Changes in fetal mannose and other carbohydrates induced by a maternal insulin infusion in pregnant sheep

Background： The importance of non-glucose carbohydrates, especially mannose and inositol, for normal development is increasingly recognized. Whether pregnancies complicated by abnormal glucose transfer to the fetus also affect the regulation of non-glucose carbohydrates is unknown. In pregnant sheep, maternal insulin infusions were used to reduce glucose supply to the fetus for both short （2-wk） and long （8-wk） durations to test the hypothesis that a maternal insulin infusion would suppress fetal mannose and inositol concentrations. We also used direct fetal insulin infusions （1-wk hyperinsulinemic-isoglycemic clamp） to determine the relative importance of fetal glucose and insulin for regulating non-glucose carbohydrates. Results： A maternal insulin infusion resulted in lower maternal （50%, P 〈 0.01） and fetal （35-45%, P 〈 0.01） mannose concentrations, which were highly correlated （r^2 = 0.69, P 〈 0.01）. A fetal insulin infusion resulted in a 50% reduction of fetal mannose （P 〈 0.05）. Neither maternal nor fetal plasma inositol changed with exogenous insulin infusions. Additionally, maternal insulin infusion resulted in lower fetal sorbitol and fructose （P 〈 0.01）. Conclusions： Chronically decreased glucose supply to the fetus as well as fetal hyperinsulinemia both reduce fetal non-glucose carbohydrates. Given the role of these carbohydrates in protein glycosylation and lipid production, more research on their metabolism in pregnancies complicated by abnormal glucose metabolism is clearly warranted.

To balance the manufacturing cost and customizability of automotive parts,a hybrid manufacturing process combining die-casting and selective laser melting(SLM)is proposed:starting with a conventional cast substrate,SLM is utilized to add additional geometric elements on top of *** this hybrid process,the first priority is to prepare a substrate surface suitable for the subsequent SLM addition of the top-on *** this study,the original cast surface of AlSi7Mg was processed by sandblasting,wire electro-discharge machining,and laser remelting,***,additional AlSi7Mg components were built on both the original cast and treated surfaces through *** hybrid builds,these surfaces and resultant interfaces were examined by optical and scanning electron *** indicate that the defect-free metallurgical joint between the cast and additively added parts can be formed on all surfaces except for the one processed by electro-discharge *** observed epitaxial grain growth crossing the interface implies a strong connection between the cast and the SLMed *** these benefits,also mismatches in microstructure,residual stress level and element distribution between the two parts are *** a comprehensive assessment,laser remelting with no additional machining is recommended as the optimal surface treatment preceding SLM fabrication,because of its user-friendly operation,low cost,and high industrial feasibility.

Reduction of low-density lipoprotein-cholesterol through statin therapy has only modestly decreased coronary heart disease (CHD)-associated mortality in developed countries, which has prompted the search for alternative therapeutic strategies for CHD. Major efforts are now focused on therapies that augment high-density lipoprotein (HDL)-mediated reverse cholesterol transport (RCT), and ultimately increase the fecal disposal of cholesterol. The process of RCT has long been thought to simply involve HDL-mediated delivery of peripheral cholesterol to the liver for biliary excretion out of the body. However, recent studies have revealed a novel pathway for RCT that does not rely on biliary secretion. This nonbiliary pathway rather involves the direct excretion of cholesterol by the proximal small intestine. Compared to RCT therapies that augment biliary sterol loss, modulation of non-biliary fecal sterol loss through the intestine is a much more attractive therapeutic strategy, given that excessive biliary cholesterol secretion can promote gallstone formation. However, we are at an early stage in understanding the molecular mechanisms regulating the non-biliary pathway for RCT, and much additional work is required in order to effectively target this pathway for CHD prevention. The purpose of this review is to discuss our current understanding of biliary and nonbiliary contributions to RCT with particular emphasis on the possibility of targeting the intestine as an inducible cholesterol secretory organ.

Loffeld’s bistatic formula (LBF) is the first two-dimensional analytic point target reference spectrum derived for general bistatic SAR frequency domain *** phase history is expanded in Taylor series around the individual points of stationary phase of the trans- mitter-target and target-receiver phase histories,respectively,and thus the common bistatic stationary phase point can be obtained using the method of stationary ***,it shows limitations for extreme bistatic configurations,namely the highly squinted mode and space-surface *** weighted LBF (WLBF) is proposed in this paper based on the different contributions of total phase modulation from the transmitter and *** formulae we derived are compared with that of the original *** extreme bistatic stripmap SAR data can be focused using WLBF,which accommodates the spaceborne squint geometry using the modified effec- tive velocity solution.A point target simulation example is presented to verify the accuracy of the new WLBF spectrum.

The Hoekebrown criterion was introduced in 1980 to provide input for the design of underground excavations in *** criterion now incorporates both intact rock and discontinuities,such as joints,characterized by the geological strength index(GSI),into a system designed to estimate the mechanical behaviour of typical rock masses encountered in tunnels,slopes and *** strength and deformation properties of intact rock,derived from laboratory tests,are reduced based on the properties of discontinuities in the rock *** nonlinear Hoekebrown criterion for rock masses is widely accepted and has been applied in many projects around the ***,in general,it has been found to provide satisfactory estimates,there are several questions on the limits of its applicability and on the inaccuracies related to the quality of the input *** paper introduces relatively few fundamental changes,but it does discuss many of the issues of utilization and presents case histories to demonstrate practical applications of the criterion and the GSI system.

A Bayesian approach is proposed for the inference of the geotechnical parameters used in slope *** methodology involves the construction of posterior probability distributions that combine prior information on the parameter values with typical data from laboratory tests and site investigations used in *** posterior distributions are often complex,multidimensional functions whose analysis requires the use of Markov chain Monte Carlo(MCMC)*** procedures are used to draw representative samples of the parameters investigated,providing information on their best estimate values,variability and *** paper describes the methodology to define the posterior distributions of the input parameters for slope design and the use of these results for evaluation of the reliability of a slope with the first order reliability method(FORM).The reliability analysis corresponds to a forward stability analysis of the slope where the factor of safety(FS)is calculated with a surrogate model from the more likely values of the input *** Bayesian model is also used to update the estimation of the input parameters based on the back analysis of slope *** this case,the condition FS?1 is treated as a data point that is compared with the model prediction of *** analysis requires a sufficient number of observations of failure to outbalance the effect of the initial input *** parameters are updated according to their uncertainty,which is determined by the amount of data supporting *** methodology is illustrated with an example of a rock slope characterised with a Hoek-brown rock mass *** example is used to highlight the advantages of using Bayesian methods for the slope reliability analysis and to show the effects of data support on the results of the updating process from back analysis of failure.
