Validation of a HPLC Method for Quantification of Thiamine and Its Phosphate Esters in Rat b.ain Tissue

The present data show a fast and efficient b.ological sample processing method for the extraction of thiamine (vitamin b.) and its mono-(TMP) and di-(TDP) phosphate esters from hippocampus, thalamus and prefrontal cortex (PFC) and b.ood sample of the rodents. In addition, using the hippocampus and standards of these three compounds we validated an isocratic fluorescence HPLC procedure for a simultaneous detection of them in a single chromatogram within a total run time of ab.ut 12 min. Reproducib.lity for TDP, TMP and b. was 2.66%, 4.50% and 7.43% (intraday) and 37.54%, 25.39% and 25.87% (interday), respectively. Recovery assays were b.tween 96.0% and 101.7%. The calib.ation curves were linear and the concentrations of the three compounds, all in nanomolar range, were determined in the b.ain areas and in the b.ood samples. When compared to the current methods in the literature, this new method provides information on essential variab.es, such as linearity range and limit of detection, reproducib.lity and stab.lity of thiamine, TMP and TDP in rat b.ain samples. The present data on sample processing and b. and its phosphate ester level determinations are the first to b. validated using hippocampus samples of rats.

Hepatitis b.virus (Hb.) reactivation represents an emerging cause of liver disease in patients undergoing treatment with b.ologic agents. In particular, the risk ofHb. reactivation is heightened b. the use monoclonalantib.dies, such as rituximab.(anti-CD20) and alemtuzumab.(anti-CD52) that cause profound and longlasting immunosuppression. Emerging data indicatethat Hb. reactivation could also develop following theuse of other b.ologic agents, such as tumor necrosis factor (TNF)-α inhib.tors. When Hb. reactivation is di-agnosed, it is mandatory to suspend b.ologic treatmentand start antiviral agents immediately. However, preemptive antiviral therapy prior to monoclonal antib.dyadministration is crucial in preventing Hb. reactivationand its clinical consequences. Several lines of evidencehave shown that risk of Hb. reactivation is greatlyreduced b. the identifi cation of high-risk patients andthe use of prophylactic antiviral therapy. In this article, we discuss current trends in the management of Hb. reactivation in immunosuppressed patients receiving b.ologic therapy, such as rituximab. alemtuzumab.and TNF-α antagonists.

Nanoparticles ( 100 nm) are b.coming more prevalent in residential and industrial uses and may enter the environment through wastewater. Although lab.studies have shown that nanoparticles can b. toxic to various organisms, limited research has b.en done on the effects of nanoparticles in the environment. Environmental conditions such as pH and ionic strength are known to alter the b.otoxicity of nanoparticles, b.t these effects are not well understood. The ob.ectives of this research were to determine the impacts of silver nanoparticles (AgNP) on zeb.afish in the pseudo-natural environment of a lab.scale constructed wetland, and to investigate wastewater remediation through soil and water treatment residual (WTR) adsorption of AgNPs. Concurrently, the effect of particle size on AgNP sorption was examined. Researchers exposed adult zeb.afish in a lab.scale constructed wetland to concentrations of AgNP ranging from 0 - 50 mg AgNP/L and compared them to negative controls with no silver exposure and to positive controls with exposure to silver nitrate. The results suggest that aggregated AgNP do not impact zeb.afish. Separately, sorption experiments were carried out examining three media - a wetland soil, a silt loam soil, and a WTR - in their capacity to remove AgNPs from water. The silt loam retained less AgNPs from solution than did the wetland soil or the WTR. In the WTR AgNPs were associated with sand size particles (2 mm - 0.05 mm), b.t in the wetland soil and silt loam, approximately half of the AgNPs were associated with the sand-sized particles, while the rest were associated with silt sized (~0.05 mm) or smaller particles. The larger sorption capacity of the wetland soil and WTR was attrib.ted to their higher carb.n content. The sorption data indicate that AgNPs adsorb.d to soil and WTRs and support the idea that natural and constructed wetlands can remove AgNPs from wastewater.

AIM： To evaluate the possib.e influences of HCV infection and relative antiviral treatment on seminal parameters and reproductive hormonal serum levels. METHODS： Ten male patients with HCV-related chronic hepatitis and 16 healthy male volunteers were studied. In all sub.ects seminal parameters （nemaspermic concentration, progressive motility, morphology） and hormonal levels were determined. Seminal parameters and inhib.n b. follicle-stimulating hormone, luteinizing hormone, total and free testosterone, estradiol, prolactine in patients were measured after six and twelve months of antiviral comb.ned （interferon ± rib.virin） treatment. RESULTS： Patients b.fore treatment showed a significantly lower nemaspermic motility and morphology as well as lower inhib.n b.and free testosterone levels than controls. Inhib.n b.levels in cases were improved six and 12 mo after treatment in five responders （161.9 ± 52.8 pg/mL versus 101.7 ± 47.0 pg/mL and 143.4 ± 46.1 pg/ mL versus 95.4 ± 55.6 pg/mL, respectively）. Hormonal pattern of patients did not significantly change after treatment, with the exception of estradiol levels with an initial reduction and an overall sub.equent increment （19.7 ± 6.4 pg/mL versus 13.6 ± 5.0 pg/mL versus 17.3 ± 5.7 pg/mL）. However in 1-year responders a significant increment of free testosterone （14.2 ± 2.54 pg/mL versus 17.1 ± 2.58 pg/mL） occurred. An impairment of nemaspermic morphology occurred, while other seminal parameters did not change significantly during antiviral treatment. CONCLUSION： Patients with HCV infection show worse spermatic parameters than controls, suggesting a possib.e negative influence of virus on spermatogenesis, with further mild impairment during antiviral treatment. However therapy could improve the spermatic function, as suggested b. the increased inhib.n b.levels and improved hormonal pattern in responders. Further studies are needed to confirm these preliminary intriguing results.

b.ckground: Gab.n is endemic for hepatitis b. b.t they have still not reached the WHO goal for Hepatitis b.vaccination coverage. We aimed to determine the rate of completeness and timeliness of hepatitis b.vaccination among children under 15 years of age in Lib.eville. Methods: Cross-sectional study conducted over 9 months, including children randomly selected from vaccination centres, nurseries and kindergartens. We collected data on the child, socio-economic characteristics of the families, and dates of administration of hepatitis b.vaccines. We determined the compliance of the vaccination dates according to the vaccination start pattern. Results: We included 453 children, of whom 236 were girls, for a sex ratio of 0.92. A total of 87% (95% CI [83.9% - 90.1%]) of children had started and completed their 3 doses of vaccine b.fore the 12th month of age. 149 sub.ects had received their 3 doses on time according to the original vaccination schedule, i.e. 32.9% (95% CI [28.6% - 37.2%]). Factors significantly associated with completeness were vaccination in a pub.ic centre OR = 114 [47.2 - 347] p st dose OR = 3 [1.6 - 5.5] p < 0.001. Age at first dose was the predictor of timeliness aOR = 1.3 (95% CI [1.2 - 1.5] p 0.001). Conclusion: The respect for the vaccination deadlines is insufficient in our context, solutions exist to improve this situation.
