Generation of Eco-Friendly and Disease-Resistant Channel Catfish(Ictalurus punctatus)Harboring the Alligator Cathelicidin Gene via CRISPR/Cas9 Engineering

As a precise and versatile tool for genome manipulation,the clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR-associated protein 9(Cas9)platform holds promise for modifying fish traits of *** the aim of reducing transgene introgression and controlling reproduction,upscaled disease resistance and reproductive intervention in catfish species have been studied to lower the potential environmental risks of the introgression of escapees as transgenic *** advantage of the CRISPR/Cas9-mediated system,we succeeded in integrating the cathelicidin gene(As-Cath)from an alligator(Alligator sinensis)into the target luteinizing hormone(lh)locus of channel catfish(Ictalurus punctatus)using two delivery systems assisted by double-stranded DNA(dsDNA)and single-stranded oligodeoxynucleotides(ssODNs),*** this study,high knock in(KI)efficiency(22.38%,64/286)but low ontarget events was achieved using the ssODN strategy,whereas adopting a dsDNA as the donor template led to an efficient on-target KI(10.80%,23/213).The on-target KI of As-Cath was instrumental in establishing the lh knockout(LH^(–)_As-Cath^(+))catfish line,which displayed heightened disease resistance and reduced fecundity compared with the wild-type(WT)sibling ***,administration of human chorionic gonadotropin(HCG)and luteinizing hormone-releasing hormone analogue(LHRHa)can restore the reproduction of the transgenic fish ***,we replaced the lh gene with an alligator cathelicidin transgene and then administered hormone therapy to move towards complete reproductive control of diseaseresistant transgenic catfish in an environmentally responsible *** strategy not only effectively improves consumer-valued traits but also guards against unwanted introgression,providing a breakthrough in aquaculture genetics to confine fish reproduction and prevent the establishment of transgenic or domestic genotypes in the natural environment.

Experimental Modelling of Transverse Oscillations in Aquaculture Netting Parallel to the Flow – Sounds Baffling

Numerous studies have been undertaken to improve the viability, durability and suitability of materials and methods used for aquaculture enclosures. While many of the previous studies considered macro-deformation of nets, there is a paucity of information on netting micro-deformation. When aquaculture pens are towed, industry operators have observed the motion described as ＂baffling＂ – the transverse oscillation of the net planes parallel and near parallel to the flow. The difficulty to observe and assess baffling motion in a controlled experimental environment is to sufficiently reproduce netting boundary conditions and the flow environment experienced at sea. The focus of the present study was to develop and assess experimental methods for visualisation and quantification of these transverse oscillations. Four netrig configurations with varied boundary conditions and model-netting properties were tested in a flume tank. While the Reynolds number was not equivalent to full-scale, usage of the pliable and fine mesh model netting that enabled baffling to develop at low flow velocities was deemed to be of a larger relevance to this initial study. Baffling was observed in the testing frame that constrained the net sheet on the leading edge, similarly to a flag attachment onto a pole. Baffling motion increased the hydrodynamic drag of the net by 35%–58% when compared to the previously developed formula for taut net sheets aligned parallel to the flow. Furthermore, it was found that the drag due to baffling decreased with the increasing velocity over the studied Reynolds numbers（below 200）; and the drag coefficient was non-linear for Reynolds numbers below 120. It is hypothesised that baffling motion is initially propagated by vortex shedding of the netting twine which causes the netting to oscillate; there after the restoring force causes unstable pressure differences on each side of the netting which excites the amplitude of the netting oscillations.

On Honesty and Deceit： An Interpretation of Difference and Repetition

Foucault famously questioned the humor, and possibly the deceit, at the heart of Deleuze＇s philosophy by asserting its sincerity and seriousness. In this paper I address how humor and sincerity, and thereby truth and falsity, are philosophical concepts at the core of Deleuze＇s thought. Using Deleuze＇s work Difference and Repetition as the focal object of my interrogation, I seek to show how truth and falsity are structured by difference and repetition. Deleuze＇s philosophy of pure difference does not merely try to critique traditional notions of truth, but attempts to define a new model of truth and falsity that bears both differences and repetitions from the previous models in the history of philosophy.

