In.the presen. study we in.estigated structural an. metabolic modification. of the brain.in.the ts65Dn.mouse model of Down.syn.rome(DS)usin. both in.vivo magn.tic reson.n.e imagin.(MRI)an. proton.magn.tic reson.n.e spectroscopy(MRS). MRI was performed for further texture an.lysis an. chan.es in.texture parameters, in.ludin. mean.grey levels, con.rast an. homogen.ity, an. they were foun. in.ts65Dn.compared to diploid littermates (2n.. These phen.typic chan.es were differen. in.the hippocampus an. cerebellum, sin.e in.ts65Dn.mean.grey levels in.reased in.the cerebellum an. decreased in.the hippocampus. In.addition. proton.n.R spectra revealed differen.es in.metabolite ratios. Levels of n.acetylaspartate(n.A)an. glutamate(Glu), were lower compared to total creatin. levels (CX), in.the ts65Dn.brain. However, the most strikin. fin.in. was an.in.rease in.the con.en.ration.of myo-in.sitol(In.)an. cholin.(Cho)in.the hippocampus, whereas the In. con.en.ration.was reduced in.the cerebellum. Overall, these data illustrate that MRI an. MRS are valuable assesmen. tools sufficien.ly sen.itive to detect associated chan.es in.differen. brain.areas, thus providin. n.w in.ight in.o the causative role of dosage-sen.itive gen.s in.the ts65Dn.DS mouse model.

Purpose: It is thought that the mechan.sms respon.ible for impaired chron.tropic respon.e to exercise are related to disturban.e of cardi ovascular auton.mic regulation.such as the baroreflex. However, it is still un.l ear whether the baroreflex mechan.sms modulate heart rate(HR) respon.es to exerc ise via vagal an./or sympathetic alteration. We therefore in.estigated the effec ts of baroreflex sen.itivity (BRS) on.the exercise HR respon.e in.the early phas e of vagal deactivation.an. in.the later phase of sympathetic excitation.via met abolic stimulation. Methods: Twen.y-four patien.s(18 males an. 6 females, age 5 9±9 years) with heart disease un.erwen. symptom-limited treadmill exercise tes tin. accordin. to the Bruce protocol, an. BRS was measured utilizin. the phen.le phrin. method. Subjects were grouped by their restin. BRS value: 12 with n.rmal BRS(≥6 ms/mm Hg) an. 12 with depressed BRS(n.e to exe rcise was assessed usin. two parameters: the in.remen. in.HR durin. exercise an. the ratio of HR respon.e to the metabolic reserve(chron.tropic in.ex). Results: (1) In.the patien.s with depressed BRS, the HR respon.es within.1 min.after the start of exercise an. from 1 min.to peak exercise were atten.ated compared with those havin. a n.rmal BRS (15±8vs. 24±8 bpm an. 36±9vs. 47±15 bpm, respecti vely). (2) The chron.tropic in.ex in.the patien.s with depressed BRS was lower t han.in.those with n.rmal BRS(0.50±0.14 vs. 0.64±0.08). Con.lusion. These fin.i n.s suggest that impaired BRS modulates both the parasympathetic in.luen.e in.ea rly exercise an. sympathetic effects in.the later phase on.HR respon.e to exerci se.

Man. local hospitals have reported a surge of cases, with most cases presen.in. with severe respiratory complication.. We aim to presen. a comprehen.ive surge filter protocol for outpatien. programs to mitigate the surge of cases admitted in.the emergen.y rooms an. in.en.ive care un.ts (ICUs) of local hospitals in.our commun.ty. Our surge filter protocol was developed based on.a detailed review of how South Korea an. Spain.moun.ed their outpatien. surge respon.es. Our prelimin.ry fin.in.s revealed that our Four-ts approach (i.e., Test, Treat, Track, an. Teach) has resulted in.n.n. of our patien.s reachin. a clin.cal requiremen. for in.atien. care.

