Cellular physiological approach for treatment of gastric cancer

Recent studies show that ion channels/transporters play important roles in fundamental cellular functions that would be involved in the cancer process. We review the evidence for their expression and functioning in human gastric cancer (GC), and evaluate the potential of cellular physiological approach in clinical management. Various types of ion channels, such as voltage-gated K+ channels, intracellular Cl- channels and transient receptor potential channels have been found to express in GC cells and tissues, and to control cell cycles. With regard to water channels, aquaporin 3 and 5 play an important role in the progression of GC. Regulators of intracellular pH, such as anion exchanger, sodium-hydrogen exchanger, vacuolar H+-ATPases and carbonic anhydrases are also involved in tumorigenesis of GC. Their pharmacological manipulation and gene silencing affect cellular behaviours, suggesting their potential as therapeutic targets for GC. Our studies indicate the intracellular Cl- concentration could act as a mediator of cellular signaling and control cell cycle progression in GC cells. Further, we demonstrate the cytocidal effects of hypotonic shock on GC cells, and indicate that the blockade of Cl- channels/transporters enhances these effects by inhibiting regulatory volume decrease. A deeper understanding of molecular mechanisms may lead to the discovery of these cellular physiological approaches as a novel therapeutic strategy for GC.

Activation of JAK-STAT pathway is required for platelet-derived growth factor-induced proliferation of pancreatic stellate cells

AM: To clarify the role of Janus kinase-signal transducers and activators of transcription (JAK-STAT) pathway in platelet-derived growth factor (PDGF) induced proliferation in activated pancreatic stellate cells (PSCs). METHODS: PSCs were isolated from rat pancreas tissue, and used in their culture-activated, myofibroblast-like phenotype. STAT-specific binding activity was assessed by electrophoretic mobility shift assay. Activation of Src, JAK2, STAT1, STAT3, and ERK was determined by Western blotting using anti-phosphospecific antibodies. Cell proliferation was assessed by measuring the incorporation of 5-bromo-2'-deoxyuridine. RESULTS: PDGF-BB induced STAT-specific binding activity, and activation of Src, JAK2, STAT1, STAB, and ERK. Ethanol and acetaldehyde at clinically relevant concentrations decreased basal activation of JAK2 and STAT3. PDGF-induced activation of STAT1 and STAT3 was inhibited by a Src inhibitor PP1 and a JAK2 inhibitor AG490, whereas PDGF-induced activation of ERK was inhibited by PP1, and not by AG490. PDGF-induced proliferation was inhibited by PP1 and AG490 as well as by STAT3 antisense oligonucleotide. CONCLUSION: PDGF-BB activated JAK2-STAT pathway via Src-dependent mechanism. Activation of 3AK2-STAT3 pathway, in addition to ERK, may play a role in PDGF-induced proliferation of PSCs.

Leaf Positioning of Arabidopsis in Response to Blue Light

Appropriate leaf positioning is essential for optimizing photosynthesis and plant growth. However, it has not been elucidated how green leaves reach and maintain their position for capturing light. We show here the regulation of leaf positioning under blue light stimuli. When 1-week-old Arabidopsis seedlings grown under white light were transferred to red light （25 i, mol m^-2 s^-1） for 5 d, new petioles that appeared were almost horizontal and their leaves were curled and slanted downward. However, when a weak blue light from above （0.1 i, mol m^-2 s^-1） was superimposed on red light, the new petioles grew obliquely upward and the leaves were flat and horizontal. The leaf positioning required both phototropinl （photl） and nonphototropic hypocotyl 3 （NPH3）, and resulted in enhanced plant growth. In an nph3 mutant, neither optimal leaf positioning nor leaf flattening by blue light was found, and blue light-induced growth enhancement was drastically reduced. When blue light was increased from 0.1 to 5 i, mol m^-2 s^-1, normal leaf positioning and leaf flattening were induced in both photl and nph3 mutants, suggesting that phot2 signaling became functional and that the signaling was independent of photl and NPH3 in these responses. When plants were irradiated with blue light （0.1 i, mol m^-2 s^- 1） from the side and red light from above, the new leaves became oriented toward the source of blue light. When we transferred these plants to both blue light and red light from above, the leaf surface changed its orientation to the new blue light source within a few hours, whereas the petioles initially were unchanged but then gradually rotated, suggesting the plasticity of leaf positioning in response to blue light. We showed the tissue expression of NPH3 and its plasma membrane localization via the coiled-coil domain and the C-terminal region. Establishment and characterization of a rat pancreatic stellate cell line by spontaneous immortalization

