Triple immunofluorescence staining has recently been developed to simultaneously identify all muscle fibers on a single cryosection which is helpful for clinical and basic research, but it has disadvantages such as fast photobleaching and unclear outlines of muscle fibers. Triple immunoenzyme staining（TIE） is likely to avoid these disadvantages. In this study, we aimed to establish a sensitive and specific TIE technique to identify fiber types in normal, denervated, and reinnervated rat muscles, and to develop a systematic sampling method for muscle fiber quantification. Tibialis anterior and soleus from normal, denervated, and reinnervated Lewis rat hind limbs were used. Five consecutive cryosections were cut from each muscle, including one for TIE and four for single immunoenzyme staining（SIE）. The TIE was performed using the polymerized reporter enzyme staining system for the first two antigens（A4.74 for My HC-IIA, BA-F8 for My HC-I） and alkaline phosphatase staining system for the third antigen（BF-F3 for My HC-IIB）, followed by corresponding detective systems and respective chromogens. The type of muscle fibers was quantified by systematic sampling at 12.5%, 25%, 33% and 50% of all muscle fibers, and was compared with that acquired from counting all the fibers（100%）. All muscle fiber phenotypes, including pure and hybrid, could be simultaneously identified on a single TIE cryosection with clear outlines. The fiber types on TIE slides matched well with their respective counterpart on the consecutive SIE slides with a 95% match rate. Systematic sampling of 12.5% fibers could represent the true fiber type distribution of the entire muscle section. Our results suggest that novel TIE can effectively visualize fiber types in normal, denervated or reinnervated rat muscles.

AIM： To report the indications for and techniques of corneal transplantation at Vietnam National Institute of Ophthalmology （VNIO） over a period of 12y （2002-2013）, METHODS： Records of patients who had undergone corneal transplantation at VNIO from j.nuary 1, 2002 to j.nuary 1, 2014 were reviewed to determine the indication for and type of corneal transplant performed. Patient age, gender, indication for corneal transplantation and surgical technique were recorded and analyzed. RESULTS： Corneal transplantation were underwent in 1390 eyes of 1278 patients with a mean age of 44.9±18.1y during the period under review. The most common indication was infectious corneal ulcer （n=670; 48.2%）, followed by corneal scar （n=333, 24.0% ）, corneal dystrophy （n=138, 9.9%） and failed graft （n=112, 8.1%）. Nearly all procedures performed were penetrating keratoplasty （n=1300, 93.5% ）, with a few lamellar keratoplasty procedures performed： lamellar keratoplasty （n=52, 3.7%）, Descemet＇s stripping automated endothelial keratoplasty （n =27, 1.9% ） and deep anterior lamellar keratoplasty （n =11, 0.8%）.CONCLUSION： While the most common indication for keratoplasty was infectious keratitis, nearly all indications for corneal transplantation were managed with penetrating keratoplasty, However, lamellar keratoplasty techniques, including deep anterior lamellar keratoplasty and Descemet＇s stripping automated endothelial keratoplasty, are being performed with increasing frequency for isolated stromal and endothelial disorders, respectively,

The middle-upper Cretaceous Ceduna River system traversed continental Australia from the NE coast to the centre of the southern coast. At its mouth, it formed a vast delta system that is similar in scale to the Niger delta of West Africa. The delta system is composed of two main lobes that represent different phases of delta construction. A recent hypothesis has challenged the traditional idea that both lobes of the delta were derived from a transcontinental river system by suggesting that the upper lobe （Santonian -Maastrichtian） is instead derived from a restricted catchment within southern Australia. Hf isotopic data presented here fingerprint the original source of the upper delta lobe zircons to NE Australia, with data comparing well with similar U-Pb and Lu-Hf isotopic data from the Lachlan Orogen, the New England Orogen, the eastern Musgraves Province and the northern Flinders Ranges. These data do not preclude a model where the lobe is derived from recycled Eromanga Basin sediments during a phase of late Cretaceous inland Australian uplift, but when coupled with reconnaissance low-temperature thermochronometry from the region of the Ceduna River course indicating widespread Triassic-j.rassic exhumation, and comparisons with detrital zircon data from the Winton Formation upstream of any proposed uplift, we suggest that both lobes of the Ceduna Delta are likely to be derived from a transcontinental Ceduna River.

