Screening Test for Antibiotics in Medicinal Plants(STAMP):Using Powdered Plant Materials Instead of Extracts

Plants are a rich source of antibiotics, but screening all the existing plant species for biological activity using current methods can be time and resource consuming. The present study is to investigate whether powdered plant materials would perform as well as plant extracts in the screening of plants with antimicrobial activity. In the new method proposed (STAMP), we compared in vitro antimicrobial activity of powdered plant materials from 12 species against bacteria and fungi. We confirmed these results with their corresponding aqueous (wet) and hydro-alcoholic extracts and one species testing the antimicrobial activity of two isolated compounds. Compared with hydro-alcoholic extracts, screening using the powdered plant materials correctly identified the majority of the species with antimicrobial activity against Candida albicans (sensitivity 91%, specificity 86%), C. parapsilosis (sensitivity 100%, specificity 67%), and Staphylococcus aureus (sensitivity 64%, specificity 86%). For bacteria, the method performed better in a pH of 9.0. The antimicrobial activity of two compounds isolated from one species (maytenin and netzahualcoyone) confirmed the results. In conclusion, the use of powdered plant materials for screening plants with antimicrobial properties is a cheap, widely available, technically easy, time sparing, reproducible, and sensitive method and can significantly shorten the time and money spent during drug development.

AIM: To investigate the expression profiles of hsa-mi R-29 c and hsa-mi R-135 b in gastric mucosal samples and their values as gastric carcinogenesis biomarkers. METHODS: The expression levels of hsa-mi R-29 c and hsa-mi R-135 b in normal gastric mucosa, non-atrophic chronic gastritis, intestinal metaplasia and intestinaltype gastric adenocarcinoma were analysed using quantitative real-time PCR. The difference between hsa-mi R-29 c and hsa-mi R-135 b expression profiles in the grouped samples was evaluated by ANOVA and Student's t-test tests. The results were adjusted for multiple testing by using Bonferroni's correction. P values ≤ 0.05 were considered statistically significant. To evaluate hsa-mi R-29 c and hsa-mi R-135 b expressions as potential biomarkers of gastric carcinogenesis, we performed a receiver operating characteristic curve analysis and the derived area under the curve, and a Categorical Principal Components analysis. In silico identification of the genetic targets of hsa-mi R-29 c and hsa-mi R-135 b was performed using different prediction tools, in order to identify possible genes involved in gastric ***: The expression levels of hsa-mi R-29 c were higher in normal gastric mucosal samples, and decreased progressively in non-atrophic chronic gastritis samples, intestinal metaplasia samples and intestinal-type gastric adenocarcinoma samples. The expression of hsa-mi R-29 c in the gastric lesions showed that non-atrophic gastritis have an intermediate profile to gastric normal mucosa and intestinal-type gastric adenocarcinoma, and that intestinal metaplasia samples presented an expression pattern similar to that in intestinal-type gastric adenocarcinoma. AIM: To evaluate in bile duct ligated rats whether there were progressive alterations of renal function without changes in histopathology. METHODS: Male Wistar rats were submitted to sham-surgery or bile duct ligation (BDL) and divided according to the post-procedure time (2, 4 and 6-wk). To determine renal function parameters, rats wereplaced in metabolic cages and, at the end of the experiment, blood and urine samples were obtained. Histology and hydroxyproline content were analyzed in liver and renal tissue. RESULTS: Rats with 2 wk of BDL increased free water clearance (P = 0.02), reduced urinary osmolality (P = 0.03) and serum creatinine (P = 0.01) in comparison to the sham group. In contrast, rats at 6 wk of BDL showed features of HRS, including signif icant increase in serum creatinine and reductions in creatinine clearance, water excretion and urinary sodium concentration. Rats with 4 wk of BDL exhibited an intermediate stage of renal dysfunction. Progressive hepatic f ibrosis according to post-procedure time was confirmed by histology. The increased levels of liver hydroxyproline contrasted with the absence of structural changes in the kidney, as assessed by histology and unchanged hydroxyproline content in renal tissue. CONCLUSION: Our data show that BDL produced progressive renal dysfunction without structural changes in the kidney, characterizing HRS. The present model will be useful to understand the pathophysiology of HRS.

