This study examined if the anti-tumorigenesis effect of Exendin-4 in HT29 and HCT116 colorectal cancer(CRC)involves modulation of SIRT1 and Akt/GSR3K/β-catenin/NF-κB ***29 and HCT116 cells were treated either with increasing levels of Exendin-4(0.0-200μM)or with Exendin-4(at its IC50)in the presence or absence of EX-527(10μM/a selective SIRT1 inhibitor)or Exendin-4(9-39)amide(E(9-39)A)(1μM/an Exendin-4 antagonist).In a dose-dependent manner,Exendin-4 inhibited cell survival,but enhanced levels of lactate dehydrogenase(LDH)and single-stranded DNA(ssDNA)in both HT29 and *** both cell lines and at it has an IC50(45μM for HT29 and 35μM for HCT1165),Exendin-4 also significantly reduced cell survival,migration,and invasion of both cell types,with no effect on the expression GLP-1 receptors(GLPRs)nor of the activity of *** these doses,Exendin-4 also increased the expression of SIRT1 but reduced the acetylation of NF-κB and the expression of Bax and cleaved caspase-3 and in both cell ***,protein levels of p-GS3Kβ(Ser9),total and acetylatedβ-catenin,and Anix2 were significantly decreased,but levels of p-GS3Kβ(Ser9)and p-β-catenin(Ser33/37/Thr41)were significantly increased in both HT29 and HCT116-exendin-4 treated *** the effects exerted by Exendin-4 were completely prevented by Ex527 or E(9-39)*** conclusion,Exendin-4 suppresses the tumorigenesis of HT29 and HCT116 CRC cell activation of GS3Kβ-induced inhibition ofβ-catenin and NF-κβin a SIRT1-dependent mechanism.

The influence of cooling rate on the microstructure and mechanical properties of two new ultrahigh-strength steels(UHSSs)with different levels of C,Cr and Ni has been evaluated for the as-cooled and untempered *** UHSS had higher contents of C and Cr,while the other one had a higher Ni *** the basis of dilatation curves,microstructures,macrohardness and microhardness,continuous cooling transformation diagrams were constructed as a guide to heat treatment *** rates(CRs)of 60,1 and 0.01°C/s were selected for more detailed *** characterization was made by laser scanning confocal microscopy,field emission scanning electron microscopy combined with electron backscatter diffraction,electron probe microanalysis and X-ray *** properties were characterized using macrohardness,tensile and Charpy V-notch impact *** with the higher C and Cr contents showed lower transformation temperatures and slower bainite formation kinetics than that with the higher Ni *** cooling rates led to lower volume fractions and carbon contents of retained austenite together with finer prior austenite grain size,as well as effective final grain size and lath *** changes were accompanied by higher yield and tensile *** best combinations of strength and toughness were obtained with martensitic microstructures and by avoiding the formation of granular bainite accompanied by proeutectoid carbides at low *** the cooling rates studied,UHSS with the higher C and Cr contents showed the higher hardness and strength but at the cost of toughness.

We propose a high-order enriched partition of unity finite element method for linear and nonlinear time-dependent diffusion *** solution of this class of problems often exhibits non-smooth features such as steep gradients and boundary layers which can be very challenging to recover using the conventional low-order finite element methods.A class of steady-state exponential functions has been widely used for enrichment and its performance to numerically solve these challenges has been ***,these enrichment functions have been used only in context of the standard h-version refinement or the so-called q-version *** this paper we demonstrate that the p-version refinement can also be a very attractive option in terms of the efficiency and the accuracy in the enriched partition of unity finite element ***,the transient diffusion problem is integrated in time using a semi-implicit scheme and the semi-discrete problem is then integrated in space using the p-version enriched finite *** results are presented for three test examples of timedependent diffusion problems in both homogeneous and heterogeneous *** computed results show the significant improvement when using the p-version refined enriched approximations in the finite element *** addition,these results support our expectations for a robust and high-order accurate enriched partition of unity finite element method.

Dipper throated optimization(DTO)algorithm is a novel with a very efficient metaheuristic inspired by the dipper throated *** has its unique hunting technique by performing rapid bowing *** show the efficiency of the proposed algorithm,DTO is tested and compared to the algorithms of Particle Swarm Optimization(PSO),Whale Optimization Algorithm(WOA),Grey Wolf Optimizer(GWO),and Genetic Algorithm(GA)based on the seven unimodal benchmark ***,ANOVA and Wilcoxon rank-sum tests are performed to confirm the effectiveness of the DTO compared to other optimization ***,to demonstrate the proposed algorithm’s suitability for solving complex realworld issues,DTO is used to solve the feature selection *** strategy of using DTOs as feature selection is evaluated using commonly used data sets from the University of California at Irvine(UCI)*** findings indicate that the DTO outperforms all other algorithms in addressing feature selection issues,demonstrating the proposed algorithm’s capabilities to solve complex real-world situations.

This paper presents an experimental investigation on the flexural behavior of cross-connected brick masonry infill wall panels supported on reinforced concrete beam grids above and below the *** experimental program was comprised of six wall *** effect of change in lower beam stiffness relative to the wall and the geometry of the main walls were *** the results of the experimental tests,the increase in the depth of the lower beam grid reduces the deflection,resulting in an increase in the load carrying capacity of the ***,the stiffness of the main walls affects the deflection and the failure load of the cross walls.

