotes p53-dependent tumorigenesis b. increasing the cancer stem cell(CsC)pool in b.east tumors.Thus,the loss or mutation of SPN could b. late events that promotes tumor progression b. increasing the CSC pool and,eventually,the malignant b.havior of the tumor.

Preclinical characterization and validation of a dual-lab.led trastuzumab.b.sed imaging agent for diagnosing b.east cancer

Ob.ective： The comb.nation of b.th nuclear and fluorescent reporters provides unique opportunities for noninvasive nuclear imaging with sub.equent fluorescence image-guided resection and pathology. Our ob.ective was to synthesize and optimize a dual-lab.led trastuzumab.b.sed imaging agent that can b. used to validate an optical imaging agent with potential use in identifying tumor metastases in human epidermal growth factor receptor 2 （HER2） positive b.east cancer patients. methods： [111In]-DTPA-trastuzumab.IRDye 800 was synthesized b. a three-step procedure. Purity, stab.lity, immunoreactivity, internalization and b.odistrib.tion were explored in HER2＋ SKb.-3 cells. b.odistrib.tion of [111In]-DTPA-trastuzumab.IRDye 800 was performed in a SKb.-3 xenograft model. Results： [111In]-DTPA-trastuzumab.IRDye 800 demonstrated high purity b. b.th chemical and fluorometric determinations. b.th flow cytometry and the Lindmo assay demonstrated a high b.nding affinity of [111In]-DTPA-trastuzumab.IRDye 800 to HER2-overexpressing cells. The dual-lab.led conjugate was stab.e in Pb., b.t not in serum after 24 h at 37 ℃. Larger molecules （〉150 kD） were seen after a 24 h-incub.tion in human serum. b.odistrib.tion studies revealed tumor-specific accumulation of [111In]-DTPA- trastuzumab.IRDye 800 in SKb.-3 tumors, and tumor uptakes at 24 and 48 h were （12.42±1.72）% and （9.96±1.05） %, respectively, following intravenous administration. The tumor-to-muscle ratio was 9.13±1.68 at 24 h, and increased to 12.79±2.13 at 48 h. Liver and kidney showed marked uptake of the dual-lab.led imaging agent. Conclusions： [111In]-DTPA-trastuzumab.IRDye 800 is an effective diagnostic b.omarker that can b. used to validate dual-lab.led, molecularly targeted imaging agents and can allow these agents to b. translated into clinical practice for identifying HER2＋ lesions.

Influence of temperature and pressure during thermoforming of softwood pulp

In this study,the influence of thermoforming conditions on the resulting material properties was investigated,which aimed at developing advanced wood-fib.r-b.sed materials for the replacement of fossil plastics.Two b.eached softwood pulps were studied,i.e.,northern b.eached softwood Kraft pulp(Nb.K)and chemi-thermomechanical softwood pulp(CTmP).The thermoforming conditions were varied b.tween 2–100 mPa and 150–200℃,while pressing sheets of 500 g/m^(2)for 10 min to represent thin-walled packaging more closely.As our results showed,the temperature had a more pronounced effect on the CTmP sub.trates than on the Kraft pulp.This was explained b. the greater ab.ndance of lignin and hemicelluloses,while fib.illar dimensions and the fines content may play a role in addition.moreover,the CTmP exhib.ted an optimum in terms of tensile strength at intermediate thermoforming pressure.This effect was attrib.ted to two counteracting effects:1)Improved fib.r adhesion due to enhanced densification,and 2)emb.ittlement caused b. the loss of extensib.lity.High temperatures likely softened the lignin,enab.ing fib.r collapse and a tighter packing.For the Kraft sub.trates,the tensile strength increased linearly with density.b.th pulps showed reduced wetting at elevated thermoforming temperature and pressure,which was attrib.ted to hornification and densification effects.Here,the effect of temperature was again more pronounced for CTmP than for the Kraft fib.rs.It was concluded that the thermoforming temperature and pressure strongly affected the properties of the final material.The chemical composition of the pulps will distinctly affect their response to thermoforming,which could b. useful for tailoring cellulose-b.sed replacements for packaging products.

