Development of a standardized in vitro approach to evaluate microphysical,chemical,and toxicological properties of combustion-derived fine and ultrafine particles

Ultrafine particles represent a growing concern in the public health community but their precise role in many illnesses is still unknown. This lack of knowledge is related to the experimental difficulty in linking their biological effects to their multiple properties, which are important determinants of toxicity. Our aim is to propose an interdisciplinary approach to study fine(FP) and ultrafine(UFP) particles, generated in a controlled manner using a mini CAST(Combustion Aerosol Standard) soot generator used with two different operating conditions(CAST1 and CAST3). The chemical characterization was performed by an untargeted analysis using ultra-high resolution mass spectrometry. In conjunction with this approach, subsequent analysis by gas chromatography–mass spectrometry(GC–MS) was performed to identify polycyclic aromatic hydrocarbons(PAH). CAST1 enabled the generation of FP with a predominance of small PAH molecules, and CAST3 enabled the generation of UFP, which presented higher numbers of carbon atoms corresponding to larger PAH molecules. Healthy normal human bronchial epithelial(NHBE) cells differentiated at the air-liquid interface(ALI) were directly exposed to these freshly emitted FP and UFP. Expression of MUC5AC, FOXJ1, OCLN and ZOI as well as microscopic observation confirmed the ciliated pseudostratified epithelial phenotype. Study of the mass deposition efficiency revealed a difference between the two operating conditions, probably due to the morphological differences between the two categories of particles. We demonstrated that only NHBE cells exposed to CAST3 particles induced upregulation in the gene expression of IL-8 and NQO1. This approach offers new perspectives to study FP and UFP with stable and controlled properties.

Hydrocarbons in particulate samples from wildfire events in central Portugal in summer 2010

In summer 2010, twenty eight （14 PM=2.5 samples plus 14 samples PM2.5-10） smoke samples were collected during wildfires that occurred in central Portugal. A portable high-volume sampler was used to perform the sampling, on quartz fibre filters of coarse （PM=2.5-10） and fine （PM2.5） smoke samples. The carbonaceous content （elemental and organic carbon） of particulate matter was analysed by a thermal-optical technique. Subsequently, the particulate samples were solvent extracted and fractionated by vacuum flash chromatog- raphy into three different classes of organic compounds （aliphatics, polycyclic aromatic hydrocarbons （PAHs） and carbonyl compounds）. The organic speciation was performed by gas chromatography-mass spectrometry （GC-MS）. Emissions were dominated by the fine particles, which represented around 92% of the PM10. A clear predominance of carbonaceous constituents was observed, with organic to elemental carbon （OC/EC） ratios ranging between 1.69 and 245 in both size fractions. The isoprenoid ketone 6,10,14-trimethyl-2-pentadecanone, a tracer for secondary organic aerosol formation, was one of the dominant constituents in both fine and coarse particles. Retene was the most abundant compound in all samples. Good correlations were obtained between OC and both aliphatic and PAH compounds. Pyrogenic processes, thermal release of biogenic com- pounds and secondary processing accounted for 97% of the apportioned PM2.5 levels.

Aljustrel mines were classified as having high environmental hazard due to their large tailings volume and high metal concentrations in waters and *** assess acid mine drainage impacted systems whose environmental conditions change quickly,the use of biological indicators with short generation time such as diatoms is *** study combined geochemical and diatom data,whose results were highlighted in 3 groups：Group 1,with low p H（1.9–5.1）and high metal/metalloid（Al,As,Cd,Co,Cu,Fe,Mn,Ni,Pb,Zn;0.65–1032 mg/L）and SO4（405–39124 mg/L）*** acidophilic species,Pinnularia aljustrelica,was perfectly adapted to the adverse conditions;in contrast,teratological forms of Eunotia exigua were found,showing that metal toxicity affected this *** low availability of metals/metalloids in sediments of this group indicates that metals/metalloids of the exchangeable fractions had been solubilized,which in fact enables metal/metalloid diatom uptake and consequently the occurrence of teratologies;Group 2,with sites of near neutral p H（5.0–6.8）and intermediate metal/metalloid（0.002–6 mg/L）and SO4（302–2179 mg/L）concentrations;this enabled the existence of typical species of uncontaminated streams（Brachysira neglectissima,Achnanthidium minutissimum）;Group 3,with samples from unimpacted sites,showing low metal/metalloid（0–0.8 mg/L）and SO4（10–315 mg/L）concentrations,high pH（7.0–8.4）and Cl contents（10–2119 mg/L）and the presence of brackish to marine species（Entomoneis paludosa）.For similar conditions of acidity,differences in diversity,abundance and teratologies of diatoms can be explained by the levels of metals/metalloids.

Reactive oxygen species(ROS)production is a by-product of mitochondrial activity and is necessary for the acquisition of the capacitated state,a requirement for functional ***,an increase in oxidative stress,due to an abnormal production of ROS,has been shown to be related to loss of sperm function,highlighting the importance of an accurate detection of sperm ROS,given the specific nature of this *** this work,we tested a variety of commercially available fluorescent probes to detect ROS and reactive nitrogen species(RNS)in human sperm,to define their *** both flow cytometry(FC)and fluorescence microscopy(FM),we confirmed that MitoSOX™Red and dihydroethidium(DHE)detect superoxide anion(as determined using antimycin A as a positive control),while DAF-2A detects reactive nitrogen species(namely,nitric oxide).For the first time,we also report that RedoxSensor™Red CC-1,CellROX®Orange Reagent,and MitoPYl seem to be mostly sensitive to hydrogen peroxide,but not ***,mean fluorescence intensity(and not percentage of labeled cells)is the main parameter that can be reproducibly monitored using this type of methodology.

