Each Epiphyte on Its Branch: A Comparative Study between Different Phorophytes

Epiphytes are plants that use the mechanical support provided by a host tree, called phorophytes, without emitting haustorial structures. Constituting an important component of the flora, playing an important role in the maintenance of ecosystems, such as water and nutrient cycling, in addition to providing resources, sometimes unique, such as food and shelter, for the canopy fauna. However, the epiphytic flora may show high beta diversity, even when compared with different phorophytes, possibly as a response to the dendromorphological characteristics of the phorophyte. Thus, this work aims to: 1) compare whether the species richness of epiphytes differs in trees with different morphological characteristics, and 2) compare species composition among three tree species. For this, 10 individuals of three tree species were selected: Guaria guidonia, Ficus sp., and Roystonea oleracea. In each individual, the number of species (richness) of vascular epiphytes was recorded and obtained from the species composition. To test differences in richness values between the three trees, we used analysis of variance (ANOVA) followed by Tukey’s test. To understand whether the epiphyte community formed separate groups according to the tree species used, we used the available non-metric multidimensional scaling method (NMDS). Thirty-four species of epiphytes were found in the 30 phorophytes surveyed. The three tree species showed significantly different richness, with the highest found in Guarea guidonea and the lowest in Roystonea oleraceae. The NMDS showed that epiphytes group the three tree species into distinct groups, although Ficus sp individuals present a transitional community between the two other species. Thus, the results suggest that the differences in the richness and composition of the epiphytic flora of the different phorophytes are influenced by their distinct morphological characteristics.

The use of natural enemies represents an important strategy in programs of Integrated Pest Management (IPM). Asopinae predators are reared with alternative prey, but supplementary plant may enhance the fecundity and longevity of their females. The objective of this research was to evaluate the development and reproduction of the predator Brontocoris tabidus Signoret, 1852 (Heteroptera: Pentatomidae) fed with Tenebrio molitor L., 1785 (Coleoptera: Tenebrionidae) pupae in a Eucalyptus grandis Hill ex Maiden (Myrtaceae) plant in the field. The duration and survival of the nymphal stage of B. tabidus were 32.07 ± 8.95 days and 77.00 ± 0.42%, respectively with 15.55 ± 8.19 egg masses, 603.00 ± 379.20 eggs and 387.45 ± 291.40 nymphs per female of this predator while longevity of its males and females was 119.80 ± 32.80 and 160.20 ± 42.13 days, respectively. Better results with E. grandis plant in the field and the fact that B. tabidus will be adapted at release time show that this predator should be reared in these conditions.

Background: Cystinosis is a multisystemic autosomal recessive deficiency of the lysosomal membrane transporter protein (cystinosin) caused by mutations in CTNS gene. Objective: This study summarizes the Portuguese experience in the diagnosis and management of patients with this rare disease over the past few years and reports recurrent mutations in the CTNS gene. Methods: Unrelated patients from different pediatric and adult hospitals all over Portugal with non-nephrotic proteinuria, hypercalciuria, hypokalemia impaired proximal reabsorption of amino acids, glycosuria and hypophosphatemia, suggestive of a Fanconi syndrome and ocular problems, were studied. Intra-leukocyte cystine levels were determined and molecular analysis was performed, to determine the presence or absence of the 57-kb deletion in CTNS, followed by direct sequencing of the coding exons of CTNS. Results: From 1998 to 2017, twenty-one cystinotic patients were biochemically diagnosed. From the remaining seventeen (four deceased), eleven were studied for CTNS gene. Five out of eleven patients were homozygous for the 57-kb deletion (10/22;45.5%), and other five were compound heterozygous for this variant (15/22;68.2%). The other mutations found were p.Q128X (c.721 C>T;2/22), p.S139F (c.755 C>T;4/22) and c.18-21delGACT (p.T7FfsX7;1/22). All of these seventeen cystinotic patients are in treatment. Approximately 84% are adults, 16% are young children, and 54.5% are kidney transplant recipient. Conclusions: The authors would like to emphasize the importance of first screening for the 57-kb deletion since it is very common in our population. This genetic study is the first in our country and it could be the basis for future genetic counseling in Portuguese population.

