The behavior of the 2006 ten Tusscher-Panfilov model of human ventricular myocytes under the impact of periodic excitation impulses was studied in the beatbox simulation environment. The cardiomyocyte model has a limited susceptibility to an forced higher frequency excitation rhythm. A high-frequency excitation rhythm can be forced by gradually increasing the frequency of excitation impulses. The mechanism of defibrillation pulse impact consists of presumably prolonging the refractoriness of cardiomyocytes which undermines their susceptibility for a long time to a forced high-frequency rhythm of fibrillation, as a result for which they hinder the propagation of a fibrillation wave. This is the only mechanism of defibrillation that was identified during the simulation. The threshold energy of a depolarizing defibrillation pulse prolonging the refractoriness of the cardio-myocyte varies depending on a delay relative to the excitation impulse (the excitation cycle phase) in a wide range (the maximum value exceeds the minimum by several thousand times). The results show differences in the mechanisms of impact on a cardiomyocyte between an excitation impulse and a monophasic defibrillation pulse.

The Kuvasali landslide disaster in Malava, Western Kenya, which caused harmful geo-environmental hazards to the villagers in August 2007, formed the premise of this paper. The existing landslide vulnerability occurrence ratings have been identified and documented using participatory approaches to ascertain the adoptive indigenous methods, which helped mitigation against the negative impacts by determining the slope steepness using Abney level. The geological set up, settlement and climatic factors, relating to landslides, revealed that the affected homesteads were on a steep slope of approximately 70%, with underlying metamorphosed basement rocks comprising ofgneisses, schists and granite intrusions. The steep escarpment overlying these rocks, where the landslide occurred, consists of volcanic rocks and black cotton clayey soils. The slope has scattered vegetation cover due to farming, while the low-lying area is planted with sugar cane and maize plantations. The landslide disaster led to loss of lives, injuries and damage to property. Natural resources, including water points, suffered siltation. Mitigation measures such as geo-hazard mapping and zoning, awareness training and campaigns for the local people focusing on hazard-prone areas resulting from poor land use practices have been recommended.

There is now strong evidence that the paternal contribution to offspring phenotype at fertilisation is more than just DNA. However, the identity and mechanisms of this nongenetic inheritance are poorly understood. One of the more important questions in this research area is： do changes in sperm DNA methylation have phenotypic consequences for offspring？ We have previously reported that offspring of obese male rats have altered glucose metabolism compared with controls and that this effect was inherited through nongenetic means. Here, we describe investigations into sperm DNA methylation in a new cohort using the same protocol. Male rats on a high-fat diet were 30% heavier than control-fed males at the time of mating （16-19 weeks old, n = 14/14）. A small （0.25%） increase in total 5-methyl-2＇-deoxycytidine was detected in obese rat spermatozoa by liquid chromatography tandem mass spectrometry. Examination of the repetitive fraction of the genome with methyI-CpG binding domain protein-enriched genome sequencing （MbD-Seq） and pyrosequencing revealed that retrotransposon DNA methylation states in spermatozoa were not affected by obesity, but methylation at satellite repeats throughout the genome was increased. However, examination of muscle, liver, and spermatozoa from male 27-week-old offspring from obese and control fathers （both groups from n = 8 fathers） revealed that normal DNA methylation levels were restored during offspring development. Furthermore, no changes were found in three genomic imprints in obese rat spermatozoa. Our findings have implications for transgenerational epigenetic reprogramming. They suggest that postfertilization mechanisms exist for normalising some environmentally-induced DNA methylation changes in sperm cells.

Placic horizons, defined as thin, wavy, hardened layers of iron and organic matter, are rare within the United States, occurring only in Washington, Alaska, and Hawaii. While ironstone is common in many soils of the southeastern United States, it is not known to contain appreciable organic matter. As a pilot study evaluating the justification for a larger study on ironstone in Louisiana, a 40 m lateral exposure with suspected placic horizons was evaluated in Vernon Parish, Louisiana. Results of laboratory analysis show elevated levels of iron and organic matter in the suspect horizons that meet the criteria of placic horizons as defined by the Soil Survey Staff. based on the results of this study, additional evaluation of multiple pedons with similar features is warranted. Should additional pedons demonstrate similar properties, a new great group of 'Petrudepts' would be needed to describe both the placic horizons in the pedon and the udic moisture regime in which they occur.

A case presentation indicating the importance of “happiness” in childhood causing memory block until the patent presented with probable mixed vascular and neurodegenerative memory loss at 60 years of age is presented to highlight the role of emotional factors in causing the disease. The question of whether Alzheimer’s disease is an adaptability disorder is raised, given the patient blocked out her memory of her childhood experience. The importance of “happiness” as a treatment goal raises issues of advocacy and Guardianship as well as capacity, which is addressed by actual case reference and court action in defence of the patient’s rights to have their wishes respected and observed. Functional mental capacity assessment, using the Functional Mental State Measure (FMSM) gives a greater indication of neuronal reserve than standard cognitive testing, as it helps to unravel the dilemma associated with pure cognitive assessment in Alzheimer’s Disease as well as vascular dementia patients and patients who, despite retained and intact functional capacity and ability to express their wishes, i.e. “best interest”, are “wrongly” placed under Guardianship. Maladaptive responses, to control the change in external environment that are sensed or perceived, and which lead to disorder or to susceptibility to disease, exemplify a General Systems Theory approach, in which appropriate and adequate responses to environmental change, in behavioural terms, by a person, whether independently, or as the recipient or giver, or both, leads to functional interaction and happiness.

background:Children with moderate-to-severe asthma continue to have disease complications despite the receipt of standardof-care *** monoclonal antibody dupilumab has been approved for the treatment of adults and adolescents with asthma as well as with other type 2 inflammatory ***:In this 52-week phase 3,randomized,double-blind,placebo-controlled trial,we assigned 408 children between the ages of 6 and 11 years who had uncontrolled moderate-to-severe asthma to receive a subcutaneous injection of dupilumab(at a dose of 100 mg for those weighing≤30 kg and 200 mg for those weighing>30 kg)or matched placebo every 2 *** the children continued to receive a stable dose of standard background *** primary end point was the annualized rate of severe asthma *** end points included the change from baseline in the percentage of predicted prebronchodilator forced expiratory volume in 1 second(ppFEV1)at week 12 and in the score on the Asthma Control Questionnaire 7 Interviewer-Administered(ACQ-7-IA)at week *** points were evaluated in the two primary efficacy populations who had either a type 2 inflammatory asthma phenotype(≥150 blood eosinophils per cubic millimeter or a fraction of exhaled nitric oxide of≥20 ppb at baseline)or a blood eosinophil count of at least 300 cells per cubic millimeter at baseline.
