Association between genome-wide epigenetic and genetic alterations in breast cancer tissue and response to HER2-targeted therapies in HER2- positive breast cancer patients: new findings and a systematic review

Recent evidence suggests that genetic and epigenetic mechanisms might be associated with acquired resistance to cancer *** aim of this study was to assess the association of genome-wide genetic and epigenetic alterations with the response to anti-HER2 agents in HER2-positive breast cancer *** was screened for articles published until March 2021 on observational studies investigating the association of genome-wide genetic and epigenetic alterations,measured in breast cancer tissues or blood,with the response to targeted treatment in HER2-positive breast cancer *** studies were included in the review along with ours,in which we compared the genome-wide DNA methylation pattern in breast tumor tissues of patients who acquired resistance to treatment (case group, n = 6) to that of patients who did not develop resistance (control group, n =6). Among genes identified as differentially methylated between the breast cancer tissue of cases and controls, oneof them, PRKACA, was also reported as differentially expressed in two studies included in the review. Althoughincluded studies were heterogeneous in terms of methodology and study population, our review suggests thatgenes of the PI3K pathway may play an important role in developing resistance to anti-HER2 agents in breastcancer patients. Genome-wide genetic and epigenetic alterations measured in breast cancer tissue or blood mightbe promising markers of resistance to anti-HER2 agents in HER2-positive breast cancer patients. Further studiesare needed to confirm these data.

Understanding of resource requirements of vulnerable species is key for conservation ***,we used sta-ble carbon(δ13C)and nitrogen isotopes(δ15N)of hair from giant armadillos(Priodontes maximus)to understand individual resource *** collected hair from 19 giant armadillos analyzing 34 samples from the Brazilian Pan-tanal and compared the resource use between sexes and age *** also assessed the relationship of isotopic values with individuals’body mass and the habitat type *** observed a wide variation in resource use by giant armadillos,showing that individuals consume prey that feeds on distinct resources and occupies different habitat types,indicating that there is individual variation in foraging *** giant armadillos presented a mixed diet(C3/C4 resources,forests/open areas),but a quarter of individuals had strictly C4 diets(open areas).Males are more prone to forage on C4 resources,presenting an isotopic niche 2.6-times larger than that of *** presented diets more associated with habitats with high arboreal cover(C3 resources,forests),while adults foraged more in open areas(C4 resources).This result is mirrored by the positive relationship betweenδ13C values and body mass,suggesting that larger giant armadillos tend to feed more in open *** observed thatδ13C values decreased as individuals increased the use of habitats with high arboreal *** stress the importance of con-serving the natural mosaic in the Pantanal landscape for the species persistence,given that giant armadillos require all habitat types to fulﬁll their dietary,spatial,and developmental needs.

Do PI3-kinase mutations drive T cells insane？

The link between the differentia- tion of effector T lymphocytes and metabolism has become an important area of research. The role played by the phosphoinositide 3-kinase （PI3K）/Akt/ mTOR （mammalian target ofrapamycin） pathway in this process was recently emphasized by Lucas et al.1 who reported a series of primary immunodeficiency patients with three different germ-line, gain-of-function mutations of the p1108 subunit of PI3K. The patients consistently presented a deficiency in naive T lymphocytes and an accumulation of highly differentiated effector T cells. This phenotype was associated with recurrent respiratory infections and with the reactivation of persistent viruses. The disease was named PASLI （p1108-activating mutation causing senescent T cells, lymphadenopathy and immunodeficiency）. The same disorder was simultaneously reported by Angulo et al.z and was named the activated PI3K-8 syndrome （APDS）. Although the accumulation of effector T cells in the state of immunodeficiency was somewhat unexpected, the description of this syn- drome provides important insight into the regulation of T-cell differentiation and T-cell memory.

