Bufalin Suppresses Esophageal Squamous Cell Carcinoma Progression through Protein Arginine Methyltransferase-6/AKT/Mammalian Target of Rapamycin Pathway Inhibition

Objective: Esophageal squamous cell carcinoma(ESCC), a commonly encountered malignant tumor in the gastrointestinal tract, poses a significant health burden. Bufalin, a pharmacologically active molecule, has been shown to exhibit antitumor activity against various types of cancers. This study investigates the molecular mechanism underpinning the effects of bufalin on ESCCs. Materials and Methods: The impact of bufalin on the proliferation and migration of ESCC cells was evaluated through the utilization of the Cell Counting Kit-8(CCK-8), scratch assay, and transwell assay. In addition, ribonucleic acid(RNA) sequencing was performed to identify genes that were abnormally expressed in response to bufalin treatment. Western blotting was utilized to ascertain the expression levels of protein arginine methyltransferase-6(PRMT6), phosphorylated AKT(p-AKT), and phosphorylated mammalian target of rapamycin(p-m TOR). Cell transfection was then performed to observe the rescue effect of PRMT6 on bufalin. Results: Bufalin displayed a significant time-dependent inhibition of the proliferation, migration, and invasion of ECA109 cells. An RNA sequencing(RNA-seq) analysis revealed that PRMT6 expression was downregulated in the cells treated with bufalin. PRMT6 promoted the proliferation, migration, and invasive potential of ECA109 cells. The overexpression of PRMT6 boosted p-AKT and p-m TOR levels in ECA109 cells and reversed bufalin inhibition. Conclusions: These findings indicate that bufalin exerts its inhibitory effects on ESCCs through the PRMT6/AKT/m TOR signaling pathway. These findings lay the groundwork for bufalin as a promising therapeutic candidate for the treatment of ESCC.

The immune system is involved in many age-related pathological changes,also plays an important role in tissue regeneration after *** no immune involvement has been discussed regarding cataract since it is presumed that lens has no source of immune cells as an avascular *** research has challenged the longstanding view of the lens as an immune-privileged tissue,revealing the presence of resident immune cells and active immune responses within the ***,we summarized the immune involvement in maintaining lens homeostasis,which may be a deleterious role in the induction of lens opacification if inappropriately ***,bioengineer-based immunomodulatory therapies to fine-tune the micro immune environment within lens may be future strategies for in situ lens regeneration,as a novel treatment for cataract.

Genome-wide identification and characterization of putative effectors in Heterodera schachtii that suppress plant immune response

The sugar beet cyst nematode(Heterodera schachtii) is one of the most destructive pathogens in sugar beet production, which causes serious economic losses every year. Few molecular details of effectors of H. schachtii parasitism are known. We analyzed the genome and transcriptome data of H. schachtii and identified multiple potential predicted proteins. After filtering out predicted proteins with high homology to other plant-parasitic nematodes, we performed functional validation of the remaining effector proteins. 37 putative effectors of H. schachtii were screened based on the Nicotiana benthamiana system for identifying the effectors that inhibit plant immune response, eventually determines 13 candidate effectors could inhibit cell death caused by Bax. Among the 13 effectors, nine have the ability to inhibit GPA2/RBP1-induced cell death. All 13 effectortriggered immunity(ETI) suppressor genes were analyzed by qRT-PCR and confirmed to result in a significant downregulation of one or more defense genes during infection compared to empty vector. For in situ hybridization,13 effectors were specifically expressed and located in esophageal gland cells. These data and functional analysis set the stage for further studies on the interaction of H. schachtii with host and H. schachtii parasitic control.

3D Printing of Tough Hydrogel Scaffolds with Functional Surface Structures for Tissue Regeneration

Hydrogel scaffolds have numerous potential applications in the tissue engineering ***,tough hydrogel scaffolds implanted in vivo are seldom reported because it is difficult to balance biocompatibility and high mechanical *** by Chinese ramen,we propose a universal fabricating method(printing-P,training-T,cross-linking-C,PTC&PCT)for tough hydrogel scaffolds to fill this ***,3D printing fabricates a hydrogel scaffold with desired structures(P).Then,the scaffold could have extraordinarily high mechanical properties and functional surface structure by cycle mechanical training with salting-out assistance(T).Finally,the training results are fixed by photo-cross-linking processing(C).The tough gelatin hydrogel scaffolds exhibit excellent tensile strength of 6.66 MPa(622-fold untreated)and have excellent ***,this scaffold possesses functional surface structures from nanometer to micron to millimeter,which can efficiently induce directional cell ***,this strategy can produce bionic human tissue with mechanical properties of 10 kPa-10 MPa by changing the type of salt,and many hydrogels,such as gelatin and silk,could be improved with PTC or PCT *** experiments show that this scaffold can effectively promote the new generation of muscle fibers,blood vessels,and nerves within 4 weeks,prompting the rapid regeneration of large-volume muscle loss injuries.

