Purpose Among the several methods to measure ^(222)Rn in water,the liquid scintillation counting(LSC)technique is one of the most sensitive and widely used when analysing non-saline waters by liquid extraction,providing excellent accuracy,precision and low-level detection *** using the liquid scintillation counter Tri-Carb 3170 TR/SL(Perkin-Elmer)it is not possible to make an automatic determination of the alpha/beta discrimination parameter,being necessary to evaluate the pulse decay discriminator(PDD)value manually to the ^(222)Rn *** this work,we describe the steps to perform the calibration of that type of *** The method consists in the preparation of standard samples from a ^(226)Ra certified standard solution and the adjustment of the PDD value by hand until the percentage of alpha events reaches a plateau of a near-constant fraction of total *** process consists in taking several measures between PDD 80 and 200 with 15 min counting time *** the plateau,the alpha/beta ratio was used to determine the plateau final range as well as the optimum PDD *** that,the calibration standards and blanks were measured and the region of interest(ROI)was established by finding the maximum value for the figure of merit(FOm)in the upper and lower *** The optimal PDD value of 148 was established by the alpha/beta ratio,and after that,it was possible to measure ^(226)Ra standards to perform the *** efficiency of(9.758±0.035)×1^(0-1) was *** routine measurements,a detection limit of about 0.50 Bq L^(-1) is achieved for a 120 min counting *** evaluate the uncertainty budget,the uncertainties of the net count rate,the efficiency,the sample amount and the decay correction factor were *** A method was developed for the calibration of LSC Tri-Carb 3170 TR/SL with the optimization of the PDD using a ^(226)Ra standard *** acquisition to obtain the plot of the variation

Objective To examine the relationship between Tpeak- Tend interval （Tpe） and Tpe/QT ratio with occurrence of ventricular premature beats （VPBs） and left ventricular remodeling in hypertension. methods A total of 52 patients with mild to moderate essential hypertension were included, undergoing echocardiography and 24-hours Holter monitoring. Ventricular remodeling was assessed by left ventricular mass index （LVmI） using the Devereux formula and diastolic fimction by transmitral E and A wave velocities and E/A ratio. Tpe was measured in the precordial leads. The end of the T wave was set by the method of the tangent to the steepest descending slope of the T wave. Results Tpe and Tpe/QT in leads V2 （r = 0.33, P = 0.01; r = 0.27, P = 0.04 respectively） and V3 （r = 0.40, P = 0.002; r = 0.40, P = 0.003, respectively） correlated significantly with LVmI. A significant inverse relationship was observed between E/A ratio and QT （r = -0.33, P = 0.01）, Tpe in V3 （r = -0.39, P = 0.003） and Tpe/QT in V3 （r = -0.31, P = 0.02）. Tpe in V3, V5, mean Tpe and maximum Tpe with cut-offvalues of 60 ms, 59 ms, 62 ms and 71 ms, respectively, associated with the occurrence of ventricular premature beats. Conclusions The repolarization parameters Tpe interval and Tpe/QT ratio correlate with LVmI and indices of left ventricular diastolic function and show better predictive values than traditional parameters such as QT interval and QT dispersion. Lead V3 is the best lead for measuring Tpe and Tpe/QT. These ECG indices can therefore be used in clinical practice to monitor LV remodeling and predict occurrence of VPBs.

Extensins （EXTs） are highly repetitive plant O-glycoproteins that require several post-translational modifi- cations （PTms） to become functional in plant cell walls. First, they are hydroxylated on contiguous proline residues; then they are O-glycosylated on hydroxyproline and serine. After secretion into the apoplast, O-glycosylated EXTs form a tridimensional network organized by inter- and intra-Tyr linkages. Recent studies have made significant progress in the identification of the enzymatic machinery required to process EXTs, which includes prolyl 4-hydroxylases, glycosyltransferases, papain-type cysteine endopeptidases, and peroxidases. EXTs are abundant in plant tissues and are particularly important in rapidly expanding root hairs and pollen tubes, which grow in a polar manner. Small changes in EXT PTms affect fastgrowing cells, although the molecular mechanisms underlying this regulation are unknown. In this review, we highlight recent advances in our understanding of EXT modifications throughout the secretory pathway, EXT assembly in cell walls, and possible sensing mechanisms involving the Catharanthus roseus cell surface sensor receptor-like kinases located at the interface between the apoplast and the cytoplasmic side of the plasma membrane.

Successful double fertilization and subsequent seed development in flowering plants requires the delivery of two sperm cells, transported by a pollen tube, into the embryo sac of an ovule. The embryo sac cells tightly control synergid cell death, and as a result the polyspermy block. Arabinogalactan proteins are highly glycosylated proteins thought to be involved in several steps of the reproductive process. We show that JAGGER, Arabinogalactan Protein 4, is an important molecule necessary to prevent the growth of multiple pollen tubes into one embryo sac in Arabidopsis thaliana. In jagger, an AGP4 knockout mutant, the pistils show impaired pollen tube blockage as a consequence of the survival of the persistent synergid. JAGGER seems to be involved in the signaling pathway that leads to a blockage of pollen tube attraction. Our results shed light on the mechanism responsible for preventing polyspermy in Arabidopsis and for safe- guarding successful fertilization of all ovules in one pistil, ensuring seed set and the next generation.
