Molecular and cellular pathophysiology of circulating cardiomyocyte-specific cell free DNA (cfDNA): Biomarkers of heart failure and potential therapeutic targets

Pathological cardiac damage during heart failure is associated with cell death and damage associated molecular patterns (DAMPs) release which triggers a viscous cycle of sterile inflammation to mediate maladaptive cardiac tissue remodelling during the progression to heart failure. DAMPs like cytokines, chemokines, and nuclear or mitochondrial genomic fragments are released in the pathological myocardium. Interestingly, circulating or cytosolic DNA fragments can play a role in the disease by interaction with nucleic acid sensors expressed in cardiomyocyte and non-myocyte neighbouring cells. The circulating cell free DNA (cfDNA) fragments have been clinically reported as markers for various diseases including cardiovascular pathophysiology. Such cfDNA within the DAMP pool can mediate intra- and inter-cellular signalling cascade to upregulate transcriptional expression of inflammatory mediators and trigger oxidative stress within cells. The cellular role of such genomic equivalents varying with chronic or acute stress might be correlated with the cell death forms encountered in myocardium during disease progression. Thus, cfDNA can be phenotypically correlated as a critical player towards upregulation of pathological processes like interstitial fibrosis, cardiomyocyte contractile dysfunction and cell death. Herein, we review the association of cfDNA with heart failure and analyse their potential usage as novel and effective therapeutic targets towards augmentation of cardiac function.

Melamine-Formaldehyde Acrylamide and Gum Polymer Impregnated Wood Polymer Nanocomposite

Melamine-formaldehyde acrylamide （MFA） copolymer was prepared and vacuum impregnated into wood in presence of 1,3-dimethylol 4,5-dihydroxyethylene urea （DMDHEU） as crosslinker, vinyltrichlorosilane （VTCS） modified montmorillonite （MMT） and Gum Polymer （GP） derived from Moringa oleifera as a flame retarding agent under catalyst heat treatment. The formation of MFA and DMDHEU was confirmed by Nuclear Magnetic Resonance （NMR） and Fourier Transform Infrared Spectroscopy （FTIR） studies. The crystallinity of the composites and interaction among them was studied by X-ray Diffrac- tometry （XRD） and FTIR study. Maximum interaction was found in wood samples treated with MFA/DMDHEU/GP （3 phr）/MMT as shown by FTIR. Scanning Electron Microscopy （SEM） revealed the presence of polymer and MMT in the composites. Transmission Electron Microscopy （TEM） study indicated the incorporation of MMT into the wood polymer composite. Thermal stability and flammability were checked by Thermogravimetric Analyser （TGA） and Limiting Oxygen Index （LOI） instrument. The treatment of wood with GP had a significant influence on the thermal stability and flame retardant properties of the composites. Remarkable improvement in water repellency and chemical resistance was found for the treated wood samples. The composites showed significant improvement in mechanical properties due to treatment.

The journey of gut microbiome - An introduction and its influence on metabolic disorders

BACKGROUND： Metabolic disorders such as Obesity, Diabetes Type 2 （T2DM） and Inflammatory Bowel Diseases （IBD） are the most prevalent globally. Recently, there has been a surge in the evidence indicating the correlation between the intestinal microbiota and development of these metabolic conditions apart from predisposing genetic and epigenetic factors. Gut microbiome is pivotal in controlling the host metabolism and physiology. But imbalances in the microbiota patterns lead to these disorders via several pathways. Animal and human studies so far have concentrated mostly on metagenomics for the whole microbiome characterization to understand how microbiome supports health in general. However, the accurate mechanisms connecting the metabolic disorders and alterations in gut microbial composition in host and the metabolites employed by the microorganisms in regulating the metabolic disorders is still vague. OBJECTIVE： The review delineates the latest findings about the role of gut microbiome to the pathophysiology of Obesity, IBD and Diabetes Mellitus. Here, we provide a brief introduction to the gut microbiome followed by the current therapeutic interventions in restoration of the disrupted intestinal microbiota. METHODS： A methodical PubMed search was performed using keywords like ＂gut microbiome,＂＂obesity, diabetes,＂＂IBD,＂ and ＂metabolic syndromes.＂ All significant and latest publications up to January 2018 were accounted for the review. RESULTS： Out of the 93 articles cited, 63 articles focused on the gut microbiota association to these disorders. The rest 18 literature outlines the therapeutic approaches in maintaining the gut homeostasis using probiotics, prebiotics and faecal microbial transplant （FMT）. CONCLUSION： Metabolic disorders have intricate etiology and thus a lucid understanding of the complex host-microbiome inter-relationships will open avenues to novel therapeutics for the diagnosis, prevention and treatment of the metabolic diseases.

Assessment of above- and belowground carbon pools in a semi-arid forest ecosystem of Delhi, India

The freshness of fruits is considered to be one of the essential characteristics for consumers in determining their quality,flavor and nutritional *** primary need for identifying rotten fruits is to ensure that only fresh and high-quality fruits are sold to *** impact of rotten fruits can foster harmful bacteria,molds and other microorganisms that can cause food poisoning and other illnesses to the *** overall purpose of the study is to classify rotten fruits,which can affect the taste,texture,and appearance of other fresh fruits,thereby reducing their shelf *** agriculture and food industries are increasingly adopting computer vision technology to detect rotten fruits and forecast their shelf ***,this research work mainly focuses on the Convolutional Neural Network’s(CNN)deep learning model,which helps in the classification of rotten *** proposed methodology involves real-time analysis of a dataset of various types of fruits,including apples,bananas,oranges,papayas and ***,machine learningmodels such as GaussianNaïve Bayes(GNB)and random forest are used to predict the fruit’s shelf *** results obtained from the various pre-trained models for rotten fruit detection are analysed based on an accuracy score to determine the best *** comparison to other pre-trained models,the visual geometry group16(VGG16)obtained a higher accuracy score of 95%.Likewise,the random forest model delivers a better accuracy score of 88% when compared with GNB in forecasting the fruit’s shelf *** developing an accurate classification model,only fresh and safe fruits reach consumers,reducing the risks associated with contaminated ***,the proposed approach will have a significant impact on the food industry for efficient fruit distribution and also benefit customers to purchase fresh fruits.