Cellular viability and nitric oxide (NO) production by J774 macrophages in the presence of orthodontic archwires

To assess, in vitro, the cellular viability in a murine macrophage cell line J774 with 9 different orthodontic wires and to evaluate the effects of its NO production. To assess cellular viability by MTT: 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl tetrazolium bromide assay in the cell line J774 with 9 different orthodontic wires and quantify NO production by these macrophages. Cell cultures were evaluated at 24, 48 and 72 hours. There was no significant difference of the means of cellular viability between the control and the group of wires in the respective time intervals. In the comparison with the control group, there was significant difference in the NO production in groups 1, 6, and 9 at 24 hours interval. Group 8 showed significant difference in relation to the control group at final time interval. Cellular viability in all groups was higher at the final time interval than at the initial time interval. This increase was significant in the control group. In the material groups, the final mean of cellular viability at 72 hours showed no significant difference when compared with the control group. NO production in all groups was higher at the final time interval than at the initial time interval. This increase was significant in the control group. In the material groups, the final mean of NO production at 72 hours was only significant in group 8 (betatitanium) when compared with the control group.

Background: The primary motor cortex (M1) stimulation (MCS) is a useful tool for attenuation of the peripheral neuropathic pain in patients with pharmacologically refractory pain. Furthermore, that neurological procedure may also cause antinociception in rodents with neuropathic pain. Cold allodynia is a frequent clinical finding in patients with neuropathic pain, then, we evaluated if an adapted model of neuropathy induced by chronic constriction injury (CCI) of the ischiadicus nervus (sciatic nerve) produces cold allodynia in an animal model of chronic pain. In addition, we also investigated the effect of the electrical stimulation of the M1 on chronic neuropathic pain condition in laboratory animals. Methods: Male Wistar rats were used. An adapted model of peripheral mononeuropathy induced by CCI was carried out by placing a single loose ligature around the right sciatic nerve. The acetone test was used to evaluate the cold allodynia in CCI or Sham (without ligature) rats. The MCS (M1) was performed at low-frequency (20 μA, 100 Hz) during 15 s by deep brain stimulation (DBS-Thomas Recording device) 21 days after CCI or Sham procedures. The cold allodynia was measured before and immediately after the neurostimulation of M1 in the following time-window: 0, 15 and 30 min after MCS. Results: Cold allodynia threshold increased in animals with chronic neuropathic pain submitted to the acetone test 21 days after the CCI surgery. The M1-stimulation by DBS procedure decreased the cold allodynia immediately and until 30 min after M1-stimulation in rats with chronic neuropathic pain. Conclusion: The current proposal for a CCI model by a single loose ligature of the sciatic nerve can be employed as an experimental model of chronic neuropathic pain in rats submitted to peripheral nervous system injury. Microsatellites and the Polyploid Guarana Plant:Diversity under a Sea of Alleles

Repeat blocks, microsatellites or simple sequence repeats (SSRs) can produce good co-dominant molecular markers for genetic diversity analysis and the determination of self-pollination rates in progenies originating from open pollination of selected genotypes. The enrichment of guarana genomic libraries was underway when it was confirmed that we are working with a complex polyploid species with 210 chromosomes. The probes (CA)12, (CT)12 and (TC)14 were used to finish the enrichment of four libraries for repeat blocks and the screening of a databank of expressed sequence tags (ESTs) from guarana seeded-fruits was accomplished as well. Fifteen clonal cultivars were genotyped with three replicas at 10 out of 27 identified loci using the 59 alleles that passed the reproducibility criterion. A large number of short repeat blocks were identified and this was considered to be a consequence of the recent polyploidization event. However, blocks with eight or more repeats ideal for genotyping were scarce. Annealing of most probes to short blocks by partial complementarity could explain the scarcity of longer blocks in genomic libraries but cannot explain why they were rare in the ESTs. Due to the complexity of the genotypes, alleles were treated as dominant traits. ESTs harboring repeat blocks had the functional annotation renewed. Locus GRN07 is inserted in a homologue of the MOTHER OF FLOWERING LOCUS T AND TFL1 (MFT), in which 3’-UTR displays clear post-transcriptional regulatory features. MFT and its variants are probably involved in the determination of seed germination and embryo growth characteristics. Other accessed loci can be involved in plant architecture and defense reactions. It was concluded that the alleles described in the present work can be used to distinguish guarana cultivars and possibly to analyze segregation using the progenies of controlled pollinations between divergent genitors. Acute effect of moderate intensity physical exercise on postprandial hyperlipaemia of individuals with central obesity