Simultaneous nephrectomy during kidney transplantation for polycystic kidney disease does not detrimentally impact comorbidity and graft survival

BACKGROUND The lack of space,as an indication for a native unilateral nephrectomy for positioning a future kidney graft in the absence of other autosomal dominant polycystic kidney disease-related symptoms,remains *** To evaluate the surgical comorbidity and the impact on graft survival of an associated ipsilateral native nephrectomy during isolated kidney transplantation in patients with autosomal dominant polycystic kidney *** One hundred and fifty-four kidney transplantations performed between January 2007 and January 2019 of which 77 without(kidney transplant alone(KTA)group)and 77 with associated ipsilateral nephrectomy(KTIN group),were retrospectively *** and surgical variables were analyzed and their respective impact on surgical comorbidity and graft *** Creation of space for future graft positioning was the main reason(n=74,96.1%)for associated ipsilateral *** significant difference in surgical comorbidity(lymphocele,wound infection,incisional hernia,wound hematoma,urinary infection,need for blood transfusion,hospitalization stay,Dindo Clavien classification and readmission rate)was observed between the two study *** incidence of primary nonfunction and delayed graft function was comparable in both groups[0%and 2.6%(P=0.497)and 9.1%and 16.9%(P=0.230),respectively,in the KTA and KTIN group].The 1-and 5-year graft survival were 94.8%and 90.3%,and 100%and 93.8%,respectively,in the KTA and KTIN group(P=0.774).The 1-and 5-year patient survival were 96.1%and 92.9%,and 100%and 100%,respectively,in the KTA and KTIN group(P=0.168).CONCLUSION Simultaneous ipsilateral native nephrectomy to create space for graft positioning during kidney transplantation in patients with autosomal dominant polycystic kidney disease does not negatively impact surgical comorbidity and short-and long-term graft survival.

Population viability analysis: using a modeling tool to assess the viability of tapir populations in fragmented landscapes

A population viability analysis(PVA)was conducted of the lowland tapir populations in the Atlantic Forest of the Pontal do Paranapanema region,Brazil,including Morro do Diabo State Park(MDSP)and surrounding forest *** from the model projected that the population of 126 tapirs in MDSP is likely to per-sist over the next 100 years;however,200 tapirs would be required to maintain a viable ***-ity analysis showed that sub-adult mortality and adult mortality have the strongest influence on the dynamics of lowland tapir *** road-kill has a major impact on the MDSP tapir population and can lead to population *** modeling showed that dispersal of tapirs from MDSP to the surrounding fragments can be detrimental to the overall metapopulation,as fragments act as ***,the model showed that under certain conditions the maintenance of the metapopulation dynamics might be determinant for the persistence of tapirs in the region,particularly in the smaller *** establishment of corridors con-necting MDSP to the forest fragments models resulted in an increase in the stochastic growth rate,making ta-pirs more resilient to threats and catastrophes,but only if rates of mortality were not increased when using *** PVA showed that the conservation of tapirs in the Pontal region depends on:the effective protection of MDSP;maintenance and,whenever possible,enhancement of the functional connectivity of the landscape,reducing mortality during dispersal and threats in the unprotected forest fragments;and neutralization of all threats affecting tapirs in the smaller forest fragments.

Dedicated population genomics for the silent world： the specific questions of marine population genetics

The life cycle of most marine species is characterized by high fecundity, dispersal during a planktonic larval phase and a sessile or sedentary adult phase with large census population size (Figure 1). Many articles on marine population genetics start by recalling these main characteristics and ensuing expectations,namely: high genetic diversity within population, low genetic differentiation between populations, high influx of effectively selected advantageous and deleterious mutations, and a high migration load opposing local adaptation. Most of the time,this reminder serves to better emphasize the paradox that observations deviate from these expectations, the most popular being genetic differentiation despite larval dispersal, challenging the idea that marine populations are open and well connected, just followed by pervasive local adaptation, challenging the idea that high migration rates prevent the establishment of locally adapted alleles in the sea, and finally low effective population size (Ne) despite large census size (N). The list of such counterexamples became so long that what was initially thought paradoxical finally became a shift to a new paradigm (Hauser and Carvalho 2008): (1) larval dispersal would be less effective than expected to homogenize allele frequencies between populations with high rates of self-recruitment, as, for example, in coral reef fishes (Palumbi and Warner 2003; Jones et al. 2005), (2) selection would be rampant in genomes and easy to identify with genome scans, as,for example, adaptation to brackish waters of the Baltic Sea that would target nothing less than ?500 loci homogeneously distributed in the herring genome (Lamichhaney et al. 2012; Martinez Barrio et al. 2016), and (3) genetic drift would be stronger than thought owing to a very low proportion of individuals effectively contributing to the following generation, as,for example, in exploited fish and shellfish stocks (Hauser et al. 2002; Ovenden et al. 2007; Pinsky and P

