目的:观察艾灸对慢性心力衰竭(CHF)大鼠心功能及心肌组织凋亡蛋白B淋巴细胞瘤-2(Bcl-2)、Bcl-2相关的X蛋白(Bax)、Fas及其配体FasL表达水平的影响,探讨艾灸防治CHF的可能机制。方法:SD大鼠随机分为正常组、模型组、艾灸组、卡托普利组、艾灸+卡托普利组(简称艾卡组),每组12只。采用隔日腹腔注射盐酸多柔比星的方法复制CHF模型。艾灸组用艾条温和灸双侧"肺俞""心俞"穴;卡托普利组用卡托普利混悬液(5mg/mL,25mL/kg)灌胃;艾卡组先灌服卡托普利混悬液,再行艾灸;均每日1次,连续3周。观察大鼠一般情况及行为体征;计算左心室质量指数(LVmI)和右心室质量指数(RVmI);HE染色观察心肌组织病理变化;酶联免疫吸附法检测血清B型脑钠肽(BNP)与N末端前脑利钠肽前体(nt-pro BNP)含量;蛋白免疫印迹法检测心肌细胞凋亡相关蛋白Bcl-2、Bax、Fas、FasL的表达水平。结果:与正常组比较,模型组大鼠心肌细胞出现横纹不清,细胞肿胀、空泡等情况,其LVmI和RVmI显著增高(Pnt-pro BNP含量明显升高(PmI及RVmI显著降低(Pnt-pro BNP含量下降(P<0.05,P<0.01)。与艾卡组比较,艾灸组与卡托普利组Bax及Fas、FasL表达水平升高(P<0.05,P<0.01),Bcl-2表达水平降低(P<0.05)。结论:艾灸可以改善CHF大鼠心肌损伤,其作用可能与降低Bax、Fas、FasL蛋白表达,上调Bcl-2表达,抑制心肌细胞凋亡有关。

The use of live animal models for testing new therapies for brain and spinal cord repair is a controversial area. Live animal models have associated ethical issues and scientific concerns regarding the predictability of human responses. Alternative models that replicate the 3 D architecture of the central nervous system have prompted the development of organotypic neural injury models. However, the lack of reliable means to access normal human neural tissue has driven reliance on pathological or post-mortem tissue which limits their biological utility. We have established a protocol to use donor cerebellar tonsillar tissue surgically resected from patients with Chiari malformation(cerebellar herniation towards the foramen magnum, with ectopic rather than diseased tissue) to develop an in vitro organotypic model of traumatic brain injury. Viable tissue was maintained for approximately 2 weeks with all the major neural cell types detected. Traumatic injuries could be introduced into the slices with some cardinal features of post-injury pathology evident. Biomaterial placement was also feasible within the in vitro lesions. Accordingly, this ‘proof-of-concept’ study demonstrates that the model offers potential as an alternative to the use of animal tissue for preclinical testing in neural tissue engineering. To our knowledge, this is the first demonstration that donor tissue from patients with Chiari malformation can be used to develop a benchtop model of traumatic brain injury. However, significant challenges in relation to the clinical availability of tissue were encountered, and we discuss logistical issues that must be considered for model scale-up.

Animals are increasingly faced with human?induced stressors that vary in space and time, thus we can expect populati on-level diverge nee in behaviors that help animals to cope with en vironme ntal change. However, empirical evidenee of behavioral trait divergence across environmental extremes is lacking. We tested for variation in behavioral traits among 2 populations of an African cichlid fish (Pseudocrenilabrus multicolor victoriae Seegers, 1990) that experience extremes of dissolved oxygen (DO) and turbidity and are known to vary in a number of physiological and life history traits associated with these stressors. Using a comm on garden reari ng experime nt, F1 progeny from wild-caught parents originating from a swamp (low DO, clear) and a river (high DO, turbid) were reared in high DO, clear water. Predator simulation assays were conducted to test for (1) variation in boldness, general activity, and foraging activity between populations,(2) differences in correlations betwee n behaviors with in and across populati ons, and (3) repeatability of behaviors. There was strong evide nee for diverge nee betwee n populati ons, with swamp fish being more bold (i.e.z leaving refuge sooner after a simulated predator attack) and active (i.e., spent more time out of refuge) than river fish. Across populations there were positive correlations between foraging activity and both boldness and general activity;however, within populations, there was only a strong positive relationship between foraging activity and boldness in the river population. Here, we have demonstrated that populations that originate from drastically different environments can produce proge ny that exhibit measurable differences in behaviors and their correlated relati on ships eve n whe n reared un der comm on conditions.

