Clinical impact of plasma TGF-β1 and circulating TGF-β1 mRNA in diagnosis of hepatocellular carcinoma

BACKGROUND:Transforming growth factor-β(TGF-β) plays an important role in the regulation of cell growth and differentiation,angiogenesis,extracellular matrix formation,immunosuppression and cancer development. In this study,we investigated the levels of TGF-β1 and TGF-β1 mRNA expression,their relationship with HBV replication,and their diagnostic value for hepatocellular carcinoma(HCC). METHODS:Total RNAs were extracted from HCC samples and matched non-tumor tissues,and from peripheral blood mononuclear cells in HCC ***-β1 mRNA was amplified by RT-PCR and confirmed by DNA sequencing. The distribution of TGF-β1 expression was assessed by *** clinical characteristics were analyzed between TGF-β1 and HBV *** diagnostic value of circulating TGF-β1 and TGF-β1 mRNA levels were investigated in HCC patients. RESULTS:The incidence of hepatic TGF-β1 expression was 83.3%in HCC samples,43.3%in the surrounding tissues, 94.7%in the HBV DNA-positive group,and 63.6%in the HBV DNA-negative *** TGF-β1 expression was associated with the degree of HCC differentiation and the status of HBV replication,but not with the size or number of *** TGF-β1 level and incidence of TGF-β1 mRNA were significantly higher in the HCC groupthan in any group of patients with benign liver disease, with a higher sensitivity of 89.5%and a specificity of 94.0% for HCC diagnosis when circulating TGF-β1 levels were >1.2μg/*** significant correlation was found between TGF-β1 expression and AFP level or tumor *** TGF-β1 level and serum AFP raised the detection rate to 97.4%. CONCLUSIONS:Abnormal expression of hepatic TGF-β1 is associated with the degree of HCC differentiation and HBV *** circulating TGF-β1 and TGF-β1 mRNA can be used as sensitive biomarkers for the diagnosis and prognosis of HBV-induced HCC.

BACKGROUND: Angiogenesis is known to be essential to the survival, growth, invasion, and metastasis of tumor cells. Vascular endothelial growth factor (VEGF) are an important angiogenic factor regulating tumor angiogenesis, but its significance and tumor pathologic features are un- clear in hepatocellular carcinoma (HCC). In the present study, we analyzed expression of tissue VEGF, alteration of microvascular density (MVD) in microvessel angiogenesis, development and metastasis of HCC, and level of serum VEGF in differential diagnosis of benign and malignant liv- er diseases. METHODS: Tumor specimens were prospectively collected from HCC patients undergoing resection. Total RNAs were extracted and the expression levels were detected from different parts of HCC tissues. The cellular distributions of VEGF and MVD of liver tumors and their paracancerous and distal cancerous tissues were investigated by streptavi- din peroxidase (S-P) immunohistochemistry, respectively. The VEGF levels of circulating blood and hepatoma tissues were measured by enzyme-linked immunosorbent assay. RESULTS: The incidence of VEGF expression was 63.9% in HCCs (23/36 cases), 78.3% in non-encapsulated HCCs (18/23), and 90.9% in HCCs with extrahepatic metastasis (10/11), respectively. The VEGF expression was tightly correlated with MVD (P <0.01). The MVD in HCC with metastasis, low differentiation or non-encapsulation was significantly higher than that in HCC with intact capsule, high differentiation, or no metastasis. No significant diffe- rence was found between VEGF, MVD, tumor size, and hepatitis virus infection. The level of total RNA in HCC tis- sues was significantly lower but the VEGF level significantly higher than those in paracancerous or distal cancerous ones (P<0.01). The abnormal expression levels of VEGF in sera of HCC patients were directly correlated with the me- tastasis and recurrence of tumors. BACKGROUND: There are many methods for myelin staining, mordant, or oil-soluble dye or the special reaction of osmic acid with lipoid is used according to different principles. The commonly used methods are classic Well staining, classic lithium carbonate-haematine staining, fast green staining, silver staining, etc. Luxol Fast Blue can brightly stain myelin sheath, and has certain specificity. The background can be very clean if there is proper differentiation, whereas Luxol Fast Blue is cheap and convenient to operate, thus it is an ideal staining reagent for routine myelin sheath. OBJECTIVE: To show the corticospinal tract of normal adult rats with Luxol Fast Blue staining method. DESIGN: A repetitive measurement design. SETTINGS: institute of Nuerobiology, nantong university; Department of Rehabilitation Medicine, Affiliated Hospital of nantong university. MATERIALS: Six healthy adult male SD rats of clean degree, weighing averagely 300 g, were provided by the experimental animal center of nantong university. 1 g/L Luxol Fast Blue solution was provided by Sigma Company; Leica CM1900 cryostat microtome by Leica Company; Leica DMR microscope by Leica Company. METHODS: The experiment was carried out in the Staff Room of Human Anatomy, nantong university in May 2005. The rats were given intraperitoneal injection of combined anesthetic (2 mL/kg), then the chest was open for perfusing saline and phosphate buffer containing formamint via heart. Brain and spinal cord were removed after 1 hour then fixed, then changed to phosphate buffer (pH 7.4) containing 300 g/L saccharu at 4 ℃, and stayed overnight, tissue blocks at pyramid, decussation of pyramid and cervical, thoracic, lumbar and sacral segments of spinal cord were removed to prepare continuous horizontal frozen sections (30 μm) after sedimentation, the sections were dried at room temperature. The corticospinal tract of normal adult rats were shown with Luxol Fast Blue staining method, and observed under Leica DMR

