Is serum copeptin a modifiable biomarker in autosomal dominant polycystic kidney disease?

The availability of disease-modifying drugs for the management of autosomal dominant polycystic kidney disease(ADPKD) has accelerated the need to accurately predict renal prognosis and/or treatment response in this condition. Arginine vasopressin (AVP) is a critical determinant of postnatal kidney cyst growth in ADPKD. copeptin (the c-terminal glycoprotein of the precursor AVP peptide) is an accurate surrogate marker of AVP release that is stable and easily measured by immunoassay. cohort studies show that serum copeptin is correlated with disease severity in ADPKD, and predicts future renal events [decline in renal function and increase in total kidney volume (TKV)]. However, serum copeptin is strongly correlated with creatinine, and its additional value as a prognostic biomarker over estimated glomerular filtration rate and TKV is not certain. It has also been suggested that copeptin could be a predictive biomarker to select ADPKD patients who are most likely to benefit from AVP-modifying therapies, but prospective data to validate this assumption are required. In this regard, long-term randomised clinical trials evaluating the effect of prescribed water intake on renal cyst growth may contribute to addressing this hypothesis. In conclusion, although serum copeptin is aligned with the basic pathogenesis of ADPKD, further rigorous studies are needed to define if it will contribute to enabling the delivery of personalised care in ADPKD.

The Role of copper and Vitamin A Deficiencies Leading to Neurological Signs in captive cheetahs(Acinonyx jubatus)and Lions(Panthera leo)in the United Arab Emirates

Neurological signs like ataxia and hind limb paresis have often been reported in cheetahs (Acinonyx jubatus), lions (Panthera leo) and snow leopards (Panthera unica). As a cause, copper and Vitamin A deficiencies have been discussed. Many cases were seen in cheetahs and lions in the United Arab Emirates (UAE) within the last years. The aim of this study was to find correlations between nutrition, serum, and tissue levels, focusing on copper and Vitamin A. Blood and tissue samples of affected and unaffected animals were analyzed at the central Veterinary Research Laboratory in Dubai, UAE. Animals were split into three different groups (A, B and c) according to their diets. Minerals were determined in serum, tissue, food and water samples, and serum was additionally analyzed for Vitamin A and E. Liver, kidney and spinal cord samples were taken for histopathological investigations. Mean serum copper and liver copper levels of animals fed pure chicken muscle meat without supplements were significantly lower (0.41 ± 0.71 μM/L;2.16 ± 0.95 ppm wet weight) than in animals fed a whole carcass prey diet (12.16 ± 3.42 μM/L;16.01 ± 17.51 ppm wet weight) (p carcass prey diets (1.85 ± 0.68;27.31 ± 5.69 μM/L). Liver zinc concentrations were highest in animals fed pure chicken meat only (43.75 ± 16.48 ppm wet weight). In histopathology, demyelination of the spinal cord was found in all of the affected animals and most commonly when fed a diet based on poultry without supplements.

Valuable Biomolecules from Nine North Atlantic Red Macroalgae: Amino Acids, Fatty Acids, carotenoids, Minerals and Metals

In modern society, novel marine resources are scrutinized pursuing compounds of use in the medical, pharmaceutical, biotech, food or feed industry. Few of the numerous marine macroalgae are currently exploited. In this study, the contents of nutritional compounds from nine common North Atlantic red macroalgae were compared: the lipid content was low and constant among the species, whereas the fatty acid profiles indicated that these species constitute interesting sources of polyunsaturated fatty acids (PUFA). The dominating essential and non-essential amino acids were lysine and leucine, aspartic acid, glutamic acid, and arginine, respectively. The amino acid score of the nine algae varied from 44% to 92%, the most commonly first limiting amino acid being histidine. Lutein, β-carotene, and zeaxanthin were the identified carotenoids. contents of all macro and trace minerals, with the exception of phosphorus, were higher than those described for conventional food. Low sodium/potassium ratios (0.08 - 2.54) suggested a potential for using the ash fraction for sodium salt replacement. The algae constituted rich sources of carbohydrates (40% to 71% of DM) which show their potential for a broader commercial exploitation. In some species, the concentrations of arsenic, cadmium, and lead exceeded limit values for application in food or feed. In conclusion, the nine algae represent promising potential sources of health promoting additives for human and animal diets, in whole or in a biorefinery concept.

The variations in the capacity of fish species to biosynthesize the n-3 highly polyunsaturated fatty acids （HUFA）, eicosapentaenoic （EPA） and docosahexaenoic （DHA） acids which are crucial to the health of higher vertebrates has been well documented. HUFA biosynthesis has been reported to involve enzyme-mediated fatty acyl desaturation and elongation processes. In previous studies, various authors have cloned, characterized and identified different genes for fatty acyl desaturase and elongase （FAD and FAE, respectively） enzymes from fish species such as Atlantic salmon and zebra fish, and have also shown the up and down regulation of the activity of these enzymes in response to fluctuations in dietary HUFA. In this paper, we report on the tissue distribution in the expression of genes （mRNA） for FAD and FAE enzymes in channa striata （Bloch, 1793） fingerlings, to evaluate the tissues of the fish which show the highest activity of these enzymes. For this purpose, mRNA from 8 different tissues of the fish （maintained on a commercial diet） were isolated and analyzed, using conventional PcR technique. The tissue distribution of the genes was estimated to be significantly （P＜0.05） higher for both FAD and FAE enzymes in the liver and brain of c. striata than in the other 6 tissues measured （muscle, ovary, testis, gill and skin） respectively. The implication of this observation with respect to the use of vegetable oils to substitute fish oil in diets for freshwater fish species is discussed accordingly.

Insights into chitosan hydrogels on dentine bond strength and cytotoxicity

contemporary dental adhesives show favorable immediate results in terms of bonding effectiveness. However, the durability of resin-dentin bonds is their major problem. Materials and Methods: Preparation of 3 chitosan-antioxidant hydrogels was achieved using modified hydrogel preparation method. Their effect on the bond strength to dentine both short term (after 24 hours) and long term (after 6 months) were evaluated using shear bond strength measurements using Instron Universal Testing Mascine). The SEM was used to study the surface of the hydrogels. The cell survival rate (cytotoxicity) of the antioxidants resveratrol, β-carotene and propolis towards Balb/c 3T3 mouse fibroblast cells was also assessed using the standard MTT assay. Results: It was found that chitosan-H treated dentine gives significantly (p β-carotene (92%) > propolis (68%) > resveratrol (33%). conclusion: the antioxidant-chitosan hydrogels significantly improved bonding to dentine with or without phosphoric acid treatment. The pH of the growth medium had a high influence on the cell survival rate of Balb/c mouse 3T3 fibroblast cells. The release of the antioxidant β-carotene would not have an influence on the pulp cells. These materials might address the current perspectives for improving bond durability.