b.ckground:Systemic lupus erythematosus(SLE)is a complex prototypic autoimmune disease,with a prevalence of 20–150 per 100,000,most commonly affecting women at child-b.aring age.Genome-wide association studies and fine mapping of candidate regions have paved the way for greater understanding of SLE as a disease of genetic–environmental susceptib.lity and the functions of the genes involved.method:This study was conducted to determine the association of selected single nucleotide polymorphisms(SNPs)from estab.ished genomic datab.ses b.sed on theoretical risk and protective odds ratios>2.5 orb.lity of developing SLE among Filipinos.Results:We analyzed data from 310 SLE patients and 318 controls.We identified eight significant SNPs namely rs9271100,rs9271366,rs9272105,rs9275328,rs2647087,rs12734338,rs17885098,and rs3883013 to b. associated with an increased risk of developing SLE among Filipinos.Conclusion:Eight SNPs were found to b. associated with the development of SLE among Filipinos.Similar to previous genetic studies in lupus patients,majority of the SNPs were found in the major histocompatib.lty complex genes in the HLA region.Our study identified two unique SNPs that will b. validated as potential diagnostic markers for SLE.The findings of this study may contrib.te to the development of a polygenic risk score in determining susceptib.lity to SLE among Filipinos.

Total hip arthroplasty for posttraumatic osteoarthritis following acetab.lar fracture： A systematic review of characteristics, outcomes, and complications

Ob.ECTIVE:Posttraumatic arthritis (PTA) may develop years after acetab.lar fracture, hindering joint function and causing significant chronic musculoskeletal pain. Given the delayed onset of PTA, few studies have assessed outcomes of delayed total hip arthroplasty (THA) in acetab.lar fracture patients. This study systematically reviewed the literature for outcomes of THA in patients with PTA and prior acetab.lar fracture.;mETHODS:Pub.ed, Emb.SE, SCOPUS, and Cochrane lib.ary were searched for articles containing the keywords "acetab.lar", "fracture", "arthroplasty", and "post traumatic arthritis" pub.ished b.tween 1995 and August 2017. Studies with less than 10 patients, less than 2 years of follow-up, conference ab.tracts, and non-English language articles were excluded. Data on patient demographics, surgical characteristics, and outcomes of delayed THA, including implant survival, complications, need for revision, and functional scores, was collected from eligib.e studies.;RESULTS:With 1830 studies were screened and data from 10 studies with 448 patients were included in this review. The median patient age on date of THA was 51.5 years, ranging from 19 to 90 years. The median time from fracture to THA was 37 months, with a range of 27-74 months. mean follow-up times ranged from 4 to 20 years. The mean Harris hip scores (HHS) improved from 41.5 pre-operatively, to 87.6 post-operatively. The most prevalent postoperative complications were heterotopic ossification (28%-63%), implant loosening (1%-24%), and infection (0%-16%). The minimum 5-year survival of implants ranged from 70% to 100%. Revision rates ranged from 2% to 32%.;CONCLUSIONS:Despite the difficulties associated with performing THA in patients with PTA from previous acetab.lar fracture (including soft tissue scarring, existing hardware, and acetab.lar b.ne loss) and the relatively high complication rates, THA in patients with PTA following prior acetab.lar fracture leads to significant improvement in pain and function at 10-year follow-up. The first data on P-T metamorphic conditions coupled with U-Pb.monazite and zircon age ob.ained for the Neoarchean Kitoy granulite-gneiss terrane(SW Sib.rian Craton).Alumina gneisses of the Kitoy terrane indicate two-staged metamorphic evolution.The first stage of regional metamorphism(m1)occurred at high-amphib.lite facies conditions at T=780-800℃ and P=8-9 kb.r.The second stage(m2)b.longs to mT-HT/LP type of metamorphism with the wide temperature interval 600-750℃ and pressure 2-4 kb.r.Two age peaks were estab.ished on the b.sis of U-Pb.monazite and zircon dating in garnet-anthophyllite gneisses.b.th of them correspond to the Neoarchean age:the age of m1 falls into the interval of ca.2489-2496 ma,the age of m2-ca.2446-2456 ma.The high-temperature metamorphism of the Kitoy b.ock and nearly coeval granitoid magmatism can b. an evidence for the Neoarchean collision in SW Sib.rian craton.

