Impact of high-fat diet and vitamin D_3 supplementation on aortic stenosis establishment in waved-2 epidermal growth factor receptor mutant mice

Objective: The use of animal models of aortic stenosis(AS) remains essential to further elucidate its pathophysiology and to evaluate new therapeutic strategies. The waved-2 mouse AS model has been proposed;data have indicated that while aortic regurgitation(Ar) is effectively induced, development of AS is *** aimed to evaluate the effect of high-fat diet(HFD) and vitamin D_3 supplementation in this ***: HFD and subcutaneous vitamin D_3 injections were initiated at the age of 6 weeks until the age of 6(n = 16, 6-month treatment group) and 9(n = 11, 9-month treatment group) months. Twelve waved-2 mice without supplementation were used as control. Echocardiography was performed at 3, 6 and 9 months. Blood serum analysis(calcium, 1,25(OH)_2D_3 and cholesterol), histology and immunohistochemistry(CD-31, CD-68 and osteopontin) were evaluated at the end of the experiment(6 or 9 months).results: Total cholesterol and 1,25(OH)_2D_3 were significantly increased relative to the control group. HFD and vitamin D_3 supplementation did result in improvements to the model, since AS was only detected in 6(15.3%) mice(2 in the 3 groups) and Ar was developed in the remaining animals. Echocardiographic parameters, fibrosis, thickness, inflammation and valvular calcification, were not significantly different between the 6-month treatment and control groups. Similar results were also observed in the 9-month treatment ***: These results suggest that HFD and vitamin D_3 supplementation have no effect in the waved-2 mouse model. This model essentially mimics Ar and rarely AS. Further studies are needed to find a reliable animal model of AS.

Al-driven peculiarities of local coordination and magnetic properties in single-phase Al_(x)-CrFeCoNi high-entropy alloys

Modern design of superior multi-functional alloys composed of several principal components requires in-depth studies of their local structure for developing desired macroscopic ***,peculiarities of atomic arrangements on the local scale and electronic states of constituent elements in the single-phase face-centered cubic(fcc)-and body-centered cubic(bcc)-structured high-entropy Alx-CrFeCoNi alloys(x=0.3 and 3,respectively)are explored by element-specific X-ray absorption spectroscopy in hard and soft X-ray energy *** based on the reverse Monte Carlo approach allow to perform a simultaneous fit of extended X-ray absorption fine structure spectra recorded at K absorption edges of each 3d constituent and to reconstruct the local environment within the first coordination shells of absorbers with high *** revealed unimodal and bimodal distributions of all five elements are in agreement with structure-dependent magnetic properties of studied alloys probed by magnetometry.A degree of surface atoms oxidation uncovered by soft X-rays suggests different kinetics of oxide formation for each type of constituents and has to be taken into account.X-ray magnetic circular dichroism technique employed at l2,3 absorption edges of transition metals demonstrates reduced magnetic moments of 3d metal constituents in the sub-surface region of in situ cleaned fcc-structured Al0.3-CrFeCoNi compared to their bulk *** to nanostructured versions of multicomponent alloys,such studies would bring new insights related to effects of high entropy mixing on low dimensions.

Cutaneous leishmaniasis in an indigenous infant with Down’s syndrome: A case report

rationale: leishmaniasis, caused by the protozoan leishmania and transmitted by sandflies, is endemic to the tropical and subtropical areas of Brazil. It is one of the neglected tropical diseases in the world. Patient concerns: A 20-month-old indigenous infant with severe malnutrition and Down’s syndrome presented with a facial ulcer for 5 months. Diagnosis: Giemsa staining of scraped ulcer tissues indicated the presence of the amastigote form of leishmania sp., and positive Montenegro’s intradermal test helped in diagnosing the condition as cutaneous leishmaniasis. Interventions: The child was hospitalized and received intravenous treatment for cutaneous leishmaniasis(1 mg/kg/day of amphotericin B). Outcomes: The condition was cured with amphotericin B(1 mg/kg/day for 14 d). lessons: Because of infanticide practices in indigenous cultures, indigenous infants with Down’s syndrome rarely survive. Thus, no similar case has been reported in the literature.

The road to sustainable use and waste management of plastics in Portugal

As a European Union(EU)member,Portugal must comply with reductions in plastic *** Portugal,the 330 items/100 m of beach litter,comprising up to 3.9 million pieces and of which 88%is plastic,is higher than the EU median(149 items/100 m)and must be reduced to 20 items/100 m(94%).Integrative measures are needed to reduce littering and improve plastics’use and disposal under the circular *** this 414 kt of plastic packaging waste,163 kt were declared plastic packaging,140 kt subjected to recycling,and 94 kt to energy *** current recycling rate of plastic packaging(34%)should be improved to reach EU recycling averages(42%)and goals and to provide widespread benefits,considering revenues of 167€/*** a net importer of waste,Portugal could benefit from the valorization of imported *** increased recycling,pyrolysis and gasification could provide short-term alternatives for producing value-added substances from plastic waste,such as hydrogen,consistent with the National Plan of Hydrogen and improving ongoing regulations on single-use *** manuscript provides an integrative view of plastics in Portugal,from use to disposal,providing specific recommendations under the circular economy.

