The equa.ions of motion a.e reduced to a.system of ordina.y differentia. equa.ions, which in turn a.e solved numerica.ly using SOR method a.d Simpson’s (1/3) rule. The results a.e ca.cula.ed for different va.ues of the pa.a.eter s (the ra.io of a.gula. velocities of disc a.d fluid) a.d the suction pa.a.eter a. Moreover, three different sets of the va.ues of non-dimensiona. ma.eria. consta.ts rela.ed to micropola. beha.ior of the fluid ha.e been chosen a.bitra.ily. The ca.cula.ions ha.e been ca.ried out using three different grid sizes to check the a.cura.y of the results. The resea.ch concludes tha. the micropola. fluids flow resembles with tha. of Newtonia. fluids when the ma.eria. consta.ts become close to zero. 控制蛋白质在材料表面的吸附和脱附在生物材料或医学等方面十分重要。在不同的情况下,对蛋白质的吸附和脱附有不同的要求。例如,对于细胞和组织在材料表面的增殖以及蛋白质的富集和分离需要蛋白质的吸附,而在生物技术和生物医学材料领域如生物传感器、含血的材料如人工血管、人造心脏等则需要抑制蛋白质的吸附。因此需要设计一种能够根据需求调控蛋白质在其上的吸附和脱附的表面,这一点可以通过同时具有刺激响应性和抗蛋白质吸附性能的表面实现。本论文发展了一种可以实现蛋白质可控吸附的新方法:在材料表面接枝上抗蛋白吸附聚合物刷(聚(2-甲基-2-噁唑啉)(PMOXa.)和刺激响应性聚合物刷(聚(4-乙烯基吡啶)(P4VP))。论文分为两个部分。(1)分别通过2-甲基-2-噁唑啉(MOXa.的阳离子开环聚合(CROP)和4-乙烯基吡啶(4VP)的可逆加成-断裂链转移(Ra.T)聚合,和随后与甲基丙烯酸缩水甘油酯的无规和嵌段共聚制备了聚(2-甲基-2-噁唑啉-r-甲基丙烯酸缩水甘油酯)(PMOXa.r-GMa.刷状共聚物和聚(4-乙烯基吡啶)-b-聚甲基丙烯酸缩水甘油酯)(P4VP-b-PGMa.。最后,将PMOXa.r-GMa.P4VP-b-PGMa.于氯仿得到的混合溶液旋涂到基底表面(硅片和玻璃片),在110℃条件下交联得到PMOXa.P4VP的二元混合刷涂层。通过水接触角(WCa.,X射线光电子能谱(XPS),可变角度椭圆偏正光谱仪(Va.E)和原子力显微镜(a.M)等对涂层进行了表征。研究结果表明,通过简单调整混合溶液组成就可以得到具有特定表面组成的涂层。混合刷的溶胀和塌缩可以随着环境pH 2～10进行调节。最后,通过异硫氰酸荧光素标记的牛血清白蛋白(FITC-BSa.吸附实验研究了 PMOXa.P4VP混合刷对牛血清白蛋白(BSa.的可调控行为。实验表明,通过调节混合刷涂层中PMOXa.r-GMa.P4VP-b-PGMa.组成和环境pH值2-10的变化,可以控制PMOXa.P4VP混合刷由低到高的吸附量和脱附率,在特定条件下89%被吸附的蛋白质可以脱附下来。(2)通过加热制备了 PMOXa.长固定而不同链长的P4VP的基于PMOXa.P4VP的涂层。通过WCa.Va.E研究了PMOXa.P4VP混合刷的调控性能和P4VP链长、pH值以及离子强度(I)之间的关系。结果表明,当P4VP链长是PMOXa.长的三倍时,改变环境条件由pH 3(I=10-3 M)到pH 9(I=10-1 M),水接触角和厚度变化值最大。最后,分别通过FITC-BSa.附实验、石英晶体微天平(QCM-D)和Va.E研究了 P4VP链长对蛋白质吸附/脱附的影响。结果表明,在pH 3及低离子强度(I=10-3 M)的条件下,BSa.附量增多,同时随P4VP链长增加BSa.附也增多。当外界条件变为pH 9且离子强度较强(I=10-1 M)时,吸附的蛋白质可有效脱附,此时PMOXa.P4VP涂层脱附率大于92%。
