Conserved Curvature of RNA Polymerase Ⅰ Core Promoter beyond rRNA Genes: The Case of the Tritryps

In trypanosomatids, the RNA polymerase I （RNAPI）-dependent promoters controlling the ribosomal RNA （rRNA） genes have been well identified. Although the RNAPI transcription machinery recognizes the DNA conformation instead of the DNA sequence of promoters, no con- formational study has been reported for these promoters. Here we present the in silico analysis of the intrinsic DNA curvature of the rRNA gene core promoters in Trypanosoma brucei, Trypanosoma cruzi, and Leiskmania major. We found that, in spite of the absence of sequence conservation, these promoters hold conformational properties similar to other eukaryotic rRNA promoters. Our results also indicated that the intrinsic DNA curvature pattern is conserved within the Leishmania genus and also among strains of T. cruzi and T. brucei. Furthermore, we analyzed the impact of point mutations on the intrinsic curvature and their impact on the promoter activity. Furthermore, we found that the core promoters of protein-coding genes transcribed by RNAPI in T. brucei show the same conserved conformational characteristics. Overall, our results indicate that DNA intrinsic curvature of the rRNA gene core promoters is conserved in these ancient eukaryotes and such con- served curvature might be a requirement of RNAPI machinery for transcription of not only rRNA genes but also protein-coding genes.

Effect of arsenic on tolerance mechanisms of two plant growth-promoting bacteria used as biological inoculants

bacterial ability to colonize the rhizosphere of plants in arsenic（As） contaminated soils is highly important for symbiotic and free-living plant growth-promoting rhizobacteria（PGPR）used as inoculants, since they can contribute to enhance plant As tolerance and limit metalloid uptake by plants. The aim of this work was to study the effect of As on growth,exopolysaccharide（EPS） production, biofilm formation and motility of two strains used as soybean inoculants, bradyrhizobium japonicum E109 and Azospirillum brasilense Az39. The metabolism of arsenate（As（V）） and arsenite（As（III）） and their removal and/or possible accumulation were also evaluated. The behavior of both bacteria under As treatment was compared and discussed in relation to their potential for colonizing plant rhizosphere with high content of the metalloid. b. japonicum E109 growth was reduced with As（III）concentration from 10 μM while A. brasilense Az39 showed a reduction of growth with As（III） from 500 μM. EPS and biofilm production increased significantly under 25 μM As（III）for both strains. Moreover, this was more notorious for Azospirillum under 500 μM As（III）,where motility was seriously affected. both bacterial strains showed a similar ability to reduce As（V）. However, Azospirillum was able to oxidize more As（III）（around 53%） than bradyrhizobium（17%）. In addition, both strains accumulated As in cell biomass. The behavior of Azospirillum under As treatments suggests that this strain would be able to colonize efficiently As contaminated soils. In this way, inoculation with A. brasilense Az39 would positively contribute to promoting growth of different plant species under As treatment.

Synthesis of activated carbons from black sapote seeds,characterization and application in the elimination of heavy metals and textile dyes

Many different techniques may be used to remove industrial pollutants from wastewater. Adsorption using activated carbon has been reported to be an effective method. This work proposes the use of a vegetable residue(black sapote seeds) as a raw material for its synthesis. These carbons were chemically activated using phosphoric acid and carbonized at 673 and 873 K. Adsorption isotherms were constructed for the textile dyes on the carbons, and this data was treated using Langmuir’s equation to quantitatively describe the adsorption process. The synthesized carbons were characterized using FTIR, EA, SEM, Nitrogen adsorption(specific surface areas of 879 and 652 m2·g-1), and their points of zero charge(2.1 and 2.3). It was possible to adsorb both heavy metals and textile dyes present in aqueous solutions and wastewaters using these activated carbons. Heavy metals were adsorbed almost completely by both carbons. Cationic dyes where adsorbed(58–59.8 mg·g-1) in greater amounts compared to anionic dyes(10–58.8 mg·g-1). The amount of anionic dyes adsorbed increased almost 30% by changing the pH of the solutions. One of the carbons was thermally regenerated on three occasions without losing its adsorption capacity and it was proved in a flow system.

Influence of surface roughness on the color of dental-resin composites

This study deals with the influence of surface roughness on the color of resin *** resin composites (microfilled,hybrid,and microhybrid) were each polished with 500-grit,1 200-grit,2 000-grit,and 4 000-grit SiC *** roughness parameter (Ra) was measured using a Plμ confocal microscope,and field-emission scanning electron microscope (Fe-SEM) images were used to investigate filler *** was measured using a spectroradiometer and a D65 standard illuminant (geometry diffuse/0° specular component excluded (SCE) mode).Surface roughness decreased with grit number and was not influenced by filler size or size distribution.A significant influence of Ra on lightness (L*) was *** increased with decreases in roughness,except for specimens that underwent polishing procedure 4 (PP4;500-grit,1 200-grit,2 000-grit,and 4 000-grit SiC papers consecutively).Generally,it was found that surface roughness influenced the color of resin *** composites that underwent PP1 (500-grit SiC paper) exhibited significant differences in chroma (C*),hue (h°),and lightness (L*) compared to composites that underwent PP3 (500-grit,1 200-grit,and 2 000-grit SiC papers consecutively) and *** difference (E*) between the polishing procedures was within acceptability thresholds in dentistry.