Abnormal expression of HSP gp96 associated with HBV replication in human hepatocellular carcinoma

BACKGROUND: Heat shock protein (HSP) gp96 is a member of the HSP90 family and presumably overexpresses as a result of stimulation by mutated or abnormal proteins. Its abnormal expression correlates with carcinogenesis, progression and prognosis of hepatocellular carcinoma (HCC). In this study, we investigated the pathological characteristics of liver gp96 expression and its relationship with hepatitis B virus (HBV) replication in HCC patients. METHODS: Tumor specimens were prospectively collected from 30 HCC patients undergoing liver resection. Total RNAs were extracted from HCC or their noncancerous tissues. The distribution of gp96 expression in hepatocytes was investigated by streptavidin peroxidase (S-P) immunohistochemistry and tissue HBV-DNA was detected by the in situ molecular hybridization technique. The association of gp96 expression with HBV replication, and the histopathological characteristics of HCC were analyzed. RESULTS: The gp96 was strongly expressed in HCC (73.3%, 22 of 30) and weakly (46.7%, 14 of 30) in non-cancerous tissues. The gp96 expression in HCC tissues was correlated with degree of tumor differentiation and tumor size, but not with tumor number (P>0.05). Immunohistochemical analysis showed that 17 of 19 HCC patients with HBVDNA -positive were strongly expressed for gp96, whereas only 5 of 11 patients with HBV-DNA-negative were positive for gp96. A significant difference was found between the two groups (89.5% vs. 45.5%, P<0.05). CONCLUSIONS: The abnormal expressions of HSP gp96 in HCC tissues are associated with HBV replication. This finding indicates that HBV infection plays an important role in the development of HCC.

The problem of fault estimation for a class of non-uniformly sampled-data systems is investigated from the time delay point of view in this ***,the output delay approach is employed to model the sampled-data system as a continuous-time one with time-varying delay ***,based on the analysis of the inapplicability of the adaptive fault diagnosis observer in such class of time-delay systems,a novel augmented fault estimation observer design method is proposed to guarantee the exponential convergence of the estimation ***,an extension to the case of time varying fault estimation for the noisy sampled-data systems is ***,simulation results of a flight control system are presented to demonstrate the effectiveness of the proposed method.

