Polymers are being used extensively in drug delivery due to their surface and bulk properties. They are being used in drug formulations and in drug delivery devices. These drug delivery devices may be in the form of implants for controlled drug delivery. Polymers used in colloidal drug carrier systems, consisting of small particles, show great advantage in drug delivery systems because of optimized drug loading and releasing property. Polymeric nano particulate systems are available in wide variety and have established chemistry. Non toxic, biodegradable and biocompatible polymers are available. Some nano particulate polymeric systems possess ability to cross blood brain barrier. They offer protection against chemical degradation. Smart polymers are responsive to atmospheric stimulus like change in temperature;pressure, pH etc. thus are extremely beneficial for targeted drug delivery. Some polymeric systems conjugated with antibodies/specific biomarkers help in detecting molecular targets specifically in cancers. Surface coating with thiolated PEG, Silica-PEG improves water solubility and photo stability. Surface modification of drug carriers e.g. attachment with PEG or dextran to the lipid bilayer increases their blood circulation time. Polymer drug conjugates such as Zoladex, Lupron Depot, On Caspar PEG intron are used in treatment of prostate cancer and lymphoblastic leukemia. Polymeric Drug Delivery systems are being utilized for controlled drug delivery assuring patient compliance.

The antioxidant status in human population based on the concept of prakruthi in Ayurveda

Objective:In the Ayurvedic system of medicine,body constitution(prakruthi) is said to play a vital role in the disease process as well as in its *** radical toxicity is also involved in the etiopathogenesis of many ***,in the present study an attempt has been made to correlate the body constitution type and the antioxidant status of that ***:Healthy volunteers from Manipal University Campus, South Karnataka,India were classified into three main human constitutions(prakruthi) and their possible combinations and their serum total antioxidant activity was ***:There was a significant negative correlation of the total antioxidant status with the pitta prakruthi whereas vata and kapha constitutions showed no significant correlation with the total antioxidant ***:Ayurveda classifies each individual based on his/her prakruthi into three categories of Doshas:Vata,Pitta and Kapha(collectively known as Tridoshas ). The three Doshas are fundamental regulatory principles responsible for maintaining organism functioning and manifesting those characteristics typifying each individual,that is,in equilibrium Tridoshas signify health and an imbalance leads to ***,in this study a significantly decreased antioxidant status in the Pitta group would render them more susceptible to damage by oxidative ***,we suggest that supplementation with antioxidants of herbal(Ayurvedic) origin to such a group would prove to be beneficial - opening up the arena for pharmacogenomics and customized antioxidant drug administration based on the prakruthi of an individual.

DNA damage in exfoliated buccal cells and antioxidant status of saliva in brain tumour patients

The present investigation was carried out in exfoliated buccal cells and saliva collected from pre-operative brain tumour patients. The DNA damage in these cells was assessed by alkaline comet assay and micronucleus (MN) assay. Salivary flow rate, pH, osmolality, total antioxidant activity (AOA) and vitamin C levels were also assessed in unstimulated whole saliva of these patients. In the comet assay a significant increase in the tail length (P<0.02) was observed when control and malignant groups were compared. A significant (P<0.02) difference in tail length was also noted between benign and malignant groups. Non significant results were found when control and benign groups were compared. Further, a marked increase in % MN (P<0.002) was observed when control and benign groups were compared. A significant increase in % MN (P<0.029) was also observed in benign cases when compared to malignant tumours. No significance was obtained when % MN in control and malignant cases was compared. Moreover, salivary flow rate and pH was significantly decreased and osmolality was markedly increased in brain tumour patients. The AOA levels in saliva were markedly decreased in brain tumours and vitamin C levels exhibited no change when compared to controls. Thus, as noted above susceptibility to free radical induced DNA damage also exists in the exfoliated buccal cells conducive to the lowered salivary antioxidant status of brain tumour patients.