AIM: Activated pancreatic stellate cells (PSCs) have been implicated in the pathogenesis of pancreatic fibrosis and inflammation. Primary PSCs can be subcultured only several times because of their limited growth potential. A continuous cell line may therefore be valuable in studying molecular mechanisms of these pancreatic disorders. The aim of this study was to establish a cell line of rat PSCs by spontaneous ***: PSCs were isolated from the pancreas of male Wistar rats, and conventional subcultivation was performed repeatedly. Telomerase activity was measured using the telomere repeat amplification protocol. Activation of transcription factors was assessed by electrophoretic mobility shift *** of mitogen-activated protein (MAP) kinases was examined by Western blotting using anti-phosphospecific antibodies. Expression of cytokine-induced neutrophil chemoattractant-1 was determined by enzyme ***: Conventional subcultivation yielded actively growing cells. One clone was obtained after limiting dilution,and designated as SIPS. This cell line has been passaged repeatedly more than 2 years, and is thus likely *** cells retained morphological characteristics of primary,culture-activated PSCs. SIPS expressed α-smooth muscle actin, glial acidic fibrillary protein, vimentin, desmin, type Ⅰ collagen, fibronectin, and prolyl hydroxylases. Telomerase activity and p53 expression were negative. Proliferation of SIPS cells was serum-dependent, and stimulated with platelet-derived growth factor-BB through the activation of extracellular signal-regulated kinase. Interleukin-1β activated nuclear factor-κB, activator protein-1, and MAP ***-1β induced cytokine-induced neutrophil chemoattractant-1 expression through the activation of nuclear factor-κB and MAP ***: SIPS cells can be useful for in vitro studies of cell biology and signal transduction of PSCs.

本文在政策分析的基础上,建立VAR-BEKK-GARCH模型进行实证分析。首先,国际玉米价格的波动幅度比中国价格波动幅度大,临储政策取消后,中国价格波动幅度变大。其次,临储期间,中国价格的预期波动可以影响国际价格的波动,而国际价格的随机波动可以影响中国价格的波动。在生产者补贴期间,中国价格的预期波动和随机波动均可影响国际价格的波动。再次,中国价格的影响力在中国市场和进口市场占据主导地位,与国产玉米供给本国市场的数量地位相符。最后,中国玉米进口的不断增加将降低对本国市场价格的主导能力,需重视通过“期货+保险”规避价格风险,并分散玉米进口源,以提高市场主导力。

Heat transfer in a tapered fluidized bed of biomass particles with pulsed gas flow

Bed-to-surface heat transfer of pure biomass particles in a pulsed fluidized bed with a tapered bottom section was investigated. Three biomass species — Douglas fir, pine, and switchgrass — were studied under various operating conditions. Their heat transfer coefficients were found to be closely associated with hydrodynamics dominated by gas pulsations. A higher superficial gas velocity generally yielded better gas–solid contact and higher heat transfer rates. A moderately increasing pulsation frequency promoted convective heat transfer of particles but also reduced pulsation intensity, leading to undesired flow behaviours such as channelling and partial defluidization. The study of the pulsation duty cycle revealed that, for cohesive particles, a smaller duty cycle was preferred to generate powerful pulsations to break up inter-particle forces. Moreover, a duty cycle increase allowed higher gas throughput as long as a suitable fluidization was maintained. The addition of finer particles to a coarse fraction increased particle mobility, and subsequently heat transfer, which also explained the higher heat transfer coefficients of switchgrass as it contained more fines compared with fir and pine. Experimental results in the tapered bed were also compared with those of non-tapered geometry where a 10%–20% increase in heat transfer was observed.

Design and Prototyping of Micro Centrifugal Compressor for Ultra Micro Gas Turbine

In order to establish the design methodology of an ultra micro centrifugal compressor, which is the most important component of an ultra micro gas turbine unit, a 10 times size of the final target compressor （impeller outer diameter 40 mm, corrected rotational speed 220,000 r/min） was designed. The problems to be solved for downsizing were examined and a 2-dimensional impeller was chosen as the first model due to its productivity. The conventional 1D prediction method and CFD were used. The prototyped compressor was tested by using cold air at the reduced speed of 110,000 r/min. Following to the 10 times model, a 5 times size of the final target model having fully 3-dimensional shape （impeller outer diameter 20mm, corrected rotational speed 500,000 r/min） was designed and tested by using hot gas at the reduced speed of 250,000 r/min.

Influence of Alloy Elements on the Osteoconductivity of Anodized Ti-29Nb-13Ta-4.6Zr Alloy

Anodizing is expected to be an effective method to improve the osteoconductivity of the Ti-29Nb-13Ta-4.6Zr (TNTZ) alloy because the bioactivity of anodized Ti is good. However, it is not known how the alloy elements influence the surface roughness, composition, hydrophilicity, and osteoconductivity of the anodized film on the Ti alloy. In this study, we investigated the effects of anodizing on the surface properties and the osteoconductivity of the anodized TNTZ alloy, focusing on the functions of the individual alloy elements. The anodized oxides of the Nb, Ta, and Zr metals were hydrophobic at all the voltages applied, in contrast to the anodized oxide of Ti. As well as pure Ti, a TiO2-based oxide film formed on TNTZ after anodizing. However, the oxide film also contained large amounts of Nb species and the molar Nb/Ti ratio in the TNTZ alloy was high, which makes the surface more hydrophobic than the anodized oxide on Ti. In vivo tests showed that the osteoconductivity of the TNTZ alloy was sensitive to both its surface roughness and hydrophilicity. When the TNTZ alloy was anodized, the process increased either the surface hydrophobicity or the surface roughness at the voltage used in this study. These changes in the surface properties did not improve its osteoconductivity.