ABS系统中主缸压力和轮缸压力具有强非线性关系,且传统的控制算法中控制参数不能根据路面的变化及时进行调整,很难在多变的路面工况下取得较好的控制效果。为提高ABS的控制效果,提出一种神经网络模糊PID控制策略。将理想滑移率与实际滑移率之差作为系统输入,经模糊化和归一化后输入神经网络,应用神经网络的自学习和误差反向传播,采用遗传算法优化网络的初始权重,利用BP算法完成网络训练实现模糊规则,使模糊规则的生成转变为加权系数初值的确定和调节,从而表示出模糊规则,寻找一个最佳的PID非线性组合控制率,调整车轮制动力矩,以适应多变的路面。仿真结果表明:该控制策略兼备了神经网络、模糊控制、PID控制的优点,能适应复杂多变的路面。

Aims: To determine correlations of insulin sensitivity to gene expression in omental and subcutaneous adipose tissue of non-obese, non-diabetic pregnant women. methods: microarray gene profiling was performed on subcutaneous and omental adipose tissue from 14 patients and obtained while fasting during non-laboring Cesarean section, using Illumina HumanHT-12 V4 Expression BeadChips. Findings were validated by real-time PCR. matusda-Insulin sensitivity index (IS) and homeostasis model assessment of insulin resistance (HOmA-IR) were calculated from glucose and insulin levels obtained from a frequently sampled oral glucose tolerance test, and correlated with gene expression. Results: Of genes differentially expressed in omental vs. subcutaneous adipose, in omentum 12 genes were expressed toward insulin resistance, whereas only 5 genes were expressed toward insulin sensitivity. In particular, expression of the insulin receptor gene (INSR), which initiates the insulin signaling cascade, is strongly positively correlated with IS and negatively with HOmA-IR in omental tissue (r = 0.84). Conclusion: Differential gene expression in omentum relative to subcutaneous adipose showed a pro-insulin resistance profile in omentum. A clinical importance of omental adipose is observed here, as downregulation of insulin receptor in omentum is correlated with increased systemic insulin resistance.

mannan-binding lectin(mBL)is a soluble innate immune protein that binds to glycosylated *** acts as an opsonin and activates complement,contributing to the destruction and clearance of infecting *** C virus(HCV)encodes two envelope glycoproteins E1 and E2,expressed as non-covalent E1/E2 heterodimers in the viral envelope.E1 and E2 are potential ligands for *** we describe an analysis of the interaction between HCV and mBL using recombinant soluble E2 ectodomain fragment,the full-length E1/E2 heterodimer,expressed in vitro,and assess the effect of this interaction on virus entry.A binding assay using antibody capture of full length E1/E2 heterodimers was used to demonstrate calcium dependent,saturating binding of mBL to HCV *** with various saccharides further confirmed that the interaction was via the lectin domain of *** binds to E1/E2 representing a broad range of virus *** was shown to neutralize the entry into Huh-7 cells of HCV pseudoparticles(HCVpp)bearing E1/E2 from a wide range of *** were neutralized to varying *** was also shown to neutralize an authentic cell culture infectious virus,strain JFH-1(HCVcc).Furthermore,binding of mBL to E1/E2 was able to activate the complement system via mBL-associated serine protease *** conclusion,mBL interacts directly with HCV glycoproteins,which are present on the surface of the virion,resulting in neutralization of HCV particles.

The free convective heat transfer to the power-law non-Newtonian flow from a vertical plate in a porous medium saturated with nanofluid under laminar conditions is investigated. It is considered that the non-Newtonian nanofluid obeys the mathematical model of power-law. The model used for the nanofluid incorporates the effects of Brown- ian motion and thermophoresis. The partial differential system governing the problem is transformed into an ordinary system via a usual similarity transformation. The numer- ical solutions of the resulting ordinary system are obtained. These solutions depend on the power-law index n, Lewis number Le, buoyancy-ratio number Nr, Brownian motion number Nb, and thermophoresis number nt. For various values of n and Le, the effects of the influence parameters on the fluid behavior as well as the reduced Nusselt number are presented and discussed.
