Genetic variation at the p53 locus of two ecologically divergent Microtus pine voles:identification of molecular markers for species assignment

The Lusitanian(Microtus lusitanicus)and the Mediterranean(Microtus duodecimcostatus)pine voles are recently diverged sister species endemic of the Iberian Peninsula that can be identiﬁed with ecological and morphological characters,but in areas where the 2 species co-occur,species designation may be difﬁ*** discrimination between *** and *** has not been achieved yet possibly because of their estimated re-cent split and an evolutionary history that includes inter-species geneﬂ*** our previous observations on exons 5–7 of the p53 gene,here we analyze the potential use of the p53 genomic region as a discrimination marker of these species by extending our analyses to several kb upstream and downstream of the p53 gene and charac-terizing the degree of genetic differentiation in 7 markers within this ***,we fully sequenced the P53 protein of both *** observed:(i)generally high differentiation in this region;(ii)*** showed in general higher values of nucleotide and haplotype diversities;(iii)the concatenated phylogenetic tree separates the 2 species;(iv)the 2 P53 proteins only differ in 1 amino acid;(v)4 of the markers,2 in p53,one in Atp1b2,and another in Wrap53,contain species-speciﬁc genetic variation thus allowing a reliable discrimination between specimens from both species,irrespective of sampling location or introgression *** provide ad-ditional data on the putative role of p53 in the evolution of these species and present researchers with a fast and cost-effective resource for *** and *** identiﬁcation.

Obstructive sleep apnea syndrome(OSAS) is a widespread disorder, characterized by recurrent upper airway obstruction during sleep, mostly as a result of complete or partial pharyngeal obstruction. Due to the occurrence of frequent and regular hypoxic events, patients with OSAS are at increased risk of cardiovascular disease, stroke, metabolic disorders, occupational errors, motor vehicle accidents and even death. Thus, OSAS has severe consequences and represents a significant economic burden. However, some of the consequences, as well as their costs can be reduced with appropriate detection and treatment. In this context, the recent advances that were made in stem cell biology knowledge and stem cell- based technologies hold a great promise for various medical conditions, including respiratory diseases. However, the investigation of the role of stem cells in OSAS is still recent and rather limited, requiring further studies, both in animal models and humans. The goal of this review is to summarize the current state of knowledge regarding both lung resident as well as circulating stem/progenitor cells and discuss existing controversies in the field in order to identify future research directions for clinical applications in OSAS. Also, the paper highlights the requisite for inter-institutional, multi-disciplinary research collaborations in order to achieve breakthrough results in the field.

The use of remote sensing to detect the consequences of erosion in gypsiferous soils

Tillage practices on sloping ground often result in unsustainable soil losses impairing soil functions such as crop productivity,water and nutrients storage,and soil organic carbon(SOC)sequestration.A sloping olive grove(10%)was planted in shallow gypsiferous soils in *** was managed by minimum tillage;the most frequent management practice in central *** consequences of erosion were studied in soil samples(at 0-10,10-20,and 20-30 cm depths)by analyzing SOC,available water and gypsum content,and by detecting spectral signatures using an ASD FieldSpecPro(R)VIS/*** Brightness index(BI),Shape index(FI),and Normalized Difference Vegetation Index(NDVI)were derived from the ASD spectral signatures and from remote sensing(Sentinel-2 image)*** development of these young olive trees was estimated from the measured diameter of the trunks(17±18 cm diameter).In 20-30 cm of the soil,the carbon stock(38±18 Mg ha-1)as well as the available water content(12±6%)was scarce,affecting the productivity of the olive *** above-mentioned indices obtained from the laboratory samples and the pixels of the Sentinel-2 image were significantly(p<0.01)correlated,with a correlation coefficient of around *** BI was related to the gypsum content and the slope of the *** FI was related to the carbon and water *** NDVI derived from the satellite image identified the influence of soil degradation on the trees and the carbon *** spatialtemporal changes of the indices might help in tracking soil changes over time.

Pancreatic cancer is one of the most aggressive and lethal malignancies. Despite remarkable progress in understanding pancreatic carcinogenesis at the molecular level, as well as progress in new therapeutic approaches, pancreatic cancer remains a disease with a dismal prognosis. Among the mechanisms responsible for drug resistance, the most relevant are changes in individual genes or signaling pathways and the presence of highly resistant cancer stem cells(CSCs). In pancreatic cancer, CSCs represent 0.2%-0.8% of pancreatic cancer cells and are considered to be responsible for tumor growth, invasion, metastasis and recurrence. CSCs have been extensively studied as of late to identify specific surface markers to ensure reliable sorting and for signaling pathways identified to play a pivotal role in CSC self-renewal. Involvement of CSCs in pancreatic cancer pathogenesis has also highlighted these cells as the preferential targets for therapy. The present review is an update of the results in two main fields of research in pancreatic cancer, pathogenesis and therapy, focused on the narrow perspective of CSCs.