The False Promise of Kant＇s Humanity Formula

Kant＇s Humanity Formula of the Categorical Imperative is arguably its most widely preferred formulation, having been defended as a moral principle and employed in the evaluation of particular moral problems by a number of leading contemporary ethicists. For them and many other readers of Kant, the idea that we are not to treat persons as mere means to our own ends but are rather to respect their rational agency as intrinsically valuable holds great promise for qualifying as, in Kant＇s words, ＂the supreme principle of morality.＂ In the present paper I argue that the Humanity Formula cannot deliver on this promise. After setting forth three conditions of adequacy for any genuine supreme principle of morality, I argue that the Humanity Formula, on three textually grounded interpretations each of which has been advocated by a prominent Kantian ethicist, does not satisfy these conditions. Whichever of these textually grounded interpretations is taken, the Humanity Formula is open to compelling counterexamples.

Metals discharged from industrial effluents,agricultural wastewater,and sewage runoff by rapid urbanization are of concern as contaminants of freshwater ecosystem because of their persistence and high toxicity to aquatic *** study attempted to identify which metal posed the greatest risk to freshwater ecosystem in the Bohai Region,*** metals arsenic(As),cadmium(Cd),chromium(Cr),copper(Cu),zinc(Zn),lead(Pb),nickel(Ni),mercury(Hg),iron(Fe),and manganese(Mn)were compared against norfloxacin and gamma-hexachlorocyclohexane(lindane).By comparing the median reported environmental and ecotoxicity concentrations,it showed that Cu,Fe,Zn,Mn,and Cr were the top five metals of *** these,Cu was deemed to represent the highest risk and Hg the lowest *** risks for all metals were higher than those for norfloxacin and *** all the metals except Hg had water concentrations that exceeded levels where ecotoxicity effects had been recorded in the literature.A comparison with the measurements across the UK rivers suggested that all metals examined had water concentrations about 5-to 10-fold higher than the UK median values except for Cu,Fe,Cd,and *** Fuyang River,a tributary of the Haihe River Basin,seemed to be the location with the highest metal ***,comparing the post-2010 period to 2000-2009,concentrations of all the metals had fallen except for Fe and Mn,so risks have decreased over the last 7 yr with the greatest improvements for Cd and *** metals still pose high risks to freshwater ecosystem in this region,there is encouragement that some control measures are taken into effect.

Pollution and climate change are among the most challenging issues for countries with developing economies,but we know little about the ecological risks that result when these pressures occur *** explored direct effects of,and interactions between,environmental pollution and climate change on ecosystem health in the Bohai Sea region of Northern *** developed an integrated approach to assess ecological risks to this region under four scenarios of climate *** ecological risks to the system from pollution alone have been declining,interactions between pollution and climate change have enhanced ecological risks to this coastal/marine *** results suggest that current policies focused strictly on pollution control alone should be changed to take into account the interactive effects of climate change so as to better forecast and manage potential ecological risks.

Diagnosis and Treatment of Tolosa-Hunt Syndrome in the Emergency Department

We describe herein a patient who presented with painful ophthalmoplegia and was ultimately diagnosed via magnetic imaging resonance studies and successfully treated for Tolosa-Hunt syndrome. Tolosa-Hunt syndrome is a rare, reversible and painful ophthalmoplegia characterized by recurrent unilateral orbital pain, ipsilateral oculomotor paralysis and prompt response to steroids. Specific criteria for its diagnosis exist and are discussed herein. Individuals affected may display signs of select cranial nerve palsies, ptosis, facial numbness, diplopia, midrosis, and proptosis. Appropriate recognition of the disease can allow for immediate intervention and thus decrease the length and severity of symptoms especially as symptoms may not spontaneously resolve without treatment which leads to unnecessary suffering through pain, anxiety, and decreased vision. We describe the case presentation and keys for diagnosis emergency medicine that physicians should know for this potentially devastating condition.