Antioxidant Effects of Dehydroepiandrosterone Are Related to Up-regulation of Thioredoxin in SH-SY5Y Cells

Neuroprotective effects of dehydroepiandrosterone（DHEA）have been shown to be associatedwith its antioxidant properties,but the mechanisms remain *** that the thioredoxin（Trx）system,an important cellular redox modulation system,changes under oxidative stress and could exertprotective effects,the relationship between the antioxidant effects of DHEA and the Trx system regulationwas *** MTT assay and morphological observation,the effects of DHEA in the model of H2O2-induced oxidative stress in SH-SY5Y cells were analyzed,then RT-PCR and Western blot assay were used todetect the alteration in mRNA and protein level of Trx,The results showed that a pre-treatment of DHEA（10-100nM）protected cells against the toxicity induced by H2O2in a dose-dependent manner,which could beconfirmed in morphological observation by phase-contract *** addition,Trx mRNA transcriptionwas inhibited by H2O2（300μM）,which could be reversed by the pre-administration of DHEA in variousconcentrations（0.1-100nM）.Western blot assay confirmed that protein level of Trx could be elevated by thepre-treatment of DHEA（10-100nM）with the exposure of *** together,these data suggest thatDHEA may be useful in treating age-related neurodegenerative diseases based on its up-regulating effects onan antioxidant and neuroprotective protein thioredoxin,a substrate in the Trx redox system.

Laser melting deposition process (LMD) is an advanced computer-aided laser additive manufacturing process (LAM). This process can produce net-shape components with fully dense microstructure and good properties based on a computer aided design (CAD) model by using a high power laser to melt the injected powders and re-solidify them in a layered way. It presents obvious superiority in direct manufacturing components with small-lot, repairing high-value components with good performance, and integrated fabricating new materials and composite structure. Deep understanding the melting process, alloying and reaction within the molten pool, solidification behavior, as well as formation and control of residual stress and defects during deposition are crucial to successful application.

Relationships between D1 protein, xanthophyll cycle and photodamage-resistant capacity in rice (Orysa sativa L.)

Relationships between D1 protein, xanthophyll cycle and subspecific difference of photodamage-resistant capacity have been studied in O. japonica rice varieties 02428 and 029 (photoinhibition-tolerance) and O. indica rice varieties 3037 and Palghar (photoinhibition-sensitivity) and their reciprocal cross F, hybrids after photoinhibitory treatment. It was shown that PS II photochemical efficiency (Fv /Fm) decreased, and xanthophyll cycle from violaxanthin (V), via anaxanthin (A), to zeaxanthin (Z) was enhanced and non-photochemical quenching (g/v) increased accordingly in SM-pretreated leaves of rice when the synthesis of D1 protein was inhibited, and that there was a decrease in qN and, as a result, more loss of D1 protein and a big decrease in Fv/Fm in DTT-pretreated leaves when xanthophyll cycle was inhibited. O. japonica subspecies had a higher maintaining capacity of D1 protein and a decrease of Fv /Fm in a more narrow range, and exhibited more resistance against photodamage, as compared with O.

Characterization of In-situ Titanium Matrix Composites Prepared by Laser Melting Deposition

Preliminary characterization of microstructure and mechanical properties of (TiB+TiC)/TC4 and TiC/Ti60 in-situ titanium matrix composites prepared by laser melting deposition is reported in this paper. The results indicate that in-situ reaction occurred during laser melting deposition of coaxially fed mixed powders from TC4 and B4C with formation of form TiB and TiC reinforcement. For TiC/Ti60 composites, there are some un-melted TiC particles and re-solidified TiC particles appeared as discontinuous chain-like morphology. Reinforcements of TiB and TiC with fraction about 25 vol% were formed with feeding 5 wt% B4C. The morphology of TiB tended to be needle-like and prismatic, while TiC appeared as granular. Small amount of un-reacted B4C with reduced size remained within the composites. A thin skull of reaction product formed around the un-reacted B4C weakened its interface bonding with the titanium alloy matrix, thus resulting in less outstanding properties of the composites. Under 600 ℃, the ultimate tensile strength of the TiCP (5wt%)/Ti60 composites was 60 MPa higher than that of Ti60 alloy, following with decreased elongation.
