Successful double fertilization and subsequent seed development in flowering plants requires the delivery of two sperm cells, transported by a pollen tube, into the em.ryo sac of an ovule. The em.ryo sac cells tightly control synergid cell death, and as a result the polyspermy block. Arabinogalactan proteins are highly glycosylated proteins thought to be involved in several steps of the reproductive process. We show that JAGGEr, Arabinogalactan Protein 4, is an important molecule necessary to prevent the growth of multiple pollen tubes into one em.ryo sac in Arabidopsis thaliana. In jagger, an AGP4 knockout mutant, the pistils show impaired pollen tube blockage as a consequence of the survival of the persistent synergid. JAGGEr seems to be involved in the signaling pathway that leads to a blockage of pollen tube attraction. Our results shed light on the mechanism.responsible for preventing polyspermy in Arabidopsis and for safe- guarding successful fertilization of all ovules in one pistil, ensuring seed set and the next generation.

Purpose: The prim.ry aim of this paper is to design, create, and im.rove functional and artificial Bruch’s mem.ranes (Bm) using bioengineering techniques, which can be applied in the treatment of maculopathies by supporting the growth and maintenance of retinal pigment epithelium (rPE) cells, thereby potentially enabling subretinal implantation in patients. methods: We fabricated by electrospinning ultrathin 3D nanofibrous mem.ranes from Polycaprolactone (PCL), and different concentrations of gelatin (5%, 15% and 30%). ArPE-19 cells were seeded onto these artificial mem.ranes. mTT assays were performed in order to evaluate ArPE-19 cell viability and cytotoxicity. IF assays were realized to observe the ArPE-19 cells onto each mem.rane. Ultrastructure of the modified Bruch’s mem.rane and ArPE-19 m.rphology after 25 days of culture were studied with transmission and scanning electron microscopy. To evaluate expression changes in m.rkers of ArPE-19 (rPE65 and ZO-1) qrT-PCr assays were realized. Data from three independent experiments were pooled and expressed as the mean SD. A confidence level of P 0.05 was considered to be statistically significant. results ArPE-19 cells grew on PCL/Gelatin mem.ranes mainly in PCL/30% gelatin combination, which had not cytotoxic effect. rPE65/cytokeratin-18/ and actin-positive ArPE-19 cells formed a correctly orientated monolayer of polygonal cells with m.rphological polarity. The apical cell surfaces exhibited abundant protruding microfolds. m.reover, zones of polygonal border look as if ArPE-19 cells fused among, suggesting the presence of tight junctions. The expression of rPE65 and ZO-1 was unchanged. Conclusions: PCL/30% Gelatine mem.ranes may imitate the natural Bm to such extent that they support rPE-cells and exhibited rPE-like m.rphology. Nitrification inhibitors are widely used in agriculture to mitigate nitrous oxide(N_(2)O)emission and increase crop ***,no concrete information on their mitigation of N_(2)O emission is available under soil and environmental conditions as in Pakistan.A field experiment was established using a silt clay loam soil from Peshawar,Pakistan,to study the effect of urea applied in combination with a nitrification inhibitor,nitrapyrin(2-chloro-6-tri-chloromethyl pyridine),and/or a plant growth regulator,gibberellic acid(GA_3),on N_(2)O emission and the nitrogen(N)uptake efficiency of *** experimental design was a randomized complete block with five treatments in four replicates:control with no N(CK),urea(200 kg N ha^(-1))alone,urea in combination with nitrapyrin(700 g ha^(-1)),urea in combination with GA_3(60 g ha^(-1)),and urea in combination with nitrapyrin and GA_*** N_(2)O emission,yield,N response efficiency,and total N uptake were measured during the experimental *** treatment with urea and nitrapyrin reduced total N_(2)O emission by 39%–43%and decreased yield-scaled N_(2)O emission by 47%–52%,relative to the treatment with urea *** maize plant biomass,grain yield,and total N uptake increased significantly by 23%,17%,and 15%,respectively,in the treatment with urea and nitrapyrin,relative to the treatment with urea alone,which was possibly due to N saving,lower N loss,and increased N uptake in the form of ammonium;they were further enhanced in the treatment with urea,nitrapyrin,and GA_3 by 27%,36%,and 25%,respectively,probably because of the stimulating effect of GA_3 on plant growth and development and the reduction in biotic and abiotic *** results suggest that applying urea in combination with nitrapyrin and GA_3 has the potential to mitigate N_(2)O emission,im.rove N response efficiency,and increase maize yield.

Background:regular physical activity(PA) is an important behavior in im.roving sleep ***,the short-term effects of PA on sleep are still *** study aimed to verify the effect of different intensities of PA practiced in different periods of the day on the subsequent sleep night in a population-based cohort of young ***::Prospective analyses were conducted for PA performed during the day and its effect on the following sleep night using data from the22-year follow-up of the 1993 Pelotas Birth Cohort in Brazil(mean age of participants-22.6 years).Wrist-worn accelerometry was usedto measure both PA and sleep *** intensity,we analyzed the sleep effect of light PA(LPA),moderate PA,and vigorous PA,stratified by *** variables were sleep time window(STW;the difference between sleep onset and sleep end),total sleep time(TST;the sum of minutes classified as sleep in STW),and sleep percent(SP;SP=(TST/STW);expressed in percentage).We performed generalized estimating equations using Stata ***::The means of STW,TST,and SP were 443.6 min/day,371.1 min/day,and 84%,*** spent in moderate PA and vigorous PA in the m.rning and afternoon was not associated with sleep *** men,10 min/day of m.rning LPA increased TST by2.56 min/*** women,10 min/day of m.rning LPA increased SP by 0.15 percentage *** LPA also increased SP by 0.09 percentage points for *** PA seems to have an inverse effect on sleep variables for any intensity and both ***::The effect of PA on sleep health is intrinsically related to the period of the day in which it is *** effect magnitude is different between *** better sleep health,it is preferable that PA be performed during the day.

Li2SnO3 branched nano- and microstructures with intense and broadband white-light emission

Exploiting the synergy between microstructure, m.rphology and dimensions by suitable nanomaterial engineeri ng, can effectively upgrade the physical properties and material performances. Li2SnO3 elongated nano- and microstructures in form of belts, wires, rods and branched structures have been fabricated by a vapor-solid method at temperatures ranging from 700 to 900℃ using metallic Sn and Li2CO3 as precursors. The achievement of these new m.rphologies can face challenging applications for Li2SnO3, not only in the field of energy storage, but also as building blocks in optoelectronic devices. The micro? and nanostructures grown at 700 and 800℃ correspond to monoclinic Li2SnO3, while at 900℃ complex Li2S nO/SnO2 core-shell microstructures are grow n, as con firmed by X-ray diffractio n and rama n spectroscopy. Transmission electron microscopy reveals structural disorder related to stacking faults in some of the branched structures, which is associated with the prese nee of the low-temperature phase of L12S nO3. The luminescent resp onse of these structures is domi nated by in tense emissi ons at 2, 2.5 and 3 eV, almost completely covering the whole range of the visible light spectrum. As a result, white-light emission is obtained without the need of phosphors or complex quantum well heterostructures. Enhanced functionality in applications such as in light-emitting devices could be exploited based on the high luminescenee intensity observed in some of the analysed Li2SnO3 structures.