Ultraviolet spectral broadening by stimulated rotational Raman scattering on nitrogen pumped with signal laser injection

We present experimental results on kilojoule ultraviolet laser output with 1%spectral *** stimulated rotational Raman scattering(SRRS)with signal laser injection,we achieve effective spectral broadening in short-range propagation,with good retention of the original near-field distribution and time *** calculations show that 2%bandwidth spectral broadening can be achieved by injecting 20 kW~cm^(2) signal light at 2.2 GW~cm^(2) flux of the pump *** addition,high-frequency modulation in the near field can be effectively avoided through replacement of the original random noise signal light by the controllable signal *** SRRS in the atmospheric environment excited with signal laser injection can provide wide-band light output with controllable beam quality without long-distance propagation,representing an important potential route to realization of broadband laser drivers.

Preparation and thermal stability of Pd_(40.5)Ni_(40.5)Si_xP_(19-x) bulk metallic glasses

With the addition of Si to replace some P,Pd40.5Ni40.5SixP19-x（x=0,2.5,5,9.5,14,and 19 in atomic number fraction） bulk glassy samples with the diameter of about 5 mm were successfully prepared by use of flux treatment and water quenching *** the increase of Si content,the glass forming ability of Pd40.5Ni40.5Si-xP19-x increases first for low Si content and then decreases for high Si content （Si≥9.5at%）.The Pd40.50Ni40.5Si5P14 glassy alloy possesses the largest supercooled liquid region△T of 119 K,the largest reduced glass transition temperature of 0.621,and the largestγparameter of 0.460,indicating that this glassy alloy possesses very large glass forming ability and very high thermal stability.

Surface modification of acrylate intraocular lenses with dielectric barrier discharge plasma at atmospheric pressure

Surface modification with dielectric barrier discharge(DBD) plasma was carried out at atmospheric pressure(argon as the discharge gas) to improve the biocompatibility of hydrophobic acrylate intraocular lens(IOL).Changes of the plasma-treated IOL surface in chemical composition,morphology and hydrophilicity were comprehensively evaluated by X-ray photoelectron spectroscopy(XPS),field emission scanning electron microscopy(FESEM),atomic force microscopy(AFM) and water contact angle(WCA) *** surface biocompatibility of the untreated and plasma-treated IOLs was compared with the adhesion behavior of platelets,macrophages and lens epithelial cells(LECs) in *** DBD plasma treatment,the hydrophilicity of the IOL surface was obviously *** changes in WCA with treatment extension may be attributed to both the introduction of oxygen or/and nitrogen-containing polar groups and the increase of surface roughness induced by plasma etching *** existence of low molecular weight oxidized material(LMWOM) was proved on the plasma-treated IOL which was caused by the chain scission effect of the plasma *** plasma-treated IOLs resisted the adhesion of platelets and macrophages *** LECs spreading and proliferation were postponed on the IOLs plasma-treated for more than 180 s,with a well maintained epithelial phenotype of *** IOL biocompatibility was improved after the DBD plasma *** speculate that slighter foreign-body reaction and later incidence of anterior capsule opacification(ACO) may be expected after implantation of the argon DBD plasma-treated IOL.

Age-related macular degeneration (AMD) is still an incurable blinding eye disease because of complex pathogenic mechanisms and unusual diseased *** the use of chemical biology tools,great progress has been achieved in improving the understanding of AMD *** severity of AMD is,at least in part,linked to the non-degradable lipofuscin bis-retinoids in retinal pigment epithelial (RPE).This material is thought to result from the lifelong accumulation of lysosomal residual bodies containing the end products derived from the daily phagocytosis of rod outer segments by RPE ***,we present previously recognized bis-retinoids with focus on structures and biosynthetic *** addition to a brief discussion on the mutual conversion relationships of bis-retinoids,future perspectives and the medical relevance of such studies on these lipofuscin constituents are also highlighted.

The exoskeleton is employed to assist humans in various domains including military missions,rehabilitation,industrial operation,and activities of daily living(ADLs).Walking is a fundamental ADL,and exoskeletons are capable of reducing the activation and metabolism of lower extremity muscles through active assistance during *** improve the performance of active assistance strategy,this article proposes a framework using an active hip ***,it correlates to an already established Bayesian-based human gait recognition algorithm,with a particular focus on linear and circular walking within industrial and ADL *** theorizing this strategy for exoskeletons,this study further reveals,in part,the activation characteristics of human hip muscles for the instruction and regulation of active assistance duration and onset *** proposed active assistance strategy provides new insights for enhancing the performance of assistive robots and facilitating human robot interaction within the context of ADLs.