Background: Preventing anterior cruciate ligament(ACL) inj.ries is important to avoid long-term adverse health consequences. Identifying barriers to implementation of these prevention programs is crucial to reducing the incidence of these inj.ries. Our purpose was to identify barriers of implementation for ACL inj.ry prevention programs and suggest mechanisms for reducing the barriers through application of a SocioEcological Model(SEM).Methods: Studies investigating ACL prevention program effectiveness were searched in Medline via PubMed and the Cochrane Library, and a subsequent review of the references of the identified articles, yielded 15 articles total. Inclusion criteria encompassed prospective controlled trials, published in English, with ACL inj.ries as the primary outcome. Studies were independently appraised by 2 reviewers for methodological quality using the PEDro scale. Barriers to implementation were identified when reported in at least 2 separate studies. A SEM was used to suggest ways to reduce the identified ***: Five barriers were identified: motivation, time requirements, skill requirements for program facilitators, compliance, and cost. The SEM suggested ways to minimize the barriers at all levels of the model from the individual through policy ***: Identification of barriers to program implementation and suggesting how to reduce them through the SEM is a critical first step toward enabling ACL prevention programs to be more effective and ultimately reducing the incidence of these inj.ries.

Optimising validity and translational potential in rodent models of brain disorders

Neurological and psychiatric disorders collectively constitute the greatest burden of disease. However, the human brain is the most complex of biological systems and therefore accurately modeling brain disorders presents enormous challenges. A large range of therapeutic approaches across a diverse collection of brain disorders have been found to show great promise in preclinical testing and then failed during clinical trials. There are a variety of potential reasons for such failures, on both the preclinical and clinical sides of the equation. In this article, I wi l l focus on the key issues of validity in animal models. I wi l l discuss two forms of construct validity,‘genetic construct validity’ and ‘environmental construct val idity’,which model specific aspects of the genome and ‘envirome’ relevant to the disorder in question. The generation of new gene-edited animal models has been facilitated by new technologies, the most notable of which are CRISPR-Cas systems. These and other technologies can be used to enhance contruct validity. Finally, I wi l l discuss how face validity can be optimized, via more sophisticated cognitive, affective and motor behavioural tests, translational tools and the integration of molecular, cellular and systems data. Predictive validity cannot yet be assessed for the many preclinical models where we currently lack effective clinical interventions, however this wi l l change as the translational pipeline is honed to deliver therapies for a range of devastating disorders.

We present new U-Pb and Hf zircon isotope data on a suite of granitoids from a hitherto unstudied area of the Palaeoproterozoic Lupa terrane of the Ubendian belt in SW *** maj.r part of the area comprises a sequence of upper amphibolite grade paragneisses and ***,the field relations show a complex history of granitoid emplacement from early pre-tectonic strongly foliated diorite-tonalite-granodiorite orthogneiss to late tectonic,weakly deformed ellipsoidal granitoid *** were followed by emplacement of undeformed K-feldspar rich porphyritic alkali granite.U-Pb zircon data on the granitoids show their emplacement ages span the period^1925 Ma to 1890 Ma and constrain the culmination of the Ubendian orogeny in the Lupa terrane to about 1900 *** undeformed K-feldspar rich Chimala granite,previously considered to be unconformably overlain by the Buanj. Group volcano-sedimentary rocks(recently shown to be^1675 Ma,not Neoproterozoic as originally thought)was dated at^1407 Ma showing that it must intrude the sedimentary rocks,although the age relationships are not exposed in the mapping *** is the first record of early Irumide/Kibaran-aged post-tectonic granites in this part of East *** isotope data from the zircons show that the granitoids have a long crustal pre-history stretching back to as far as^3.9 Ga,and showing that the Lupa terrane may form part of the"metacratonic"margin of an originally much more extensive Tanzania Craton.
