Human mesenchymal stromal cells(hMSCs)are mechanically sensitive undergoing phenotypic alterations when subjected to shear stress,cell aggregation,and substrate changes encountered in 3D dynamic bioreactor cultures.However,little is known about how bioreactor microenvironment affects the secretion and cargo profiles of hMSC-derived extracellular vesicles(EVs)including the subset,“exosomes”,which contain therapeutic proteins,nucleic acids,and lipids from the parent cells.In this study,bone marrow-derived hMSCs were expanded on 3D Synthemax II microcarriers in the PBS mini 0.1L Vertical-Wheel bioreactor system under variable shear stress levels at 25,40,and 64 RPM(0.1-0.3 dyn/cm^(2)).The bioreactor system promotes EV secretion from hMSCs by 2.5-fold and upregulates the expression of EV biogenesis markers and glycolysis genes compared to the static 2D culture.The microRNA cargo was also altered in the EVs from bioreactor culture including the upregulation of miR-10,19a,19b,21,132,and 377.EV protein cargo was characterized by proteomics analysis,showing upregulation of metabolic,autophagy and ROS-related proteins comparing with 2D cultured EVs.In addition,the scalability of the Vertical-Wheel bioreactor system was demonstrated in a 0.5L bioreactor,showing similar or better hMSC-EV secretion and cargo content compared to the 0.1L bioreactor.This study advances our understanding of bio-manufacturing of stem cell-derived EVs for applications in cell-free therapy towards treating neurological disorders such as ischemic stroke,Alzheimer’s disease,and multiple sclerosis.

该项目为佐治亚理工学院的一项重要建筑遗产注入了新的活力，将其改建为建筑学院的灵活附属建筑，包括工作室、实验室、教室和研究室。原建筑由一个50英尺高的大跨度工业顶棚，在垂直轴上重新规划，水平轴上保持开放和灵活性。不间断的地板主要作为工作室空间，但也可以重新配置用于大型组件。大跨度空间融入四方院和公共空间城市网络，最大程度的与学院建立联系。对于学校来说，重要的是新干预措施是与希恩曼早期现代主义的复合钢／砖石结构的教学阐述互相补充，而不是与其一争高下。翻修充分利用了该建筑的潜力，钢和胶合板巧妙结合，满足结构、规范、规划和可持续性的要求，获得LEED黄金级认证。

Background:Youzhi artificial intelligence(AI)software is the AI-assisted decision-making system for diagnosing skin tumors.The high diagnostic accuracy of Youzhi AI software was previously validated in specific datasets.The objective of this study was to compare the performance of diagnostic capacity between Youzhi AI software and dermatologists in real-world clinical settings.Methods:A total of 106 patients who underwent skin tumor resection in the Dermatology Department of China-Japan Friendship Hospital from July 2017 to June 2019 and were confirmed as skin tumors by pathological biopsy were selected.Dermoscopy and clinical images of 106 patients were diagnosed by Youzhi AI software and dermatologists at different dermoscopy diagnostic levels.The primary outcome was to compare the diagnostic accuracy of the Youzhi AI software with that of dermatologists and that measured in the laboratory using specific data sets.The secondary results included the sensitivity,specificity,positive predictive value,negative predictive value,F-measure,and Matthews correlation coefficient of Youzhi AI software in the real-world.Results:The diagnostic accuracy of Youzhi AI software in real-world clinical settings was lower than that of the laboratory data(P<0.001).The output result of Youzhi AI software has good stability after several tests.Youzhi AI software diagnosed benign and malignant diseases by recognizing dermoscopic images and diagnosed disease types with higher diagnostic accuracy than by recognizing clinical images(P=0.008,P=0.016,respectively).Compared with dermatologists,Youzhi AI software was more accurate in the diagnosis of skin tumor types through the recognition of dermoscopic images(P=0.01).By evaluating the diagnostic performance of dermatologists under different modes,the diagnostic accuracy of dermatologists in diagnosing disease types by matching dermoscopic and clinical images was significantly higher than that by identifying dermoscopic and clinical images in random sequence(P=0.022).The diagnostic accurac

Systemic lupus erythematosus(SLE)is an autoimmune disease that is a challenge to diagnose and treat.There is an urgent need for biomarkers to help define organ involvement,and more effective therapies.A unique population of T cells,the CD3^(+)CD4^(−)CD8^(−)(DNeg)cells,is significantly increased in lupus patients.Twentyseven cases(53%)of pediatric SLE patients had elevated DNeg cells in their peripheral blood,which correlated with kidney function(R^(2)=0.54).Significant infiltration of DNeg cells was observed in both adult and pediatric lupus kidneys by immunofluorescence.For the first time,this study provides direct evidence that DNeg cells facilitate kidney injury in preclinical 8-week-old MRL/lpr lupus mice.In lupus mice,the increase in DNeg cells tracked with worsening disease and correlated with kidney function(R^(2)=0.85).Our results show that DNeg cells per se can cause kidney dysfunction,increase in number with increase in disease pathology,and could serve as a potential biomarker.