Laser Measurements in High Speed Compressors for Rotor-Stator Interaction Analysis

This paper deals with the experimental quantification of the unsteady effects of the interactions between rotor and stator rows in high speed compressors. Due to the fact that the levels of the periodic fluctuations arising from the unsteady interaction may be low compared with the random fluctuations arising from the measurement uncertainties, it is crucial to minimize the errors inherent to the used technique. The first part of the paper concentrates on technical details relative to the experimental process. The second part is devoted to the data post-processing. Two tools for analysing the rotor-stator interactions are presented. The first tool is based on a decomposition of the flow field which was initially introduced to solve numerical problems when attempting to calculate the flow field in a multi-row configuration. The second tool is based on a spectral analysis of the signal, that qualifies the interaction in a sense of circumferential spinning lobes. Experimental results obtained within both an axial and a centrifugal high speed compressors are used to illustrate the data processing. In both cases, the effects of the unsteady interaction are quantified.

BACKGROUND Probiotics are a promising solution for managing irritable bowel syndrome(IBS).Saccharomyces cerevisiae(***)I-3856 has already demonstrated beneficial effects in IBS subjects,particularly in IBS with predominant constipation(IBS-C).AIM To confirm the efficacy of *** I-3856 in the management of gastrointestinal symptoms in *** A randomized,double-blind,placebo-controlled clinical study was performed in a total of 456 *** a run-in period,subjects were randomly assigned to the group receiving *** I-3856(8×109 CFU daily)or the placebo for 8 wk,and they performed daily self-evaluations of gastrointestinal *** primary objective was to assess the effect of the probiotic on abdominal *** secondary objectives were the evaluation of other gastrointestinal symptoms,bowel movement frequency and consistency,and quality of life(QOL).RESULTS A significantly higher proportion of abdominal pain responders was reported in the Probiotic group(45.1%vs 33.9%,P=0.017).A nonsignificant difference in the area under the curve for abdominal pain over the second month of supplementation was observed in subjects receiving probiotic vs placebo[P=0.073,95%CI:-0.59(-1.23;0.05)].No statistically significant differences were reported in the evolution of bowel movement frequency and stool consistency between the *** 8 wk of supplementation,the overall QOL score was significantly higher in the Probiotic group than in the Placebo group[P=0.047,95%CI:3.86(0.52;7.20)].Furthermore,exploratory analyses showed statistically significant and clinically relevant improvements in QOL scores in abdominal pain responders vs *** The results of this clinical study confirmed the abdominal pain alleviation properties of *** I-3856 in *** pain relief was associated with improved *** identifier:NCT03150212.

Northern Fulmars(Fulmarus glacialis)are a pelagic seabird species distributed at northern and polar *** are often used as an indicator of plastic pollution in the North Sea region,but data are lacking from higher latitudes,especially when it comes to ***,we investigated amounts of ingested plastic and their characteristics in fulmar chicks from the Faroe *** particles(≥1 mm)in chicks of two age classes were searched using a digestion method with *** addition,to evaluate if additive tissue burden reflects plastic ingestion,we measured liver tissue concentrations of two pollutant classes associated with plastic materials:polybrominated diphenyl ethers(PBDEs)and several dechloranes,using gas chromatography with high-resolution mass *** most common shape was hard fragment(81%)and the most common polymer was polyethylene(73%).Plastic contamination did not differ between either age class,and we found no correlation between neither the amount and mass of plastic particles and the concentration of *** comparison with previous studies on adult fulmars,we do not recommend using chicks for biomonitoring adults because chicks seem to ingest more plastics than adults.