Changes in social emotion recognition following traumatic frontal lobe injury

Changes in social and emotional behaviour have been consistently observed in patients with traumatic brain injury. These changes are associated with emotion recognition deficits which represent one of the major barriers to a successful familiar and social reintegration. In the present study, 32 patients with traumatic brain injury, involving the frontal lobe, and 41 age- and education-matched healthy controls were analyzed. A Go/No-Go task was designed, where each participant had to recognize faces representing three social emotions （arrogance, guilt and jealousy）. Results suggested that ability to recognize two social emotions （arrogance and jealousy） was significantly reduced in patients with traumatic brain injury, indicating frontal lesion can reduce emotion recognition ability. In addition, the analysis of the results for hemispheric lesion location （right, left or bilateral） suggested the bilateral lesion sub-group showed a lower accuracy on all social emotions.

Depression is a common,recurrent mental disorder and one of the leading causes of disability and global burden of disease *** to 15%-40%of cases do not respond to diverse pharmacological treatments and,thus,can be defined as treatment-resistant depression(TRD).The development of biomarkers predictive of drug response could guide us towards personalized and earlier *** evidence points to the involvement of the glutamatergic system in the pathogenesis of ***,the N-methyl-D-aspartic acid receptor(NMDAR)andα-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid receptor(AMPAR),which are targeted by ketamine and esketamine,are proposed as promising pathways.A literature search was performed to identify studies on the genetics of the glutamatergic system in depression,focused on variables related to NMDARs and *** review highlights GRIN2B,which encodes the NR2B subunit of NMDAR,as a candidate gene in the pathogenesis of *** addition,several studies have associated genes encoding AMPAR subunits with symptomatic severity and suicidal *** genes encoding glutamatergic receptors could,therefore,be candidate genes for understanding the etiopathogenesis of TRD,as well as for understanding the pharmacodynamic mechanisms and response to ketamine and esketamine treatment.

XEN^(■) implant and trabeculectomy medium-term quality of life assessment and comparison of results

AIM: To evaluate and compare the quality of life of patients submitted to XEN~? implant or trabeculectomy and the relationship with potentially involved ***: A cross-sectional study of patients with advanced open-angle glaucoma who underwent implantation of XEN~?(group 1) and trabeculectomy(group 2) between October 2015 and February 2017. The studied variables were: age, gender, follow-up time, need of topical anti-hypertensive therapy, visual acuity and intraocular pressure(IOP). The quantification of the quality of life was attained through the Glaucoma Symptom Scale(GSS) questionnaire. RESULTS: Totally 34 eyes(34 patients) were included, 17 in each group. The mean GSS scores for group 1 were 42.6±6.8(median, 47; p25, 36.5; p75, 48.5) and for group 2 it was 41.6±7.0(median, 43; p25, 36.5; p75, 47.0; P=0.34). There was a strong negative correlation between the need for topical anti-hypertensive drugs and the GSS result in both groups(r=-0.88, Panalysis demonstrates the noninferiority of medium-term quality of life of one group in relation to the other(XEN~? implant and trabeculectomy). The number of topical anti-hypertensive drugs and IOP negatively influenced the quality of life.

A GIS Methodology for Planning Sustainable Renewable Energy Deployment in Portugal

A Geographical Information System (GIS) methodology was developed to?identify and characterize suitable areas for deploying renewable energy projects. The methodology enables to compute the sustainable renewable energy potential in an area under study and can be implemented for different spatial scales, ranging from local to national levels, while operating with different restriction layers. This GIS-based method has been successfully applied to wind energy deployment studies in Continental Portugal and other foreign countries?(e.g. Venezuela, Mozambique among others). Results from several development plans using this methodology enable to conclude it is an adequate tool for planning sustainable renewable energy deployment both for onshore and offshore regions.

Space-Use analysis in the Design of Neighborhood Walking Paths

SUA （space-use analysis） is about techniques that objectively describe environments and relate this description to specific problems of use. The purpose of this paper is to explore a space-use analytical tool--POE （post-occupation evaluation）--regarding a neighborhood accessibility condition. Based on a site-specific approach, the main research question is focused on how far neighborhood walking paths are compatible with different user needs, guaranteeing their safety and autonomy, as well as easiness of perception and use, thus providing access to local services and resources they need for participation in the day-to-day activities of community life. This paper is organized in three parts： The first one introduces the main concepts and explains how to ensure it in a practical application in urban design; The second part presents the practical approach, referring to the used methodology, the specific case-study and reached results; The third one establishes correlations between the two approaches and discusses the main findings.

Identification of the Sustainable Wind and Solar Potential in Urban Areas Using a GIS Methodology

In recent years the publication of new legislation in the renewables sector, namely the one related to microgeneration systems, has contributed to an increase in the installation of renewable energy systems in Portugal. Nevertheless, the investment in these systems is still high and the risk of considering sites with reduced resource constitutes a barrier to the development of this sub-sector. In the sequence of the previous work developed in the wind energy planning area, a methodology to identify the sustainable wind and solar potential in urban areas was developed, in order to reduce the pre-installation costs related to a less suitable site selection, and also to identify the real potential of the urban areas. In order to do so, a geographical information system was used and a set of tools was developed. The methodology was structured to enable its replication in other geographical locations and is sufficiently interactive to allow for the variation of the main input data. A case study in two urban environments is presented in this document to illustrate the developed methodology.