通过克隆人质子感知受体G2A基因、构建G2A基因的表达载体并瞬时转染293T细胞,建立G2A受体调控机理及其功能研究的转基因细胞模型,并检测G2A介导的细胞应答.从人脐静脉内皮细胞(Human.Umbilical Vein.En.othelial Cells,HUVECs)中提取总Rn.,反转录为cDn.为模板,设计一对针对G2A基因的引物,克隆不含终止密码子的G2A基因并亚克隆到pMD-19T载体.将鉴定出的阳性质粒和表达载体pEGFP-n.用Hin.Ⅲ和BamHⅠ双酶切回收后连接获得阳性重组质粒pEGFP-n.-G2A,并测序鉴定;重组载体以lipofectamin.2000介导转染293T细胞.Real time PCR法检测外源基因在293T细胞中的表达;ELISA检测转染与未转染G2A基因的细胞内三磷酸肌醇(In.sitol-1,4,5-triphosphosate,IP3)的含量.测序和酶切证实了克隆到G2A基因并获得pEGFP-n.-G2A表达载体;荧光成像与Real time PCR证实在293T细胞中过表达G2A基因;ELISA检测在酸性刺激下转基因细胞内IP3含量(应答)明显升高,而脂质配体LPC则抑制其IP3的积累,本研究为G2A受体介导信号机制及其功能研究提供一个真核细胞模型.

Polypyrrole (Ppy) was syn.hesized usin. Ferric Chloride (FeCl3) as oxidan.. The ratio of mon.mer to oxidan. was 1:2.4. The polymerization.was carried out at 5。C. Attempts were made to in.rease the electrical con.uc-tivity by usin. various dopan.s viz. Lithium per Chlorate (LiClO4), para-Toluen. Sulfon.te (p-ts) an. n.p-thalen. Sulfon.c acid (n.A). The materials were characterized usin. FTIR, X-Ray diffraction.an. SEM. The electrical con.uctivity was measured by two probe method an. was foun. to be in.the ran.e of 10-3 to 10-2 S/cm. Thin.films of these preparation. were casted on.the in.erdigited electrodes to study the detection.of gases such as ammon.a an. LPG. It was foun. that for the pure Ppy an. on. doped with LiClO4, when.ammon.a gas was allowed to flow in. there was a sudden.in.rease in.the curren., which decreased rapidly when.gas was stopped. This behavior is in.con.radiction.to that observed in.earlier in.estigation. where Ppy was prepared by electrochemical method. However when.Ppy doped with p-ts an. n.A were used, the tren. was reversed. Similarly it was n.ted that for pure Ppy an. on. doped with LiClO4, p-ts an. n.A there was an.in.rease in.the curren. when.LPG gas was brought in. However the respon.e to LPG was somewhat low as compared ammon.a gas.

For the effect of thermal treatmen. on.the mode-I fracture toughn.ss(FT), three crystallin. rocks(two basalts an. on. ton.lite) were experimen.ally in.estigated. Semi-circular ben. specimen. of the rocks were prepared followin. the method suggested by the In.ern.tion.l Society for Rock Mechan.cs(ISRM)an. were treated at various temperatures ran.in. from room temperature(25 ℃) to 600 ℃. Mode-I FT was correlated with ten.ile stren.th(ts), ultrason.c velocities, an. Youn.’s modulus(YM). Addition.lly,petrographic an. X-ray diffraction.an.lyses were carried out to fin. the chemical chan.es resultin. from the heat treatmen.. Further, scan.in. electron.microscopy(SEM) was con.ucted to observe the micro structural chan.es when.subjected to high temperatures. These experimen.s demon.trate that heat treatmen. has a stron. n.gative impact on.the FT an. mechan.cal properties of the rocks. From room temperature to 600 ℃, mode-I FT values of massive basalt, gian. plagioclase basalt, an. ton.lite were reduced by n.arly 52%, 68%, an. 64%, respectively. Also, at all temperature levels, FT an. mechan.cal properties are foun. to be expon.n.ially correlated. However, the exact n.ture of the relation.hip main.y depen.s on.rock type. Besides, ts was foun. to be a better in.icator of degradation.degree than.the mode-I FT. SEM images show that micro crack den.ity an. structural disin.egration.of the min.ral grain. in.rease with temperature. These physical chan.es con.irm the observed reduction.in.the stiffn.ss of heat-treated crystallin. rocks.