Man-made environments such as tropical hydroelectric reservoirs alter the preexisting carbon (C) cycle and remove C from circulation through burial in sediments. Carbon burial (CB) was measured using the silica-tracer method during four field surveys in the less than six-year-old Belo Monte tropical reservoir. Fresh C sedimentation was also measured. Belo Monte’s CB median rate 276 (n = 84;min 0;max 352,625 mg C·m-2·d-1) is within the range (230 to 436 mg C·m-2·d-1) of CB rates measured further downstream at the Xingu Ria and higher than the averaged over 50 years oceanic rate 244 mg C·m-2·d

通过对12个不同剪枝后废弃物堆肥(PWC)样品理化性质分析,评价它们是否适合作为观赏植物的生长基。样品定期地采集于相同的堆肥处,堆肥时间大于18个月,考察季节变化对堆肥的影响。除了剪枝后废弃物堆肥外,加拿大泥炭藓和商业生长基(CGM)被用作参考材料。PWC堆肥的性质如下:pH值在8以上,显著高于泥炭和商业生长基的pH值;所有堆肥样品都显示出丰富的有机质和相当高的阳离子交换量。C/N比值在22～48之间,显著高于15～20最优范围。虽然对堆肥进行抽样时处于不同季节,但其化学性质并无显著差异,仅持水量随季节有微小变化。与加拿大泥炭和商业生长基相比,修剪下来的植物残枝可以作为盆栽观赏植物生长基的组成部分。

Prognostic implication of the coronary microvascular dysfunction in patients with isolated left bundle branch block

The present study aims to determine the influence of microvascular dysfunction (MVD) in the prognosis of patients presenting isolated left bundle branch block (LBBB). Methods: We studied 30 patients (pts), 22 males, 8 females, mean age 57 ± 4 years, with isolated LBBB, with a mean follow up of 48 ± 6 months. The control group consisted of 20 healthy individuals, 12 males, mean age 52 ± 10 years. Both groups were screened for cardiovascular risk factors (RF);they also had an echocardiogram and Coronary CT Scan, ruling out both structural heart disease and obstructive lesions of the epicardial coronary arteries. A myocardial perfusion study was then performed, with two groups emerging according to these results: Group A, 8 pts (26%), with reversible perfusion defects, in which the diagnosis of MVD was suspected, and Group B, 22 pts (74%), with either normal perfusion or minor septal/apical reversible defects (related to LBBB). All Group A pts, and 9 of the Group B pts, underwent coronary arteriography, with intracoronary acetylcholine and nitroglycerine infusion, thus evaluating vasomotor response as endothelium dependent (acetylcholine) or endothelium independent (nitroglycerine). During follow up, we reviewed functional class, 12 lead ECG and echocardiogram on a yearly basis. Results: All Group A patients had an abnormal acetylcholine response;only three of them had abnormal response to nitroglycerine infusion, suggesting endothelium dependent MVD. Of those in Group B, only one patient had abnormal acetylcholine response. At the end of the follow up period, 3 pts (37%) in Group A, showed functional class decrease vs 5 pts (22%) of those in Group B. In Group A, a significant increase of End Diastolic Left Ventricle Diameter (EDLVD) was found (51.6 ± 3.6 vs 59.3 ± 6.8 mm;p < 0.05) with significant decrease in LVEF (62 ± 4.8 vs 46% ± 3.7%, p variation. Impairment of apoptosis promotes abnormal cellular proliferation and accumulation of genetic alterations. Identifying bioactive compounds extracted from seaweeds that induce apoptosis in cancer cells may be explored as new agents for cancer chemoprevention and/or chemotherapy. The present study aimed to determine the chemical composition and biological activity of the chloroform crude extract and fractions from Antarctic seaweed Desmarestia anceps. The chloroform extract was obtained by three consecutive macerations and the fractionation by vacuum liquid chromatography. The chemical characterization of the extract and fractions was performed through Gas Chromatography. The cytotoxicity of the crude extract and fractions was evaluated by the MTT assay. Cell death and cell cycle evaluation after 24 hours of exposure to chloroform extract were performed by flow cytometry. A total of 48 compounds were identified. The results indicate that chloroform extract and its fractions presented cytotoxic activity against HCT 116 cell line in a dose dependent-manner. Proapoptotic events were observed after chloroform extract exposition, which promoted an increase of multinucleated cells and reduced cell viability. This study was the first to explore cytotoxic potential of seaweed D. anceps fractions against HCT colorectal cancer cell line, suggesting that these macroalgae may be a promising candidate against anticancer activity.