Assessment of periportal fibrosis in Schistosomiasis mansoni patients by proton nuclear magnetic resonance-based metabonomics models

BACKGrOUND The evaluation of periportal fibrosis(PPF)is essential for a prognostic assessment of patients with Schistosomiasis *** WHO Niamey Protocol defines patterns of fibrosis from abdominal ultrasonography,^(1)H-nuclear magnetic resonance(NMr)-based metabonomics has been employed to assess liver fibrosis in some *** To build 1H-NMr-based metabonomics models(MM)to discriminate mild from significant periportal PPF and identify differences in the metabolite *** A prospective cross-sectional study was performed on schistosomiasis patients at a University Hospital in Northeastern *** evaluated 41 serum samples from 10 patients with mild PPF(C Niamey pattern)and 31 patients with significant PPF(D/E/F Niamey patterns).MM were built using partial least squares-discriminant analysis(PlS-DA)and orthogonal projections to latent structures discriminant analysis(OPlS-DA)*** PlS-DA and OPlS-DA resulted in discrimination between mild and significant PPF groups with r2 and Q2 values of 0.80 and 0.38 and 0.72 and 0.42 for each model,*** OPlS-DA model presented accuracy,sensitivity,and specificity values of 92.7%,90.3%,and 100%to discriminate significant *** metabolites identified as responsible by discrimination were:Nacetylglucosamines,alanine,glycolaldehyde,carbohydrates,and *** MMs discriminated mild from significant PPF patterns in patients with Schistosomiasis mansoni through identification of differences in serum metabolites profiles.

The stone marten(Martes foina)is a carnivorous mammal that often consumes fleshy fruits,thus potentially promoting seed *** present study was developed in Bussaco National Forest,central Portugal,and aimed to assess the potential role of the stone marten in dispersing native and exotic plants in different forest landscape *** from stone marten scats and fleshy fruits were collected monthly and were thereafter identified and sowed in a nursery,following a randomized experimental *** emergence was monitored *** linear models were used to test for differences in time and success of emergence between seeds from scats and fruits of 3 native species(rubus ulmifolius,Arbutus unedo and Celtis australis)and 1 exotic plant species with invasive behavior(Prunus laurocerasus).Fruit consumption by the stone martens significantly increased and accelerated the germination of the native *** but had no effect on the other 2 native species or on the invasive *** suggested that stone martens contribute to gene flow and forest regeneration by dispersing native plant ***,although the germination was not enhanced in the invasive species,the preference of stone martens for these fruits may potentially contribute to the proliferation of *** study represents a contribution to better understanding the fauna and flora interactions,enabling for a more conscious and effective decision-making in forest management.

Amyotrophic lateral sclerosis: a complex syndrome that needs an integrated research approach

Amyotrophic lateral sclerosis, the most common neurodegenerative disease affecting motor neurons, lacks an effective treatment. A small fraction of amyotrophic lateral sclerosis cases have a familial origin, related to mutations in causative genes, while the vast majority of amyotrophic lateral sclerosis cases are considered to be sporadic, resulting from the interaction between genes and environmental factors in predisposed individuals. During the past few years, dozens of drugs have been postulated as promising strategies for the disease after showing some beneficial effects in preclinical cellular and murine models. However, the translation into clinical practice has been largely unsuccessful and the compounds failed when were tested in clinical trials. This might be explained, at least partially, by the enormous complexity of the disease both from clinico-epidemiological and a pathogenic points of view. In this review, we will briefly comment on the complexity of the disease focusing on some recent findings, and we will suggest how amyotrophic lateral sclerosis research might be reoriented to foster the advance in the diagnostic and therapeutic questions.

NEW HYBrID CONJUGATE GrADIENT METHOD AS A CONVEX COMBINATION OF lS AND Fr METHODS

In this paper, we present a new hybrid conjugate gradient algorithm for unconstrained optimization. This method is a convex combination of liu-Storey conjugate gradient method and Fletcher-reeves conjugate gradient method. We also prove that the search direction of any hybrid conjugate gradient method, which is a convex combination of two conjugate gradient methods, satisfies the famous D-l conjugacy condition and in the same time accords with the Newton direction with the suitable condition. Furthermore, this property doesn't depend on any line search. Next, we also prove that, moduling the value of the parameter t,the Newton direction condition is equivalent to Dai-liao conjugacy *** strong Wolfe line search conditions are *** global convergence of this new method is *** comparisons show that the present hybrid conjugate gradient algorithm is the efficient one.