Ki-67 immunohistochemical expression in mandibular ameloblastoma: A prognostic indicator for local recurrence

Objective: This study used Ki-67 to evaluate the proliferative activity of ameloblastoma in an attempt to provide a scientific basis for any differences in the biologic behaviour that exists between the different types of ameloblastoma and correlate the results with the recurrence rate. Study design: We assessed Ki-67 expression by immunohistochemistry in 32 cases of mandibular ameloblastoma. Formalin-fixed, paraffinembedded blocks were sectioned and used for Haematoxylin & Eosin (H&E) staining. H&E slides were used to re-evaluate the histological types of the ameloblastoma. Immunohistochemical analysis for Ki-67 with a labelled streptavidin-biotin-peroxidase complex technique was performed on tumour sections using Monoclonal Mouse Anti-Human Ki-67 Antigen (clones MIB1, code M7240, Dako Corporation). Lymph nodes (lymphoma) were used as positive control for Ki-67. Results: The specimens comprised 18 cases of Follicular ameloblastoma, 7 cases of Cystic ameloblastoma, 4 cases of plexiform ameloblastoma, and 3 cases of Acanthotic ameloblastoma. The results showed that cellular proliferative activity varied within the ameloblastoma types. There was no statistically significant difference in the Ki-67 Acanthotic, cystic and follicular types of ameloblastoma, while plexiform type showed statistically significantly higher levels than the other 3 types. The mean Ki-67 labelling index of plexiform ameloblastoma ranged from 13 to 30 with a mean of 19.75. The mean values of Ki-67 were statistically significantly higher in recurrent than?non-recurrent cases. Conclusion: The significant expression of Ki-67 could provide useful prognostic markers for proliferative activity and good prognostic indicators for recurrence rate of mandibular ameloblastoma.

This paper aims to assess soil fertility and risk contamination parameters in response to increased amendment rates of sewage sludge application,under semi-arid climatic conditions of Morocco,using two soil textures:soil A(clay-silty)and soil B(sandy-clay)over two *** this end,sewage sludge and soils treatments physicochemical parameters,heavy metals content,degree of pollution and associated risks were evaluated and compared to controlled *** results showed that all studied parameters changed significantly(pPb>Zn>Ni>Cd>***,their concentrations were below the critical values established by the European Union for the agronomic use of *** obtained results of assessed parameters indicating SS degree of pollution and associated risks showed a low to a moderate contamination risks of heavy ***,environmental risks of sewage sludge application will be related to increase of heavy metals mainly in a long-term agricultural ***,optimal management of sewage sludge application is recommended to ensure sustainability and benefits at the same time to master the contamination risks.

Network management and multimedia data mining techniques have a great interest in analyzing and improving the network traffic *** recent times,the most complex task in Software Defined Network(SDN)is security,which is based on a centralized,programmable ***,monitoring network traffic is significant for identifying and revealing intrusion abnormalities in the SDN ***,this paper provides an extensive analysis and investigation of the NSL-KDD dataset using five different clustering algorithms:K-means,Farthest First,Canopy,Density-based algorithm,and Exception-maximization(EM),using the Waikato Environment for Knowledge Analysis(WEKA)software to compare extensively between these five ***,this paper presents an SDN-based intrusion detection system using a deep learning(DL)model with the KDD(Knowledge Discovery in Databases)***,the utilized dataset is clustered into normal and four major attack categories via the clustering ***,a deep learning method is projected for building an efficient SDN-based intrusion detection *** results provide a comprehensive analysis and a flawless reasonable study of different kinds of attacks incorporated in the KDD ***,the outcomes reveal that the proposed deep learning method provides efficient intrusion detection performance compared to existing *** example,the proposed method achieves a detection accuracy of 94.21%for the examined dataset.

This study highlights the regulatory potential antibacterial and antiradical of Origanum elongatum essential oil(EO),an endemic medicinal plant of Morocco used for its various *** chemical composition of the EO was characterized using gas chromatography-mass spectrometry(GC-MS).The antibacterial activity against different agricultural phytopathogens was determined by disc diffusion and microatmosphere methods,as well as by the determination of minimum inhibitory concentrations(MIC)and minimum bactericidal concentration(MBC),while the antioxidant activity was evaluated by DPPH and FRAP *** complement the experimental analyses,a molecular docking approach was used to predict and elucidate the mechanisms of action of the identified bioactive com-pounds,both for their antioxidant and antibacterial *** GC-MS analysis revealed a chemical composition dominated by the major compounds:p-cymene-2-ol(25.31%),thymol(23.88%),andγ-terpinene(19.26%).Furthermore,antibacterial analyses performed using different methodological approaches(disc diffusion,microatmosphere,MIC,and MBC)showed significant inhibitory activity against all phytopathogens ***,*** EO exhibited interesting antioxidant ability with an IC_(50) value of 168.251.14μg/mL for DPPH assay and EC_(50) value±of 164.221.04μg/mL for FRAP ***,in silico molecular docking demonstrated further insights into±the interactions between the oil’s active components and bacterial targets,supporting its mode of *** in-depth characterization highlights the potential of *** EO as a natural alternative for the biocontrol of plant *** opens new perspectives for developing natural solutions to protect crops against plant diseases.
