Adjustment of Forest Ecosystem Root Respiration as Temperature Warms

Adjustment of ecosystem root respiration to warmer climatic conditions can alter the autotrophic portion of soil respiration and influence the amount of carbon available for biomass production. We examined 44 published values of annual forest root respiration and found an increase in ecosystem root respiration with increasing mean annual temperature （MAT）, but the rate of this cross-ecosystem increase （Q10 = 1.6） is less than published values for short-term responses of root respiration to temperature within ecosystems （Q10 = 2-3）. When specific root respiration rates and root biomass values were examined, there was a clear trend for decreasing root metabolic capacity （respiration rate at a standard temperature） with increasing MAT. There also were tradeoffs between root metabolic capacity and root system biomass, such that there were no instances of high growing season respiration rates and high root biomass occurring together. We also examined specific root respiration rates at three soil warming experiments at Harvard Forest, USA, and found decreases in metabolic capacity for roots from the heated plots. This decline could be due to either physiological acclimation or to the effects of co-occurring drier soils on the measurement date. Regardless of the cause, these findings clearly suggest that modeling efforts that allow root respiration to increase exponentially with temperature, with Q10 values of 2 or more, may over-predict root contributions to ecosystem CO2 efflux for future climates and underestimate the amount of C available for other uses, including net primary productivity.

A generic framework for geotechnical subsurface modeling with machine learning

This study introduces a generic framework for geotechnical subsurface modeling, which accounts for spatial autocorrelation with local mapping machine learning(ML) methods. Instead of using XY coordinate fields directly as model input, a series of autocorrelated geotechnical distance fields(GDFs) is designed to enable the ML models to infer the spatial relationship between the sampled locations and unknown locations. The whole framework using GDF with ML methods is named GDF-ML. This framework is purely data-driven which avoids the tedious work in the scale of fluctuations(SOFs)estimating and data detrending in the conventional spatial interpolation methods. Six local mapping ML methods(extra trees(ETs), gradient boosting(GB), extreme gradient boosting(XGBoost), random forest(RF), general regression neural network(GRNN) and k-nearest neighbors(KNN)) are compared in the GDF-ML framework. The results show that the GDFs are better than the conventional XY coordinate fields based ML methods in both accuracy and spatial continuity. GDF-ML is flexible which can be applied to high-dimensional, multi-variable and incomplete datasets. Among these six methods, GDF with ET method(GDF-ET) clearly shows the best accuracy and best spatial continuity. The proposed GDF-ET method can provide a fast and accurate interpretation of the soil property profile. Sensitivity analysis shows that this method is applicable to very small training dataset size. The associated statistical uncertainty can also be quantified so that the reliability of the subsurface modeling results can be estimated objectively and explicitly. The uncertainty results clearly show that the prediction becomes more accurate when more sampled data are available.