Introduction: High postprandial triglyceride levels and obesity are associated with coronaropathy, and influence the pathogenesis and progression of atherosclerosis. Objective: To evaluate the effect of immediate acute physical exercise on postprandial hyperlipaemia in individuals with central obesity. Methods: Study included sedentary, normolipidemic individuals aged 18 - 30 years, with waist circumference >80 in for women and >94 for men. The physical exercise was performed for 45 minutes with 55% - 60% of the heart rate reserve. Triglyceride level tests were performed after fasting, and at time intervals of 1h and 3 h after lipid overload, a week before and immediately after a session of physical exercise. Results: Thirty-six individuals were evaluated (mean age 23.74 ± 4.05 years, 88.2% women, BMI = 28.4 ± 3.4). Fasting and post-prandial triglyceride levels were similar before and after the exercise (p = 0.87). A smaller rise in triglyceride levels was observed in the 1st hour in obese individuals (5%) compared with over-weight individuals (21%) p = 0.001. Conclusion: In individuals with central obesity, moderate intensity physical exercise performed immediately before lipid overload does not reduce the hyperlipidemic peak.

Spacial and Temporal Variation of Degradation of Organic Matter by Benthic Macroinvertebrates

Leaf litter input from riparian landscapes has been identified as both a major energy flow to stream ecosystems and as a food source for stream macroinvertebrates. In this study the benthic macroinvertebrate community was used to evaluate the decomposition of organic matter in river systems. The aim of this work was to study the decomposition of organic matter using the benthic macroinvertebrate colonization. The research was developed over five months (January 2011 to May 2011) in four different sites, distributed along the River ferreira terminal area, northern Portugal. A litter bag experiment was used to examine the role of macroinvertebrate communities in the processing of organic material on the river. Litter bags were placed in the water and collected every 7, 15, 30, 60, 90 and 120 days. In litter bags was observed a high abundance of Oligochaeta and Chironomidae, which are characterized by being detritivorous-herbivores and filtering collectors, respectively, indicating the benthic macroinvertebrate more involved on the decomposition of organic matter. These results contributed to increase current knowledge about benthic macroinvertebrate communities and may serve as incentive for future research works.

Fluorescence of Chlorophyll a for Discovering Inhibitors of Photosynthesis in Plant Extracts

Aims: Twelve crude extracts were prepared from different parts of Pluchea sagittalis, Cecropia palmata and Brachiaria brizantha, and their effect on the light reaction of photosynthesis. Study Design: Use of fluorescence of ChL a (Chlorophyll a) to detect inhibitors of photosynthesis in plant extracts. Place and Duration of Study: Instituto de Ciências Ambientais, Químicas e Farmacêuticas, Departamento de Ciências Exatas e da Terra, Universidade Federal de Sao Paulo (UNIFESP), between July 2015 and January 2016. Methodology: The samples had their effect on the light reaction of photosynthesis studied by Chlorophyll a (Chl a) fluorescence transient by the use of chloroplasts isolated from spinach leaves. As negative controls we used the same amounts of DMSO that have been employed to solubilize the samples, and as positive control we have used 10 μM of the commercial herbicide DCMU (Diuron). Results: The extracts inhibited the photosynthesis, and their inhibition sites were located in the range of electron flow from OEC complex and between P680 to QA of PS II, and inhibited the photosystem II (PS II) by inducing the appearance of the transient bands K and J. Conclusion: Our results suggest an indicative that the photochemical apparatus probably is failing at the donor and acceptor sides of PSII, causing damage to electron transport during the photosynthesis phenomenon.