T follicular helper expansion and humoral-mediated rejection are independent of the HVEM/BTLA pathway

The molecular pathways contributing to humoral-mediated allograft rejection are poorly defined. In this study, we assessed the role of the herpesvirus entry mediator/B- and T-lymphocyte attenuator （HVEM/BTLA） signalling pathway in the context of antibody-mediated allograft rejection. An experimental setting was designed to elucidate whether the blockade of HVEM/BTLA interactions could modulate de novo induction of host antidonor-specific antibodies during the course of graft rejection. To test this hypothesis, fully allogeneic major histocompatibility complex-mismatched skin grafts were transplanted onto the right flank of recipient mice that were treated with isotype control, anti-CD40L or modulatory antibodies of the HVEM/BTLA signalling pathway. The frequencies of CD4 T follicular helper （Tfh） cells （B220-, CD4＋ CXCR5＋ PD-lhigh）, extrafollicular helper cells （B220-, CD4＋ CXCR5- PD-1＋ and PD-1-） and germinal centre （GC） B cells （B220＋Fas＋ GL7＋） were analysed by flow cytometry in draining and non-draining lymph nodes at day 10 post transplantation during the acute phase of graft rejection. The host antidonor isotype-specific humoral immune response was also assessed. Whereas blockade of the CD40/CD40L pathway was highly effective in preventing the allogeneic humoral immune response, antibody-mediated blockade of the HVEM/BTLA-interacting pathway affected neither the expansion of Tfh cells nor the expansion of GC B cells. Consequently, the course of the host antidonor antibody-mediated response proceeded normally, without detectable evidence of impaired development. In summary, these data indicate that HVEM/BTLA interactions are dispensable for the formation of de novo host antidonor isotype-specific antibodies in transplantation.

Esophageal atresia (EA) with or without tracheoesophageal fistula (TEF) is the most common congenital anomaly of the esophagus. The improvement of survival observed over the previous two decades is multifactorial and largely attributable to advances in neonatal intensive care, neonatal anesthesia, ventilatory and nutritional support, antibiotics, early surgical intervention, surgical materials and techniques. Indeed, mortality is currently limited to those cases with coexisting severe life-threatening anomalies. The diagnosis of EA is most commonly made during the first 24 h of life but may occur either antenatally or may be delayed. The primary surgical correction for EA and TEF is the best option in the absence of severe malformations. There is no ideal replacement for the esophagus and the optimal surgical treatment for patients with long-gap EA is still contro-versial. The primary complications during the postoperative period are leak and stenosis of the anastomosis, gastro-esophageal reflux, esophageal dysmotility, fistula recurrence, respiratory disorders and deformities of the thoracic wall. Data regarding long-term outcomes and follow-ups are limited for patients following EA/TEF repair. The determination of the risk factors for the complicated evolution following EA/TEF repair may positively impact long-term prognoses. Much remains to be studied regarding this condition. This manuscript provides a literature review of the current knowledge regarding EA.

Chemical proﬁles of non-invasive biological material,such as feces,have great potential to study elusive animals or those with low population ***,we use a metabolomic approach to evaluate Neotropical mustelids as a biological model to describe the diversity of the metabolites present in fecal samples,as well as to evaluate the potential of chemical proﬁles for taxonomic *** collected fecal samples from captive individuals of 5 species of mustelids occurring in Brazil and analyzed them by liquid chromatography coupled to high-resolution mass *** 200 compounds have been annotated;“bile acids,alcohols and derivatives”was the most expressive class in the metabolome of all the *** successfully discriminated 3 taxonomic groups:1—tayra(Eira barbara);2—otters(Lontra longicaudis and Pteronura brasiliensis;1);and 3—grisons(Galictis vittata and Galictis cuja).Several compounds seemed to be associated with food intake and the digestive process,while others were found for theﬁrst time in Neotropical *** concluded that mustelids show high metabolome diversity and that species-speciﬁc identiﬁcation through metabolomic proﬁles is possible,thus contributing to the development and implementation of additional non-invasive approaches in the study of mustelids.

Optimization of InAs/GaAs quantum-dot structures and application to 1.3-μm mode-locked laser diodes

The self-assembled growth of InAs/GaAs quantum dots by molecular beam epitaxy is conducted by optimizing several growth parameters, using a one-step interruption method after island formation. The dependence of photoluminescence on areal quantum-dot density is systematically investigated as a function of InAs deposition, growth temperature and arsenic pressure. The results of this investigation along with time-resolved photoluminescence measurements show that the com- bination of a growth temperature of 490℃, with a deposition rate of 0.02 ML/s, under an arsenic pressure of 1×10^-6 Torr （1 Torr = 1.33322×10^2 Pa）, provides the best compromise between high density and the photoluminescence of quantum dot structure, with a radiative lifetime of 780 ps. The applicability of this 5-layer quantum dot structure to high-repetition-rate pulsed lasers is demonstrated with the fabrication and characterization of a monolithic InAs/GaAs quantum-dot passively mode-locked laser operating at nearly 1300 nm. Picosecond pulse generation is achieved from a two-section laser, with a 19.7-GHz repetition rate.

A data fusion-based framework to integrate multi-source VGI in an authoritative land use database

Updating an authoritative Land Use and Land Cover(LULC)database requires many *** geographic information(VGI)involves citizens in the collection of data about their spatial *** is a growing interest in using existing VGI to update authoritative *** paper presents a framework aimed at integrating multi-source VGI based on a data fusion technique,in order to update an authoritative land use *** VGI data source is considered to be an independent source of information,which is fused together using Dempster-Shafer Theory(DST).The framework is tested in the updating of the authoritative land use data produced by the French National Mapping *** data sets were collected from several in-situ and remote campaigns run between 2018 and 2020 by contributors with varying *** data fusion approach achieved an overall accuracy of 85.6%for the 144 features having at least two contributions when the confidence threshold was set to *** the heterogeneity and limited amount of VGI used,the results are promising,with 99%of the LU polygons updated or *** results show the potential of using multi-source VGI to update or enrich authoritative LU data and potentially LULC data more generally。