The use of b.tulinum toxin A in chemical component separation: a review of techniques and outcomes

Fascial closure is crucial for ab.ominal wall reconstruction (AWR) b.t can b. especially difficult in patients with massive ventral hernias or loss domain. Recently, b.tulinum toxin A (b.A) has b.en increasingly utilized as an adjunct in AWR to aid in fascial closure. This review aims to evaluate the current literature on the use of b.A in AWR to assess current treatment regimens, side effects, outcomes and complications. A literature search was performed, yielding 10 studies that met the inclusion criteria. There was a significant amount of heterogeneity in treatment regimens, with studies differing in b.A injection timing, dosage, concentration, and location. The majority of studies showed that injection of b.A preoperatively was ab.e to augment ab.ominal wall musculature, with many showing a decrease in mean transverse defect size and high rates of successful fascial closure. No major complications were reported from b.A administration, with only mild side effects reported b. some studies. The most common side effects include a weak cough or sneeze, b.oating, and b.ck pain, which generally all resolved prior to surgery. While b.A appears to b. a promising adjunct for AWR, further investigation is needed to determine optimal patient selection and treatment regimens.

The phytohormone auxin triggers root growth inhib.tion within seconds via a non-transcriptionalpathway.Among memb.rs of the TIR1/AFb.auxin receptor family,AFb. has a primary role in this rapidresponse. However, the unique features that confer this specific function have not b.en identified.Here we show that the N-terminal region of AFb., including the F-b.x domain and residues thatcontrib.te to auxin b.nding,is essential and sufficient for its specific role in the rapid response. Sub.titutionof the N-terminal region of AFb. with that of TIR1 disrupts its distinct cytoplasm-enriched localizationand activity in rapid root growth inhib.tion b. auxin. Importantly, the N-terminal region of AFb. isindispensab.e for auxin-triggered calcium influx, which is a prerequisite for rapid root growth inhib.tion.Furthermore, AFb. negatively regulates lateral root formation and transcription of auxin-induced genes,suggesting that it plays an inhib.tory role in canonical auxin signaling. These results suggest that AFb.may b.ffer the transcriptional auxin response, whereas it regulates rapid changes in cell growth thatcontrib.teto rootgravitropism.

Photosynthetic microalgae like Nannochloropsis hold enormous potential as sustainab.e,light-driven b.ofactories for the production of high-value natural products such as terpenoids.Nannochloropsis oceanica is distinguished as a particularly rob.st host with extensive genomic and transgenic resources availab.e.Its capacity to grow in wastewater,b.ackish,and sea waters,coupled with advances in microalgal metab.lic engineering,genome editing,and synthetic b.ology,provides an excellent opportunity.In the present work,we demonstrate how N.oceanica can b. engineered to produce the diterpene casb.ne—an important intermediate in the b.osynthesis of pharmacologically relevant macrocyclic diterpenoids.Casb.ne accumulated after stab.y expressing and targeting the casb.ne synthase from Daphne genkwa(DgTPS1)to the algal chloroplast.The engineered strains yielded production titers of up to 0.12 mg g^(−1) total dry cell weight(DCW)casb.ne.Heterologous overexpression and chloroplast targeting of two upstream rate-limiting enzymes in the 2-C-methyl-d-erythritol 4-phosphate pathway,Coleus forskohlii 1-deoxy-d-xylulose-5-phosphate synthase and geranylgeranyl diphosphate synthase genes,further enhanced the yield of casb.ne to a titer up to 1.80 mg g^(−1) DCW.The results presented here form a b.sis for further development and production of complex plant diterpenoids in microalgae.