We report the case of a Nigerian adolescent recently arrived to Spain, who presented at the emergency room with severe respiratory distress. She had been previously diagnosed of HIV-1 discontinuing antiretroviral therapy, what was hidden by the family. This case illustrates the difficulties in management and stigma in HIV-infected adolescents, particularly immigrants and the need to collect all the information available before starting antiretroviral therapy.

Fluorescence of Chlorophyll a for Discovering Inhibitors of Photosynthesis in Plant Extracts

Aims: Twelve crude extracts were prepared from different parts of Pluchea sagittalis, Cecropia palmata and Brachiaria brizantha, and their effect on the light reaction of photosynthesis. Study Design: Use of fluorescence of ChL a (Chlorophyll a) to detect inhibitors of photosynthesis in plant extracts. Place and Duration of Study: Instituto de Ciências Ambientais, Químicas e Farmacêuticas, Departamento de Ciências Exatas e da Terra, Universidade Federal de Sao Paulo (UNIFESP), between July 2015 and January 2016. Methodology: The samples had their effect on the light reaction of photosynthesis studied by Chlorophyll a (Chl a) fluorescence transient by the use of chloroplasts isolated from spinach leaves. As negative controls we used the same amounts of DMSO that have been employed to solubilize the samples, and as positive control we have used 10 μM of the commercial herbicide DCMU (Diuron). Results: The extracts inhibited the photosynthesis, and their inhibition sites were located in the range of electron flow from OEC complex and between P680 to QA of PS II, and inhibited the photosystem II (PS II) by inducing the appearance of the transient bands K and J. Conclusion: Our results suggest an indicative that the photochemical apparatus probably is failing at the donor and acceptor sides of PSII, causing damage to electron transport during the photosynthesis phenomenon.

【目的】研究中国咖啡主栽区咖啡种质资源圃中由专性寄生病原真菌咖啡驼孢锈菌(Hemileia vastatrix *** Br.)引起的咖啡黄锈病(coffee leaf rust,CLR)致病类型,对保护中国咖啡种质资源以及指导主栽区抗性品种的布局和选育具有重要意义。【方法】收集分布在中国咖啡主栽区4个具有代表性的咖啡种质资源圃CLR病样,在超感病基因型寄主上进行扩繁及单孢分离,在特定基因型寄主上对咖啡驼孢锈菌的毒力基因进行鉴定,并推导出相应的致病类型。【结果】57份样品中,有42份样品(占73.68%)被成功扩繁,有27份样品(占64.29%)被保存。根据样品信息录入23个CIFC条目,共鉴定出3种致病类型:XXXⅦ(v_(2,5,6,7,9)),占86.96%,为优势致病类型;New race(v_(2,5,6,7)),占8.69%,为在中国种质资源圃CLR调查的首次报道;XXXIV(v_(2,5,7,9)),占4.35%。【结论】中国咖啡主栽区咖啡驼孢锈菌群体遗传结构正在发生改变,原有致病类型已渗入位于主栽区内的种质资源圃,并在基因型相近的寄主上加速进化;新的致病类型在主栽区内的咖啡基地和种质资源圃之间随机流动,并对当前主栽品种的抗性造成威胁。研究结果将为中国咖啡种质资源圃的科学管理提供重要依据,为CLR防控提供理论支持。