Effect of feeding broilers diets differing in susceptible phytate content

Measurements of total phytate phosphorus content of diets may be deceptive as they do not indicate substrate availability for phytase; it may be that measurements of phytate susceptible to phytase effects are a more accurate measure of phosphorus(P) availability to the bird. To verify this hypothesis, an experiment was conducted to compare diets formulated to contain either high or low susceptible phytate, supplemented with either 0 or 500 FTU/kg phytase. Susceptible phytate was determined by exposing the feed samples to conditions that mimicked the average pH of the proximal gastrointestinal tract(pH 4.5) and the optimum temperature for phytase activity(37℃) and then measuring phytate dissolved. Ross 308 birds(n = 240) were fed one of 4 dietary treatments in a 2 x 2 factorial design; 2 diets with high(8.54 g/kg, 57.90% of total phytate) or low(5.77 g/kg, 46.33% of total phytate) susceptible phytate, containing 0 or 500 FTU/kg phytase. Diets were fed to broilers(12 replicate pens of 5 birds per pen) from d 0 to 28 post hatch. Birds fed diets high in susceptible phytate had greater phytate hydrolysis in the gizzard(P jejunum(P jejunum and ileum(P < 0.05) and resulting greater tibia and femur Ca and P(P < 0.05) content than those fed the low susceptible diets. All the susceptible phytate was fully degraded in the tract in the absence of added phytase, suggesting the assay used in this study was able to successfully estimate the amount of total dietary phytate that was susceptible to the effects of phytase when used at standard levels. Background and Aims:High-intensity interval training(HIIT)is a therapeutic option for people with nonalcoholic steatohepatitis(NASH).However,the perspectives and experiences of HIIT for people with NASH are unknown,limiting translation of *** explored the experiences and perspectives of both professionally supervised and self-directed HIIT in people with NASH and evaluated participant-reported knowledge,barriers,and enablers to commencing and sustaining ***:Twelve participants with NASH underwent 12 weeks of supervised HIIT(3 days/week,4×4 minutes at 85–95%maximal heart rate,interspersed with 3 minutes active recovery),followed by 12-weeks of selfdirected(unsupervised)***-on-one,semistructured participant interviews were conducted by exercise staff prior to HIIT and following both supervised and self-directed HIIT to explore prior knowledge,barriers,enablers,and outcomes at each *** were audio-recorded,transcribed,coded,and thematically analyzed by two independent ***:Four dominant themes were identified:(1)no awareness of/experience with HIIT and ambivalence about exercise capabilities;(2)multiple medical and social barriers to commencing and continuing HIIT;(3)exercise specialist support was a highly valued enabler,and(4)HIIT was enjoyed and provided holistic ***:People with NASH may lack knowledge of and confidence for HIIT,and experience multiple complex barriers to commencing and continuing *** specialist support is a key enabler to sustained *** factors need to be addressed in future clinical programs to augment the uptake and long-term sustainability of HIIT by people with NASH so they can experience the range of related benefits.

Versican belongs to the family of the large aggregating chondroitin sulfate proteoglycans located primarily within theextracellular matrix (ECM). Versican, like other members of its family, has unique N- and C-terminal globular regions,each with multiple motifs. A large glycosaminoglycan-binding region lies between them. This review will begin byoutlining these structures, in the context of ECM proteoglycans. The diverse binding partners afforded to versican byvirtue of its modular design will then be examined. These include ECM components, such as hyaluronan, type Icollagen, tenascin-R, fibulin-1, and -2, fibrillin-1, fibronectin, P- and L-selectins, and chemokines. Versican also binds tothe cell surface proteins CD44, integrin β1, epidermal growth factor receptor, and P-selectin glycoprotein *** multiple interactors play important roles in cell behaviour, and the roles of versican in modulating such processesare discussed.

High-throughput quantification of microbial birth and death dynamics using fluorescence microscopy

Background: Microbes live in dynamic environments where nutrient concentrations fluctuate. Quantifying fitness in terms of birth rate and death rate in a wide range of environments is critical for understanding microbial evolution and ecology. Methods^ Here, using high-throughput time-lapse microscopy, we have quantified how Saccharomyces cerevisiae mutants incapable of synthesizing an essential metabolite (auxotrophs) grow or die in various concentrations of the required metabolite. We establish that cells normally expressing fluorescent proteins lose fluorescence upon death and that the total fluorescence in an imaging frame is proportional to the number of live cells even when cells form multiple layers. We validate our microscopy approach of measuring birth and death rates using flow cytometry, cell counting, and chemostat culturing. Results: For lysine-requiring cells, very low concentrations of lysine are not detectably consumed and do not support cell birth, but delay the onset of death phase and reduce the death rate compared to no lysine. In contrast, in low hypoxanthine, hypoxanthine-requiring cells can produce new cells, yet also die faster than in the absence of hypoxanthine. For both strains, birth rates under various metabolite concentrations are better described by the sigmoidal-shaped Moser model than the well-known Monod model, while death rates can vary with metabolite concentration and time. Conclusions: Our work reveals how time-lapse microscopy can be used to discover non-intuitive microbial birth and death dynamics and to quantify growth rates in many environments.
